Insertion sequences (ISs) are ubiquitous and abundant mobile genetic elements in prokaryotic genomes. ISs often encode only one protein, the transposase, which catalyzes their transposition. Recent studies have shown that transposases of many different IS families interact with the β sliding clamp, a DNA replication factor of the host. However, it was unclear to what extent this interaction limits or favors the ability of ISs to colonize a chromosome from a phylogenetically-distant organism, or if the strength of this interaction affects the transposition rate. Here we describe the proliferation of a member of the IS1634 family in Acidiphilium over ~600 generations of cultured growth. We demonstrate that the purified transposase binds to the β sliding clamp of Acidiphilium, Leptospirillum and E. coli. Further, we also demonstrate that the Acidiphilium IS1634 transposase binds to the archaeal sliding clamp (PCNA) from Methanosarcina, and that the transposase encoded by Methanosarcina IS1634 binds to Acidiphilium β. Finally, we demonstrate that increasing the strength of the interaction between β and transposase results in a higher transposition rate in vivo. Chagas disease is caused by Trypanosoma cruzi infection, being cardiomyopathy the more frequent manifestation. New chemotherapeutic drugs are needed but there are no good biomarkers for monitoring treatment efficacy. There is growing evidence linking immune response and metabolism in inflammatory processes and specifically in Chagas disease. Thus, some metabolites are able to enhance and/or inhibit the immune response. Metabolite levels found in the host during an ongoing infection could provide valuable information on the pathogenesis and/or identify deregulated metabolic pathway that can be potential candidates for treatment and being potential specific biomarkers of the disease. To gain more insight into those aspects in Chagas disease, we performed an unprecedented metabolomic analysis in heart and plasma of mice infected with T. cruzi. Many metabolic pathways were profoundly affected by T. cruzi infection, such as glucose uptake, sorbitol pathway, fatty acid and phospholipid synthesis that were increased in heart tissue but decreased in plasma. Tricarboxylic acid cycle was decreased in heart tissue and plasma whereas reactive oxygen species production and uric acid formation were also deeply increased in infected hearts suggesting a stressful condition in the heart. While specific metabolites allantoin, kynurenine and p-cresol sulfate, resulting from nucleotide, tryptophan and phenylalanine/tyrosine metabolism, respectively, were increased in heart tissue and also in plasma. Negli ultimi decenni le nuove scoperte mediche e il miglioramento dello stile di vita nei paesi occidentali hanno determinato un aumento del consumo di sostanze terapeutiche, nonché della gamma di prodotti farmaceutici e parafarmaceutici comunemente utilizzati. Gli studi di cinetica dei farmaci dimostrano con certezza che essi, seppur metabolizzati, mantengono inalterato il loro potere d'azione, rimanendo biodisponibili anche una volta escreti dall'organismo. A causa della loro eccessiva polarità tali molecole sono inoltre difficilmente trattenute dai convenzionali impianti di depurazione dei reflui urbani, dai quali confluiscono inevitabilmente verso le acque fluviali e/o costiere le quali risultano. In ragione di ciò, la valutazione degli effetti che la loro presenza può provocare sulla qualità dei sistemi di approvigionamento idrico e sulla biologia delle specie tipiche degli ecosistemi acquatici, ha classificato tali composti come una nuova classe di inquinanti emergenti, il cui impatto ambientale non risulta ancora del tutto arginato attraverso adeguate contromisure legislative. I farmaci sono sostanze bioattive progettate per avere effetti specifici a bassissime concentrazioni negli organismi target attraverso specifici meccanismi d'azione. Nel caso in cui i bersagli cellulari su cui agiscono siano evolutivamente conservati negli organismi non target, essi possono esercitare le proprie funzioni attraverso i medesimi meccanismi di regolazione fisiologica attivati nelle specie target, dando origine a effetti specifici, o anche aspecifici, nel caso in cui tali bersagli siano deputati alla regolazioni di funzioni differenti. Pertanto lo scopo del presente lavoro di tesi è stato quello di analizzare le possibili alterazioni di carattere fisiologico in individui di mitilo mediterraneo (Mytilus galloprovincialis) esposti a concentrazioni ambientali di fluoxetina, farmaco antidepressivo appartenente alla classe degli inibitori selettivi del riassorbimento presinaptico della serotonina (SSRI). Più nel dettaglio, a seguito di un'esposizione per 7 giorni a dosaggi compresi tra 0.03 e 300 ng/L di FX, sono stati analizzati i livelli intracellulari di AMPc e l'attività dell'enzima PKA nei diversi trattamenti sperimentali effettuati. Inoltre sono stati valutati i livelli di espressione genica del recettore serotoninergico 5HTmyt1 e della la P-glicoproteina (Pgp, gene ABCB1), trasportatore di membrana responsabile del sistema di detossificazione noto come Multi-xenobiotic resistance (MXR). Nella ghiandola digestiva, la FX causa una diminuzione statisticamente significativa dei livelli di AMPc, dell'attività della PKA e dell'espressione del gene ABCB1 rispetto al controllo. Al contrario nel mantello il farmaco non produce alterazioni dei