Un approccio multidisciplinare per l'identificazione dello stock del nasello (Merluccius merluccius, Linnaeus 1758) nei mari sardi
The "stock" concept is fundamental to fisheries management. To manage a fishery responsibly it is important to understand its stock structure and how fishing effort and mortality are distributed (Begg e Waldman, 1999). The failure to comply the true stock structure and the ineffective fisheries management of an exploited species can lead to overfishing and depletion of the stock (Begg et al., 1999a). In the last years, scientific community has pointed out that the most successful way to address stock identification studies is the application of a holistic approach, thus involving a broad selection of complementary techniques (Begg et al., 1999a; Abaunza, 2008a; Cadrin et al., 2013). The European hake (Merluccius merluccius, Linnaeus 1758) represents a basic component of the demersal communities and one of the target species in the Mediterranean Sea (Lo Brutto et al., 2004; Ligas et al., 2011). For this reason it has been extensively studied. Furthermore, hake stocks are generally considered over-exploited both in the Atlantic Ocean (ICES, 2011; 2012) and in the Mediterranean Sea (STECF, 2013; GFCM, 2014). At present, European hake populations in the Mediterranean Sea are managed considering several relatively small geographical sub-areas (GSAs), which are identified mainly on the basis of political considerations (Ragonese et al., 2012; Vittori et al., 2013). In this partition, Sardinian seas are represented by GSAs 11.1 and 11.2 and are split between two FAO Statistical Divisions (Balearic Islands and Sardinia). It is known that the complexity of many marine species stocks is often underestimated in modern stock management processes (Begg et al., 1999a). As regards the Mediterranean sea, various studies suggested a geographical distribution of the stocks that appears considerably different from the current management units partition (Abaunza et al., 2000; Abaunza et al., 2008c; Murta et al., 2008), and this fact have been observed also for the European hake (Castillo et al., 2004; Mattiucci et al., 2004; Tanner et al., 2012; Fiorentino et al., 2014). The need of the achievement of a comprehensive knowledge about the stock structure of a commercially important species like M. merluccius, has led to the development of this study, which aims, through the employment of a multidisciplinary approach, at a better geographical definition of this species stock in the waters around Sardinia. A first insight into the complexity of the European hake population in the area has been supplied by the analysis and comparison of its length structure in the western and eastern coasts of Sardinia, showing a degree of differentiation that has been subsequently deepened through the application of various techniques. The analysis of the variability in the growth of the species hasn't indicated a meaningful differentiation of the populations in the two sides of the island, but has suggested that the presence of many spawning areas in the western coast could be the cause of the difference in the 0 age-class mean length obtained, as well as in the length structure above-mentioned. On the contrary, the implementation of a geometric morphometric technique to study the shape of individuals has led to the discovery of a significant, even though of moderate extent, differentiation between the two coasts. However, as remarked by Tudela (1999) and Levi et al. (2004) in other case-studies, these differences were believed to be linked only to environmental influences, without a connection to those biological parameters that are fundamental in the definition of a stock. This hypothesis has been made also considering that the observed difference in shape didn't seem to affect individuals' growth. Lastly, the parasitological exam of specimens collected in the whole area of Sardinian seas, has supported the hypothesis of homogeneity of the populations in the area. Indeed, no significant differences have been found in the relative abundance and prevalence of the various parasite taxa examined. Comparing these results with those obtained in a similar study carried out in the Mediterranean Sea (Mattiucci et al., 2004) it has been possible to highlight a considerable similarity of our study area with the central area of Mediterranean sea, and, conversely, a clear differentiation between Sardinian seas and the eastern part of Mediterranean basin. Similar considerations has been made in the context of StockMed project (Fiorentino et al., 2014). This study has therefore provided a solid evidence of the homogeneity of the European hake stock in the waters around Sardinia, leading to the conclusion that this population should be managed jointly. Moreover, it has supported the recently expressed opinion that its stock (as well as those of other important species) could be wider than expected, underlining the need of improving current management units boundaries.

