Organochlorine compounds (OCs), including pesticides and polychlorobiphenyls (PCBs), were determined in the serum of 499 cord blood samples from infants born during 2004−2006 in Valencia (Spain). The concentrations were found to be correlated with the region of origin of the mothers. Those from Latin-America gave birth to infants with higher 4,4′-DDE and 4,4′-DDT concentrations than those from Europe. On the contrary, European mothers had children with higher concentrations of hexachlorobenzene and PCBs than Latin-American mothers. These associations were independent of maternal period of stage in Valencia. Data examination with a multivariate model including maternal region of origin described 39−44% of the concentration variability of 4,4′-DDE and total PCBs with statistic significance. Other maternal variables found to significantly influence OC burden in newborns were age, education level, and body mass index (BMI). In general, older women, with higher education standards and higher BMI had infants with higher OC serum concentrations than younger women, with lower education scores and low BMI. 6 páginas, 2 figuras, 2 tablas. ; Polybromodiphenyl ethers (PBDEs) were analyzed in blood serum of pregnant women and in cord blood serum of their newborns from a general population cohort (n = 174; Valencia, Spain). The most abundant PBDE congeners identified were BDE 47, BDE 99, BDE 153, BDE 154 and BDE 209. Their cord blood serum concentrations were about 45% of those in maternal serum but after lipid normalization median of total PBDEs was 9.6 ng/g lipid in cord serum (concentrations encompassing between not detected and 140 ng/g lipid) and 9.6 ng/ lipid in maternal serum(concentrations covering between not detected and 120 ng/g lipid). The distributions of these compounds were dominated by BDE 47 in both cases. In cord blood serum the decreasing order of abundance was BDE 47 > BDE 99 > BDE 209 > BDE 153 > BDE 154. The congener composition in maternal serum followed a similar trend: BDE 47 > BDE 153 > BDE 154 > BDE 209 > BDE 99. The congener concentrations exhibited a higher degree of correlation in cord blood than in maternal serum. Use of the maternal determinants for categorization of the observed maternal and fetal PBDE concentrations only showed significant associations for the levels in umbilical cord. Neonates from rural areas exhibited statistically significantly lower concentrations than those from urban, semi-urban or metropolitan sites. Maternal serum also showed this difference but the higher dispersion of the concentrations in maternal serum did not afford its recognition with statistical significance. The lower qualitative and quantitative variability in the PBDE concentrations of cord blood serum than maternal serum suggest that the latter is reflecting PBDE contributions from a wider diversity of sources than the former whereas cord blood sera seems to represent the long term standing stock of these compounds accumulated in the maternal tissues. ; The authors are grateful to the mothers who participated in the study and to Hospital La Fe. En este estudio se investigan los valores de trihalometanos en el agua de consumo suministrada por varios sistemas de abastecimiento de la provincia de Granada, en el área de actuación de la cohorte madres-hijos de la Red INMA (Infancia y Medio Ambiente). ; Métodos: Se analizaron 82 muestras de agua de consumo en dos campañas de muestreo en invierno y verano de 2006. Se determinó la concentración de cloroformo, romodiclorometano, dibromoclorometano y bromoformo, siguiendo un procedimiento optimizado basado en cromatografía de gases y espectrometría de masas. ; Resultados: El rango de concentración de trihalometanos totales se situó entre 0,14 y 18,75 ug/l en la campaña de invierno y entre 0,01 y 31,87 ug/l en la de verano. El compuesto mayoritario fue cloroformo. La concentración media de trihalometanos en agua de origen superficial y subterráneo fue de 10,13 y 1,41 ug/l, respectivamente. ; Conclusiones: Los valores de trihalometanos encontrados son muy inferiores a la concentración máxima admisible (100 ug/l) establecida por la Unión Europea para estos compuestos. La presencia de trihalometanos en la zona es menor a la descrita en otras regiones españolas. ; [ENG] Objectives: Drinking water chlorination generates potentially harmful by-products, such as trihalomethanes. Trihalomethane levels are high in some parts of Spain. The aim of the present