livelli intracellulari di AMPc e dell'attività della PKA mentre si apprezza una sottoespressione del gene ABCB1 nei trattamenti a dosaggi intermedi. In entrambi i tessuti si nota un aumento dell'espressione genica di 5HTmyt1 alle minori concentrazioni di FX, mentre ai dosaggi più alti non si registrano alterazioni significative rispetto al controllo. Nel loro insieme i risultati indicano che nei mitili, concentrazioni ambientali di FX producono significative alterazioni di diversi parametri fisiologici attraverso una modulazione specifica dei medesimi bersagli molecolari coinvolti nella terapia umana. La riduzione dei livelli di AMPc/PKA apprezzata nella ghiandola digestiva risulta in linea con la funzione inibitoria svolta dal recettore 5HTmyt1 su tale via di trasduzione, mentre l'assenza di variazioni significative registrata nel mantello supporta l'ipotesi di un'interazione tra il sistema serotoninergico e catecolaminergico nella regolazione dei processi legati al ciclo riproduttivo che si verificano in tale tessuto. In conclusione, i dati dimostrano che l'espressione del gene codificante la proteina Pgp è regolata dalla FX attraverso uno specifico meccanismo d'azione AMPc-dipendente modulato dalla serotonina; tuttavia, è ipotizzabile anche un effetto non specifico indotto dalla FX stessa, per esempio attraverso l'induzione di stress ossidativo. Inoltre essi evidenziano la presenza di un meccanismo di regolazione retroattivo sulla espressione dei recettori 5HTmyt1 in funzione delle concentrazioni extracellulari di serotonina modulate dall'azione della FX.

Agri-environmental schemes (AES) of the Common Agricultural Policy (CAP) aims at reversing the negative effects of agricultural intensification on biodiversity and ecosystem services. Landscape context may modulate, and even constraint, AES effectiveness. We evaluate AES effectiveness on ant abundance, diversity and community composition. Ants are an ecologically dominant group whose response to conservation efforts in farmland has been rarely evaluated, despite its role in weed control, particularly in Mediterranean farmland. Ants were sampled in the edge and in the centre of paired cereal fields, managed with and without AES in three study areas along a landscape complexity gradient. AES application had no significant effects on ant species richness or ant community composition. Richness increased in fields and landscapes with higher amounts of complex edges and decreased towards the centre of the fields. Specialist granivorous ants (harvester ants, Messor spp.) were the most abundant. Abundance of foraging ants increased with the amount of complex edges around fields and in the landscape. AES application increased ant abundance close to field edges but not in field centers. AES fields had less specialist granivorous foraging in their centers than in control field centers. Ant communities in Mediterranean cereal cropland were mostly constrained by the availability of complex edges, needed for nest building. AES increased the abundance of foraging ants, mostly specialist harvester ants, and its potential service of weed control, but close to field edges mainly. By controlled hydrolysis of chitosan or chitin with different enzymes, three types of chitooligosaccharides (COS) with MW between 0.2 and 1.2 kDa were obtained: Fully deacetylated (fdCOS), partially acetylated (paCOS), and fully acetylated (faCOS). The chemical composition of the samples was analyzed by high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD) and MALDI-TOF mass spectrometry. The synthesized fdCOS was basically formed by GlcN, (GlcN)2, (GlcN)3, and (GlcN)4. On the contrary, faCOS contained mostly GlcNAc, (GlcNAc)2 and (GlcNAc)3, while paCOS corresponded to a mixture of at least 11 oligosaccharides with different proportions of GlcNAc and GlcN. The anti-inflammatory activity of the three COS mixtures was studied by measuring their ability to reduce the level of TNF-α (tumor necrosis factor) in murine macrophages (RAW 264.7) after stimulation with a mixture of lipopolysaccharides (LPS). Background: Tumour-derived exosomes can be released to serum and provide information on the features of the malignancy, however, in order to perform systematic studies in biological samples, faster diagnostic techniques are needed, especially for detection of low abundance proteins. Most human cancer cells are positive for at least one ligand for the activating immune receptor NKG2D and the presence in plasma of NKG2D-ligands can be associated with prognosis. Methods: Using MICA as example of a tumour-derived antigen, endogenously expressed in metastatic melanoma and recruited to exosomes, we have developed two immunocapture-based assays for detection of different epitopes in nanovesicles. Although both techniques, enzyme-linked immunosorbent assay (ELISA) and Lateral flow immunoassays (LFIA) have the same theoretical basis, that is, using capture and detection antibodies for a colorimetric read-out, analysis of exosome-bound proteins poses methodological problems that do not occur when these techniques are used for detection of soluble molecules, due to the presence of multiple epitopes on the vesicle. Results: Here we demonstrate that, in ELISA, the signal obtained was directly proportional to the amount of epitopes per exosome. In LFIA, the amount of detection antibody immobilized in Au-nanoparticles needs to be low for efficient