Many immune diseases occur at different rates among people with schizophrenia compared to the general population. Here, we evaluated whether this phenomenon might be explained by shared genetic risk factors. We used data from large genome-wide association studies to compare the genetic architecture of schizophrenia to 19 immune diseases. First, we evaluated the association with schizophrenia of 581 variants previously reported to be associated with immune diseases at genome-wide significance. We identified five variants with potentially pleiotropic effects. While colocalization analyses were inconclusive, functional characterization of these variants provided the strongest evidence for a model in which genetic variation at rs1734907 modulates risk of schizophrenia and Crohn's disease via altered methylation and expression of EPHB4—a gene whose protein product guides the migration of neuronal axons in the brain and the migration of lymphocytes towards infected cells in the immune system. Next, we investigated genome-wide sharing of common variants between schizophrenia and immune diseases using cross-trait LD score regression. Of the 11 immune diseases with available genome-wide summary statistics, we observed genetic correlation between six immune diseases and schizophrenia: inflammatory bowel disease (rg = 0.12 ± 0.03, P = 2.49 × 10−4), Crohn's disease (rg = 0.097 ± 0.06, P = 3.27 × 10−3), ulcerative colitis (rg = 0.11 ± 0.04, P = 4.05 × 10–3), primary biliary cirrhosis (rg = 0.13 ± 0.05, P = 3.98 × 10−3), psoriasis (rg = 0.18 ± 0.07, P = 7.78 × 10–3) and systemic lupus erythematosus (rg = 0.13 ± 0.05, P = 3.76 × 10–3). With the exception of ulcerative colitis, the degree and direction of these genetic correlations were consistent with the expected phenotypic correlation based on epidemiological data. Rheumatoid arthritis (RA) is a polygenic disease associated with accelerated atherosclerosis and increased cardiovascular (CV) mortality. JAK/STAT signalling pathway is involved in autoimmune diseases and in the atherosclerotic process. JAK3 is a highly promising target for immunomodulatory drugs and polymorphisms in JAK3 gene have been associated with CV events in incident dialysis patients. Therefore, the aim of this study was to assess the potential role of JAK3 polymorphisms in the development of CV disease in patients with RA. 2136 Spanish RA patients were genotyped for the rs3212780 and rs3212752 JAK3 gene polymorphisms by TaqMan assays. Subclinical atherosclerosis was evaluated in 539 of these patients by carotid ultrasonography (US). No statistically significant differences were found when each polymorphism was assessed according to carotid intima-media thickness values and presence/absence of carotid plaques in RA, after adjusting the results for potential confounders. Moreover, no significant differences were obtained when RA patients were stratified according to the presence/absence of CV events after adjusting for potential confounders. Introduction. Rheumatoid arthritis (RA) is a polygenic disease associated with accelerated atherosclerosis and increased cardiovascular (CV) mortality. Recent studies have identified the ABO rs579459, PPAP2B rs17114036, and ADAMTS7 rs3825807 polymorphisms as genetic variants associated with coronary artery disease and the PIK3CG rs17398575 and EDNRA rs1878406 polymorphisms as the most significant signals related to the presence of carotid plaque in nonrheumatic Caucasian individuals. Accordingly, we evaluated the potential relationship between these 5 polymorphisms and subclinical atherosclerosis (assessed by carotid intima-media thickness (cIMT) and presence/absence of carotid plaques) and CV disease in RA. Material and Methods. 2140 Spanish RA patients were genotyped for the 5 polymorphisms by TaqMan assays. Subclinical atherosclerosis was evaluated in 620 of these patients by carotid ultrasonography technology. Results. Extragalactic astronomy.-- et al. ; The Andromeda galaxy (M 31) is one of a few galaxies that has sufficient angular size on the sky to be resolved by the Planck satellite. Planck has detected M 31 in all of its frequency bands, and has mapped out the dust emission with the High Frequency Instrument, clearly resolving multiple spiralarms and sub-features. We examine the morphology of this long-wavelength dust emission as seen by Planck, including a study of its outermost spiral arms, and investigate the dust heating mechanism across M 31. We find that dust dominating the longer wavelength emission (≳0.3 mm) is heated by the diffuse stellar population (as traced by 3.6 μm emission), with the dust dominating the shorter wavelength emission heated by a mix of the old stellar population and star-forming regions (as traced by 24 μm emission). We also fit spectral energy distributions for individual 5′ pixels and quantify the dust properties across the galaxy, taking into account these different heating mechanisms, finding that there is a linear decrease in temperature with galactocentric distance for dust heated by the old stellar population, as would be expected, with temperatures ranging from around 22 K in the nucleus to 14 K outside of the 10 kpc ring. Finally, we measure the integrated spectrum of the whole galaxy, which we find to be well-fitted with a global dust temperature of (18.2 ± 1.0) K with a spectral index of 1.62 ± 0.11 (assuming a single modified blackbody), and a significant amount of free-free emission at intermediate frequencies of 20–60 GHz, which corresponds to a star formation rate of around 0.12 M⊙ yr-1. Interstellar and circumstellar matter.