study was to investigate trihalomethane concentrations in drinking water from distinct water supplies in the province of Granada, within the framework of the Childhood and Environment (INMA) study. ; Methods: Eighty-two tap water samples were collected in two campaigns during the winter and summer of 2006. An optimized procedure based on gas chromatography and mass spectrometry was used to determine concentrations of chloroform, bromodichloromethane, dibromochloromethane, and bromoform in the samples. ; Results: Total trihalomethane concentrations ranged from 0.14 to 18.75 ug/l in winter samples and from 0.01 to 31.87 ug/l in summer samples. The most abundant compound was chloroform. Mean trihalomethane concentrations were 10.13 in surface waters and 1.41 ug/l in ground waters. ; Conclusions: The trihalomethane levels found were considerably below the maximum permitted level of 100 ug/l in the European Union. The values obtained varied widely according to the type of water source: the highest concentrations were found in urban and sub-urban areas, where the water is largely of surface origin. Despite reductions in their use and fugitive release, they remain one of the most important groups of persistent pollutants to which humans are exposed, primarily through dietary intake. We designed a case-control study to assess the risk of colorectal cancer with exposure to these chemicals, and their potential interactions with genetic alterations in the tumors. A subsample of cases (n = 132) and hospital controls (n = 76) was selected from a larger case-control study in Barcelona, Catalonia, Spain. We measured concentrations in serum of several organochlorines by gas chromatography. We assessed point mutations in K-ras and p53 genes in tissue samples by polymerase chain reaction/single-strand conformation polymorphism and assessed expression of p53 protein by immunohistochemical methods. An elevated risk of colorectal cancer was associated with higher serum concentrations of mono-ortho polychlorinated biphenyl (PCB) congeners 28 and 118. The odds ratio for these mono-ortho PCBs for middle and higher tertile were, respectively, 1.82 [95% confidence interval (CI) , 0.90-3.70] and 2.94 (95% CI, 1.39-6.20) . alpha-Hexachlorocyclohexane, hexachlorobenzene, and p,p´-DDE (4,4´-dichlorodiphenyltrichloroethene) showed nonsignificant increases in risk. Risk associated with mono-ortho PCBs was slightly higher for tumors with mutations in the p53 gene but was not modified by mutations in K-ras. Mono-ortho PCBs were further associated with transversion-type mutations in both genes. These results generate the hypothesis that exposure to mono-ortho PCBs contributes to human colorectal cancer development. Background Occupational exposure to pesticides has been reported among general population worldwide. However, little is known about the associations between non-occupational exposure to pesticides, and biological markers of health and their response by sex. Objectives We aimed to assess the associations between non-occupational overall pesticide exposure, length of exposure and specific pesticides reported with 35 biological markers of health representing cardiometabolic, haematological, lung function, sex hormones, liver and kidney function profiles, and vitamin D in Finnish cohort. Methods 31-year cross-sectional examination of the Northern Finland Birth Cohort 1966 provided blood samples for biomarker measurements in 1997–1998. Number of subjects varied between 2361 and 5037 for given exposures and certain outcome associations. Multivariable regression analyses were performed to examine associations between overall pesticide exposure (OPE), length of pesticide exposure in months (PEM), in years (PEY), and specific pesticides use (PEU) or not with cardiometabolic [SBP, DBP, TC, LDL, HDL, triglycerides, fasting glucose, insulin, HOMA-IR, HOMA-B, HOMA-S, hs-CRP], hematological [WBC, RBC, Hb, HCT, MCV, MCH, MCHC, platelets], lung function (FVC, FEV1), sex hormones [luteinizing hormone (LH), testosterone (TT), sex-hormone binding globulin (SHBG)], liver and kidney function profiles [total protein, albumin, globulin, ALP, ALT, GGT, urea, creatinine], and vitamin D adjusting for sex, BMI, socioeconomic position (SEP) and season of pesticide use. Results This cohort study on up to 5037 adults with non-occupational OPE, PEM, PEY and PEU differed by sex and SEP. In regression analyses, all the exposures were positively