▪ Abstract  Nucleic acids are preserved in prehistoric samples under a wide range of depositional environments. The development of new molecular methods, especially the polymerase chain reaction, has made possible the recovery and manipulation of these molecules, and the subsequent molecular genetic characterization of the ancient samples. The analysis of ancient (a)DNA is complicated by the degraded nature of ancient nucleic acids, as well as the presence of enzymatic inhibitors in aDNA extracts. We review aspects of ancient DNA preservation, a variety of methods for the extraction and amplification of informative DNA segments from ancient samples, and the difficulties encountered in documenting the authenticity of ancient DNA template. Studies using aDNA to address questions in human population history or human evolution are reviewed and discussed. Future prospects for the field and potential directions for future aDNA research efforts in physical anthropology are identified.

Ancient DNA Typing is a comprehensive manual about the analysis of ancient and degraded DNA. The book contains a detailed protocol chapter which serves as a lab manual with the most important methods for DNA extraction, PCR and electrophoretic analysis of degraded samples. The appendix helps to set up a PCR lab specializing in ancient DNA research and answers the most frequent questions concerning technical problems in investigations of degraded DNA

Evolutionary processes, including selection, can be indirectly inferred based on patterns of genomic variation among contemporary populations or species. However, this often requires unrealistic assumptions of ancestral demography and selective regimes. Sequencing ancient DNA from temporally spaced samples can inform about past selection processes, as time series data allow direct quantification of population parameters collected before, during, and after genetic changes driven by selection. In this Comment and Opinion, we advocate for the inclusion of temporal sampling and the generation of paleogenomic datasets in evolutionary biology, and highlight some of the recent advances that have yet to be broadly applied by evolutionary biologists. In doing so, we consider the expected signatures of balancing, purifying, and positive selection in time series data, and detail how this can advance our understanding of the chronology and tempo of genomic change driven by selection. However, we also recognize the limitations of such data, which can suffer from postmortem damage, fragmentation, low coverage, and typically low sample size. If most academic debates surrounding the recent boom of ancient DNA (aDNA) so far have concerned conflicting research epistemologies, this article is a call for taking aspects of media and communication more seriously. Analyzing the fates of two recent research papers on Viking Age Scandinavia, we show how aDNA research is communicated, narrated and infused with meaning in the public sphere, particularly in relation to popular narratives and political debates. We observe significant interlacing of scientific, political and media discourses in and around the papers, and conclude that archaeogenetics is a highly mediatized scientific field. ; Code, Narrative, History: Making Sense of Ancient DNA in Contemporary Culture

The integration of massively parallel sequencing (MPS) technology into forensic casework has been of particular benefit to the identification of unknown military service members. However, highly degraded or chemically treated skeletal remains often fail to provide usable DNA profiles, even with sensitive mitochondrial (mt) DNA capture and MPS methods. In parallel, the ancient DNA field has developed workflows specifically for degraded DNA, resulting in the successful recovery of nuclear DNA and mtDNA from skeletal remains as well as sediment over 100,000 years old. In this study we use a set of disinterred skeletal remains from the Korean War and World War II to test if ancient DNA extraction and library preparation methods improve forensic DNA profiling. We identified an ancient DNA extraction protocol that resulted in the recovery of significantly more human mtDNA fragments than protocols previously used in casework. In addition, utilizing single-stranded rather than double-stranded library preparation resulted in increased attainment of reportable mtDNA profiles. This study emphasizes that the combination of ancient DNA extraction and library preparation methods evaluated here increases the success rate of DNA profiling, and likelihood of identifying historical remains.