-- et al. ; Within ten nearby (d < 450 pc) Gould belt molecular clouds we evaluate statistically the relative orientation between the magnetic field projected on the plane of sky, inferred from the polarized thermal emission of Galactic dust observed by Planck at 353 GHz, and the gas column density structures, quantified by the gradient of the column density, NH. The selected regions, covering several degrees in size, are analysed at an effective angular resolution of 10′ FWHM, thus sampling physical scales from 0.4 to 40 pc in the nearest cloud. The column densities in the selected regions range from NH≈ 1021 to1023 cm-2, and hence they correspond to the bulk of the molecular clouds. The relative orientation is evaluated pixel by pixel and analysed in bins of column density using the novel statistical tool called "histogram of relative orientations". Throughout this study, we assume that the polarized emission observed by Planck at 353 GHz is representative of the projected morphology of the magnetic field in each region, i.e., we assume a constant dust grain alignment efficiency, independent of the local environment. Within most clouds we find that the relative orientation changes progressively with increasing NH, from mostly parallel or having no preferred orientation to mostly perpendicular. In simulations of magnetohydrodynamic turbulence in molecular clouds this trend in relative orientation is a signature of Alfvénic or sub-Alfvénic turbulence, implying that the magnetic field is significant for the gas dynamics at the scales probed by Planck. OBJECTIVES: To determine whether the interleukin-33 (IL-33)-interleukin-1 receptor like 1 (IL-1RL1) signaling pathway is implicated in the risk of subclinical atherosclerosis in patients with rheumatoid arthritis (RA). METHODS: A total of 576 Spanish RA patients from Northern Spain were genotyped for 6 well-known IL33-IL1RL1 polymorphisms (IL33 rs3939286, IL33 rs7025417, IL33 rs7044343, IL1RL1 rs2058660, IL1RL1 rs2310173 and IL1RL1 rs13015714) by TaqMan genotyping assay. The presence of subclinical atherosclerosis was determined by the assessment of carotid intima-media thickness (cIMT) by carotid ultrasound (US). RESULTS: RA patients carrying the TT genotype of the IL33 rs3939286 polymorphism had lower cIMT values than those homozygous for the CC genotype (mean ± standard deviation (SD): 0.71 ± 0.14 mm versus 0.76 ± 0.16 mm, respectively) while patients carrying the CT genotype had intermediate cIMT values (mean ± SD: 0.73 ± 0.17 mm). Moreover, RA patients carrying the mutant allele T of the IL33 rs3939286 polymorphism exhibited significantly lower cIMT values than those carrying the wild allele C (mean ± SD: 0.72 ± 0.16 mm versus 0.75 ± 0.18 mm respectively; p = 0.04). The association of both genotype and allele frequencies of IL33 rs3939286 and cIMT levels remained statistically significant after adjustment for sex, age at the time of US study, follow-up and center (p = 0.006 and p = 0.0023, respectively), evidencing that the potential effect conferred by IL33 rs3939286 may be independent of confounder factors. No association with other IL33-IL1RL1 genetic variants was observed. In the years to come, the Javalambre-Physics of the Accelerated Universe Astrophysical Survey (J-PAS) will observe 8000 deg2 of the northern sky with 56 photometric bands. J-PAS is ideal for the detection of nebular emission objects. This paper presents a new method based on artificial neural networks (ANNs) that is aimed at measuring and detecting emission lines in galaxies up to zRlarr2 =Rlarr2 0.35. These lines are essential diagnostics for understanding the evolution of galaxies through cosmic time. We trained and tested ANNs with synthetic J-PAS photometry from CALIFA, MaNGA, and SDSS spectra. To this aim, we carried out two tasks. First, we clustered galaxies in two groups according to the values of the equivalent width (EW) of Hα, Hβ, [N II], and [O III] lines measured in the spectra. Then we trained an ANN to assign a group to each galaxy. We were able to classify them with the uncertainties typical of the photometric redshift measurable in J-PAS. Second, we utilized another ANN to determine the values of those EWs. Subsequently, we obtained the [N II]/Hα, [O III]/Hβ, and O 3N 2 ratios, recovering the BPT diagram ([O III]/Hβ versus [N II]/Hα). We studied the performance of the ANN in two training samples: one is only composed of synthetic J-PAS photo-spectra (J-spectra) from MaNGA and CALIFA (CALMa set) and the other one is composed of SDSS galaxies. We were able to fully reproduce the main sequence of star-forming galaxies from the determination of