detection, otherwise steric hindrance results in lower signal. We describe the conditions for detection of MICA in exosomes and prove, for the first time using both techniques, the co-existence in one vesicle of exosomal markers (the tetraspanins CD9, CD63 and CD81) and an endogenously expressed tumour-derived antigen. The study also reveals that scarce proteins can be used as targets for detection antibody in LFIA with a better result than very abundant proteins and that the conditions can be optimized for detection of the protein in plasma. The extreme genetic diversity of the protozoan Trypanosoma cruzi has been proposed to be associated with the clinical outcomes of the disease it provokes: Chagas disease (CD). To address this question, we analysed the similarities and differences in the CD pathophysiogenesis caused by different parasite strains. Using syngeneic mice infected acutely or chronically with 6 distant parasite strains, we integrated simultaneously 66 parameters: parasite tropism (7 parameters), organ and immune responses (local and systemic; 57 parameters), and clinical presentations of CD (2 parameters). While the parasite genetic background consistently impacts most of these parameters, they remain highly variable, as observed in patients, impeding reliable one-dimensional association with phases, strains, and damage. However, multi-dimensional statistics overcame this extreme intra-group variability for each individual parameter and revealed some pathophysiological patterns that accurately allow defining (i) the infection phase, (ii) the infecting parasite strains, and (iii) organ damage type and intensity. Retina ganglion cell (RGC) axons grow along a stereotyped pathway undergoing coordinated rounds of fasciculation and defasciculation, which are critical to establishing proper eye– brain connections. How this coordination is achieved is poorly understood, but shedding of guidance cues by metalloproteinases is emerging as a relevant mechanism. Secreted Frizzled Related Proteins (Sfrps) are multifunctional proteins, which, among others, reorient RGC growth cones by regulating intracellular second messengers, and interact with Tolloid and ADAM metalloproteinases, thereby repressing their activity. Here, we show that the combination of these two functions well explain the axon guidance phenotype observed in Sfrp1 and Sfrp2 single and compound mouse mutant embryos, in whichRGCaxons make subtle but significant mistakes during their intraretinal growth and inappropriately defasciculate along their pathway. The distribution of Sfrp1 and Sfrp2 in the eye is consistent with the idea that Sfrp1/2 normally constrain axon growth into the fiber layer and the optic disc. Disheveled axon growth instead seems linked to Sfrp-mediated modulation of metalloproteinase activity. Indeed, retinal explants from embryos with different Sfrp-null alleles or explants overexpressing ADAM10 extend axons with a disheveled appearance, which is reverted by the addition of Sfrp1 or an ADAM10-specific inhibitor. For a long time, most attention has focused on neurons in a neuronocentric conceptualization of central nervous system development, these cells fulfilling an intrinsic program that establishes the brain's morphology and function. By contrast, glia have mainly been studied as support cells, offering guidance or as the cells that react to brain injury. However, new evidence is appearing that demonstrates a more fundamental role of glial cells in the control of different aspects of neuronal development and function, events in which the influence of neurons is at best weak. Moreover, it is becoming clear that the function and organization of the nervous system depends heavily on reciprocal neuron-glia interactions. During development, neurons are often generated far from their final destination and while intrinsic mechanisms are responsible for neuronal migration and growth, they need support and regulatory influences from glial cells in order to migrate correctly. Similarly, the axons emitted by neurons often have to reach faraway targets and in this sense, glia help define the way that axons grow. Moreover, oligodendrocytes and Schwann cells ultimately envelop axons, contributing to the generation of nodes of Ranvier. Finally, recent publications show that astrocytes contribute to the modulation of synaptic transmission. In this sense, purinergic receptors are expressed widely by glial cells and neurons, and recent evidence points to multiple roles of purines and purinergic receptors in neuronal development and function, from neurogenesis to axon growth and functional axonal maturation, as well as in pathological conditions in the brain. Colorectal and pancreatic cancers remain important contributors to cancer mortality burden and, therefore, new therapeutic approaches are urgently needed. Rosemary (Rosmarinus officinalis L.) extracts and its components have been reported as natural potent antiproliferative agents against cancer cells. However, to potentially apply rosemary as a complementary approach for cancer therapy, additional information regarding the most effective composition, its antitumor effect in vivo and its main molecular mediators is still needed. In this work, five carnosic acid-rich supercritical rosemary extracts with different chemical compositions have been assayed for their antitumor activity both in vivo (in nude mice) and in vitro against colon and