associated with total cholesterol and low-density lipoprotein cholesterol, and PEU was negatively associated with high-density lipoprotein cholesterol in females. OPE and PEM were positively associated with haematocrit in females and PEU with platelets in males. PEU was negatively associated with mean corpuscular haemoglobin. OPE and PEM were positively associated with LH in males. OPE was negatively associated with total protein and albumin in males. Discussion In Finnish young adults, non-occupational overall pesticide exposure, length of exposure and specific pesticides were associated with multiple biological markers of health. In this study, we analyzed the sources of ambient PM inorganic and organic components near a cement plant using fossil fuels as well as alternative fuels, such as sewage sludge and biomass. Source apportionment methodologies, i.e., principal component analysis (PCA) and multivariate curve resolution by alternating least squares (MCR-ALS), and carbon isotope analysis (δ13C) were used to determine the potential sources and their contributions. Four sources of PM10 main tracer compounds constituents were identified: Marine and secondary inorganic aerosol, cement plant/industrial, traffic and crustal. The contributions of those sources varied significantly depending on the period of the year. However, the inorganic tracer PM species in the area were mainly released by combustion sources, namely traffic and the activity of the cement plant, especially in winter months. The analyses of tracer organic compounds also indicated combustion sources, i.e., biomass burning and fossil fuel combustion, as the predominant contributors to ambient air PM (62, 59 and 69%, in PM10, PM2.5 and PM1, respectively). Organic dust was a significant source of PM10 (33%) while its contribution was found to be minor in the finest fractions (9 and 2% in PM2.5 and PM1, respectively). Urinary metabolites of organophosphate (OP) and pyrethroid (PYR) pesticides and biomarkers of effects were studied in a population (n=40) residing in an important agricultural area of the province of Córdoba (Argentina). Detection frequencies (DF) higher than 85% were observed for the metabolites of pirimiphos (2-diethylamino-6-methylpyrimidin-4-ol –DEAMPY-, median 7.5 μg/g creatinine, DF: 100%), parathion (p-nitrophenol, 0.99 μg/g creatinine, 100%), and chlorpyrifos (3,5,6-trichloro-2-pyridinol, 0.25 μg/g creatinine, 85%). The DEAMPY concentrations doubled the levels found in other studies and were negatively associated with Er-AChE activity, suggesting the appearance of health effects already in environmental exposure levels below established acceptable daily intakes (ADIs). 3-Phenoxybenzoic acid, the metabolite of several PYR pesticides, was also found in all samples. In the present study, we evaluated the concentrations of PAHs in 3 PM fractions (10, 2.5 and 1) collected in the surroundings of a cement plant located in Barcelona. PAH content and speciation were developed for the three fractions to elucidate their distribution among different sizes. 11 páginas, 4 figuras, 6 tablas. ; Prenatal levels of organohalogen compounds (OHCs), including polybromodiphenyl ethers (PBDEs), polychlorobiphenyls (PCBs), hexachlorobenzene (HCB), hexachlorocyclohexanes and DDTs have been investigated in two different cohorts from the Spanish Mediterranean area, Valencia and Menorca, by analysis of cord blood serum. Comparison of the observed median concentrations to other cohorts show low PBDE levels (2.8-6.8 ng/g lipid) but high values of HCB (0.68 ng/ml, 260 ng/g lipid) and PCBs (0.65 ng/ml, 240 ng/g lipid) in the cohort of Menorca indicating that Mediterranean areas, and not only high latitude regions, may contain population groups highly exposed to some of these pollutants. Significant differences in the concentrations of these compounds have been found between the two cohorts. Differences in maternal population such as age or body mass index cannot explain this variation. One possible cause of variability is the seven year time period elapsed between the two cohort recruitments but the strong differences observed do not allow exclusion of local diet differences. Thus, the different average PCB congener distributions between the two cohorts suggests an influence from materials containing diverse PCB mixtures. The congener mixtures found in Menorca could reflect a diet contribution from some western Mediterranean fish species.