the EWs. With the CALMa training set, we reached a precision of 0.092 and 0.078 dex for the [N II]/Hα and [O III]/Hβ ratios in the SDSS testing sample. Nevertheless, we find an underestimation of those ratios at high values in galaxies hosting an active galactic nuclei. We also show the importance of the dataset used for both training and testing the model. Such ANNs are extremely useful for overcoming the limitations previously expected concerning the detection and measurements of the emission lines in such surveys as J-PAS. Furthermore, we show the capability of the method to measure a EW of 10 Å in Hα, Hβ, [N II] and [O III] lines with a signal-to-noise ratio (S/N) of 5, 1.5, 3.5, and 10, respectively, in the photometry. Finally, we compare the properties of emission lines in galaxies observed with miniJPAS and SDSS. BAFF, APRIL and BAFF-R are key proteins involved in the development of B-lymphocytes and autoimmunity. Additionally, BAFF, APRIL and BAFFR polymorphisms were associated with immune-mediated conditions, being BAFF GCTGT>A a shared insertion-deletion genetic variant for several autoimmune diseases. Accordingly, we assessed whether BAFF, APRIL and BAFFR represent novel genetic risk factors for Immunoglobulin-A vasculitis (IgAV), a predominantly B-lymphocyte inflammatory condition. BAFF rs374039502, which colocalizes with BAFF GCTGT>A, and two tag variants within APRIL (rs11552708 and rs6608) and BAFFR (rs7290134 and rs77874543) were genotyped in 386 Caucasian IgAV patients and 806 matched healthy controls. No genotypes or alleles differences were observed between IgAV patients and controls when BAFF, APRIL and BAFFR variants were analysed independently. Likewise, no statistically significant differences were found in the genotype and allele frequencies of BAFF, APRIL or BAFFR when IgAV patients were stratified according to the age at disease onset or to the presence/absence of gastrointestinal (GI) or renal manifestations. Similar results were disclosed when APRIL and BAFFR haplotypes were compared between IgAV patients and controls and between IgAV patients stratified according to the clinical characteristics mentioned above. Our results suggest that BAFF, APRIL and BAFFR do not contribute to the genetic network underlying IgAV. ; We are indebted to the patients and healthy controls for their essential collaboration to this study. Cytokines signalling pathway genes are crucial factors of the genetic network underlying the pathogenesis of Immunoglobulin-A vasculitis (IgAV), an inflammatory vascular condition. An influence of the interleukin (IL)33- IL1 receptor like (IL1RL)1 signalling pathway on the increased risk of several immune-mediated diseases has been described. Accordingly, we assessed whether the IL33-IL1RL1 pathway represents a novel genetic risk factor for IgAV. There is currently no approved treatment for primary Sjögren's syndrome, a disease thatprimarily affects adult women. The difficulty in developing effective therapies is -in part-because of the heterogeneity in the clinical manifestation and pathophysiology of the disease.Finding common molecular signatures among patient subgroups could improve our under-standing of disease etiology, and facilitate the development of targeted therapeutics. Here,we report, in a cross-sectional cohort, a molecular classification scheme for Sjögren's syn-drome patients based on the multi-omic profiling of whole blood samples from a Europeancohort of over 300 patients, and a similar number of age and gender-matched healthyvolunteers. Using transcriptomic, genomic, epigenetic, cytokine expression andflow cyto-metry data, combined with clinical parameters, we identify four groups of patients withdistinct patterns of immune dysregulation. A genetic component influences the development of atherosclerosis in the general population and also in rheumatoid arthritis (RA). However, genetic polymorphisms associated with atherosclerosis in the general population are not always involved in the development of cardiovascular disease (CVD) in RA. Accordingly, a study in North-American RA patients did not show the association reported in the general population of coronary artery disease with a series of relevant polymorphisms (TCF21, LPA, HHIPL1, RASD1-PEMT, MRPS6, CYP17A1-CNNM2-NT5C2, SMG6-SRR, PHACTR1, WDR12 and COL4A1-COL4A2). In the present study, we assessed the potential association of these polymorphisms with CVD in Southern European RA patients. We also assessed if polymorphisms implicated in the increased risk of subclinical atherosclerosis in non-rheumatic Caucasians (ZHX2, PINX1, SLC17A4, LRIG1 and LDLR) may influence the risk for CVD in RA. 2,609 Spanish patients were genotyped by TaqMan assays. Subclinical atherosclerosis was determined in 1,258 of them by carotid ultrasonography (assessment of carotid intima media thickness and presence/absence of carotid plaques). No statistically significant differences were found when each polymorphism was assessed according to the presence/absence of cardiovascular events and subclinical atherosclerosis, after adjustment for potential confounder factors.
