ABSTRACT - NECTAR 2018 Parkinson's Disease (PD) is characterized by dopaminergic neuronal loss in the substantia nigra (SN) resulting in a dopamine deficiency in the striatum.1 We developed a pathologically relevant rat model of PD using viral vectors to overexpress mutant alpha-synuclein.2 Our aims were to evaluate the dopaminergic changes and neuronal loss at different time-points in this model with two different presynaptic PET tracers, and relate imaging results with behavioural and histological changes. Therefor rats were unilaterally injected in the SN with AAV2/6 viral vector coding for mutated (A53T) human alpha-synuclein, and were studied at 6 or 12 weeks post-injection (wpi). PET imaging was performed using 6-[18F]fluoro-L-m-tyrosine3 (FMT), a substrate for AADC and [18F]-LBT9994 (LBT), a radioligand for dopamine transporter (DAT). Quantitative uptake images were calculated using Logan and Patlak graphical methods, with a cerebellar reference. At 6wpi we did not observe any asymmetry in the striatum; neither with FMT (n=5;p=0.421) nor LBT (n=6;p=0.310). At 12wpi we observed an asymmetry with LBT (n=4;p=0.003), but not with FMT (n=2;p=0.336). Histological evaluation showed 28% loss of TH-positive cells in the SN at 12wpi (n=8;p=0.031), but not at 6wpi (n=6;ns). A motor deficit was observed with the cylinder test at both time-points (n=7/8;p<0.001). Our results indicate that, in contrast to FMT, LBT parametric PET imaging is sufficiently sensitive to detect dopaminergic alterations in vivo in a rat model characterized by progressive and mild neurodegeneration of the nigro-striatal pathway. This PET tracer may be of particular interest to test neuroprotective or neuro-restorative therapeutic strategies in future studies using our PD rat model. References: 1. "Die multiple Imputation ist ein Verfahren zum Umgang mit fehlenden Werten bei der Datenanalyse, also dann, wenn die fehlenden Werte bereits entstanden sind. Unter multipler Imputation wird die Ergänzung (Imputation) fehlender Werte durch mehrere (multiple) 'plausible' Werte unter einem für die Daten geeigneten Modell verstanden. Multiple Imputationen können als eine Abfolge von drei Schritten dargestellt werden: Im Imputationsschritt werden für jeden fehlenden Wert D>1 Werte generiert und damit D>1 vervollständigte Datensätze erstellt, die keine fehlenden Werte mehr enthalten. Im Analyseschritt werden die D vervollständigten Datensätze getrennt mit Verfahren zur Analyse kompletter Daten ausgewertet. Im Integrationsschritt werden die Ergebnisse der D Einzelanalysen zu einer gemeinsamen Schätzung (Punktschätzer und Standardfehler) kombiniert. Der theoretisch und rechentechnisch anspruchsvollste Schritt ist die Erzeugung korrekter Imputationen. Wichtig ist, dass es sich bei den ergänzten Werten nicht um ad hoc, beliebig oder willkürliche eingesetzte Werte handelt, sondern um Werte, die unter einem spezifizierten Modell für die Daten auf Basis der beobachteten Werte wahrscheinlich sind. Multiple Imputationen sind dem listenweisen Ausschluss von fehlenden Werten (Listwise Deletion) überlegen, weil letzterer eine restriktivere Annahme über die fehlenden Werte trifft." Die immer umfangreichere Datenflut steht in einem symbiotischen Verhältnis zum Leitgedanken der Transparenz: Einerseits führt die Akkumulation von Daten zur Forderung, diese auch öffentlich zugänglich zu machen. Umgekehrt werden immer mehr Daten produziert, um Transparenz herzustellen. Üblicherweise sieht man die Herausforderung im Umgang mit den Datenströmen in der Kunst des Differenzierens: Transparenz und Datenerfassung sollen dort zum Einsatz kommen, wo es sinnvoll ist (z.B. im Open Government), sind aber abzulehnen, wo sie sich als problematisch zu erkennen geben (etwa in der naiven Ausrufung eines Zeitalters der Post-Privacy). In den letzten Jahren irritieren jedoch vermehrt Stimmen, die den Leitgedanken der Transparenz grundsätzlich in Frage stellen: so etwa das französische Autorenkollektiv Tiqqun (2007) oder der Philosoph Byung-Chul Han (2012). Kommt hier einfach nur Fortschrittsfeindlichkeit zum Ausdruck? Welche Überlegungen stehen hinter der Zurückweisung des Transparenzparadigmas?

Die Autoren untersuchen die Funktionsweise von Datenmärkten. Besonderes Augenmerk wird dabei auch auf Substitute zu einem "Handel mit Daten" im engeren Sinne gelegt. Die Unterscheidung zwischen personenbezogenen und nicht personenbezogenen Daten ist für die Analyse der Funktionsweise und möglicher Funktionsdefizite von Datenmärkten von zentraler Bedeutung. Die Funktionsweise von Märkten für personenbezogene Daten kann insbesondere durch das Recht auf Datenportabilität (Art. 20 DSGVO) nachhaltig geprägt werden. Dies hängt allerdings von der konkreten Ausgestaltung des Rechts auf Datenportabilität ab. Funktional kann dieses Recht unter anderem als eine Abhilfe für eine durch das Datenschutzrecht bedingte partielle Marktbehinderung verstanden werden. Die Einführung eines - allerdings nur bei Marktmachtlagen zwingenden - Rechts auf Datenportabilität kann auch für nicht personenbezogene Daten sinnvoll sein. Es würde dann auf eine andere Form von Marktversagen reagieren. Im Übrigen sehen die Autoren weder bei personenbezogenen noch bei nicht personenbezogenen Daten einen gesetzgeberischen Handlungsbedarf zur allgemeinen Förderung des Datenhandels. ; The authors examine the functioning of data markets. Particular attention is paid to substitutes for "data trading" in a narrow sense. In the analysis of the functioning and possible functional deficiencies of data markets, it is necessary to distinguish between markets for personal data and markets for non-personal data. In the functioning of markets for personal data, the right to data portability (Article 20 of the GDPR) may play an important role. Functionally, the right to data portability can be understood as a remedy for the restrictions to trade that follow from data protection law. There may be a good case for introducing a right to data portability also for non-personal data. However, it would react to a different form of market failure and should be mandatory only in the presence of market power. Apart from this, the authors currently do not see a need for legislative action to promote data trading - this holds for personal as well as for non-personal data.

In today's fast-paced business environment, leaders who fail to recognize and respond to the emotions of their employees can result in stagnation or a lack of growth in their organization. In light of this, the current study presents a method for identifying leadership styles based on the perspectives of employees, exploring their perceptions of specific leadership traits. Drawing upon the Multifactor Leadership Questionnaire framework, this study aims to identify the leadership style of Tony Wedding Studio by conducting in-depth interviews with its employees. Through sentiment and cluster analyses of the collected data, the study identifies Idealized Influence Attributes and Laissez-faire as key leadership style traits. Additionally, this study sheds light on how employees react to and feel about their leader's leadership style, providing valuable insights for organizational development and leadership training. Overall, this study emphasizes the importance of understanding and responding to employee perspectives in effective leadership.