pancreatic cancer cells. We found that the antitumor effect of carnosic acid together with carnosol was higher than the sum of their effects separately, which supports the use of the rosemary extract as a whole. In addition, gene and microRNA expression analyses have been performed to ascertain its antitumor mechanism, revealing that up-regulation of the metabolic-related gene GCNT3 and down-regulation of its potential epigenetic modulator miR-15b correlate with the antitumor effect of rosemary. Moreover, plasmatic miR-15b down-regulation was detected after in vivo treatment with rosemary. A presente pesquisa tem por objetivo compreender como discentes do curso de especialização em Ensino de Biologia, modalidade a distância, discutem a temática de gênero e sexualidades em sua prática docente. Para tanto, foi realizada a aplicação de questionário via Google Docs a discentes matriculados nos 8 polos que ofertam o referido curso de especialização pelo período de 18 a 25 de maio de 2019. Partimos da premissa de que a escola se constitui enquanto heteronormativa e LGBTfóbica. Além disso, o discurso biológico se configura como um regulador de práticas humanas. Desta forma, docentes da disciplina de biologia possuem um importante papel na desconstrução de práticas com enfoque no determinismo biológico para a compreensão de gênero e sexualidades, bem como no combate a preconceitos e discriminações. Os resultados evidenciam que, em sua maioria, os docentes discutem tranquilamente as temáticas, tornando-se mobilizadores neste processo de abertura para tais abordagens em âmbito educacional, mesmo com a falta de preparo e omissão por parte de outros docentes e da equipe pedagógica. Por fim, a maioria dos participantes concordam com a importância dos temas em âmbito escolar, utilizando das mais variadas metodologias e estratégias para a promoção do respeito e ao enfrentamento da LGBTfobia.

[EN] DELLA proteins are the negative regulators of the gibberellin (GA) signaling pathway. GAs have a pervasive effect on plant physiology, influencing processes that span the entire life cycle of the plant. All the information encoded by GAs, either environmental or developmental in origin, is canalized through DELLAs, which modulate the activity of many transcription factors and transcriptional regulators. GAs unlock the signaling pathway by triggering DELLA polyubiquitination and degradation by the 26S proteasome. Recent reports indicate, however, that there are other pathways that trigger DELLA polyubiquitination and degradation independently of GAs. Moreover, results gathered during recent years indicate that other post-translational modifications (PTMs), namely phosphorylation, SUMOylation and glycosylation, modulate DELLA function. The convergence of several PTMs in DELLA therefore highlights the strict regulation to which these proteins are subject. Biomedical engineering (BME) has the potential of transforming medical care towards universal healthcare by means of the democratization of medical technology. To this end, innovative holistic approaches and multidisciplinary teams, built upon the gathering of international talent, should be encouraged within the medical industry. However, these transformations can only be accomplished if BME education also continuously evolves and focuses on the internationalization of students, the promotion of collaborative design strategies and the orientation towards context relevant medical needs. In this study we describe an international teaching-learning experience, the ''UBORA (Swahili for 'excellence') Design School''. During an intensive week of training and collaboration 39 engineering students lived through the complete development process for creating innovative open-source medical devices following the CDIO (''conceivedesign-implement-operate'') approach and using the UBORA e-infrastructure as a co-design platform. Our post-school survey and analyses showed that this integral teaching-learning experience helped to promote professional skills and could nurture the future generation of biomedical engineers, who could transform healthcare technology through collaborative design oriented to open source medical devices.

Despite increasing work detailing the presence of foraging specializations across a range of taxa, limited attention so far has been given to the role of spatiotemporal variation in food predictability in shaping individual resource selection. Here, we studied the exploitation of human-provided carrion resources differing in predictability by Canarian Egyptian vultures (Neophron percnopterus majorensis). We focussed specifically on the role of individual characteristics and spatial constraints in shaping patterns of resource use. Using high-resolution GPS data obtained from 45 vultures tracked for 1 year, we show that individual vultures were repeatable in both their monthly use of predictable and semi-predicable resources (feeding station vs. farms) and monthly levels of mobility (home range size and flight activity).However, individual foraging activities were simultaneously characterized by a high degree of (temporal) plasticity in the use of the feeding station in specific months. Individual rank within dominance hierarchy revealed sex-dependent effects of social status on resource preference in breeding adults, illustrating the potential complex social mechanisms underpinning status-dependent resource use patterns.