Studio della dinamica di sfruttamento di Merluccius merluccius (Linneo, 1758) nel Mar Tirreno settentrionale in relazione alle politiche di gestione nel settore della pesca
Lo sfruttamento eccessivo delle risorse alieutiche ha portato negli anni molti stock ittici in situazioni critiche e prossime al collasso. Si sono susseguiti una serie di regolamenti sia nazionali che comunitari, rivolti a una migliore gestione delle risorse. Questo lavoro di tesi si inserisce all'interno del progetto europeo MINOUW (Minimizing unwanted catches in European fisheries) nell'ambito degli obiettivi di Horizon 2020, volto a incoraggiare l'utilizzo di tecniche che riducano le catture indesiderate. L'obiettivo è quello di valutare l'efficacia delle politiche di gestione messe in atto per la riduzione dello scarto e la tutela dei giovanili. Come specie target è stato scelto il nasello, Merluccius merluccius, come area di riferimento la GSA 9, che comprende il Mar Ligure e il Mar Tirreno settentrionale e come indicatore i quantitativi di scarto. I dati raccolti provengono da diverse campagne di ricerca, effettuate dal 1995 sino al 2016. I campioni di nasello provengono sia da acquisti presso i mercati ittici sia da campionamenti effettuati a bordo delle imbarcazioni commerciali. In laboratorio sono stati determinati i parametri biologici di taglia (lunghezza totale LT), peso (al decimo di grammo), sesso e stadio maturativo delle gonadi. I dati sulle catture sono stati standardizzati all'ora di pesca e sono stati utilizzati per realizzare gli indici di biomassa e abbondanza, sono state poi effettuate ulteriori elaborazioni statistiche con il pacchetto R tramite la funzione GAM (Generalized Additive Models) su quattro variabili spazio-temporali (profondità, durata della cala, stagione e periodo). Le distribuzioni di frequenza delle catture sono state scomposte nella frazione commercializzata e scartata e ne è stata calcolata la L50 di scarto. Tramite l'analisi delle taglie catturate è emerso come la prima taglia a comparire nella rete vari tra 3,5 e 5,5 cm. In seguito alle politiche di gestione è aumentata negli anni la taglia media sia della frazione scartata, che quella commercializzata. Questo andamento è evidenziato anche dall'aumento della L50 di scarto che passa da 10,69 cm nel 1995 a 17,48 cm nel 2016. Tuttavia, lo scarto del nasello è ancora elevato, in termini di biomassa oscilla tra il 10 e il 50% delle catture totali mentre in termini di abbondanza i valori sono nettamente più alti variando tra il 30 e il 90%, inoltre sul mercato sono ancora presenti esemplari inferiori alla taglia minima di sbarco. Tramite l'analisi GAM delle variabili spazio-temporali è emerso come i tassi di scarto siano maggiori alle profondità comprese tra 150 e 200 m e nel periodo estivo-autunnale. Da questo lavoro emerge come le politiche che si sono susseguite nel corso degli anni non abbiano portato un effettivo vantaggio per la specie in termini di riduzione della mortalità dovuta alla pesca, in quanto i tassi di scarto non sono stati ridotti.

DNA-based methods for the control of seafood frauds in Italy and China
The globalization of the seafood supply chain, together with the difficulty in visual identification of prepared seafood products, makes quite easy to commit frauds by substituting high-value species with less expensive ones. The traceability systems implemented at international level should be the answer to solve this issue and the accurate labeling of seafood species throughout the supply chain has become quite important for the protection of the consumers. Currently, the European Union (EU) has established a legal framework for the management of seafood label (Reg. No. 1379/2013), while in China, despite its increasingly important role in the world's seafood market, a specific vertical regulation for fishery product traceability and an official system for the assignment of the trade names are still not available. Meanwhile, it is also necessary to develop rapid, economic and highly sensitive methods for the identification of seafood species. DNA-based methods are the most applied for species identification due to the high resistance of nucleic acid to physical-chemical treatments that allow to recover diagnostic fragment of DNA even in processed products. However, the most appropriate DNA based method to be used should be selected according to the species and the kind of products to be analyzed. In this perspective, the research activities were aimed: (I) to develop a simple and cost-effective bead-milling method for DNA extraction from fish muscles, (II) to develop a DNA-based method to verify the labeling accuracy in the light of the European provisions, (III-IV) to investigate the phenomenon of species substitution in China, (V) to assess the possible implementations of the Chinese labeling system through the introduction of official denominations, in light of EU. The bead-based method developed in the first work allowed to obtain acceptable amounts of DNA, with good purity and amplificability. Although the salting-out method has been confirmed as the most effective protocol in terms of pure performances, the bead-milling procedure can be considered a valid alternative. In the second work the analyisis of canned pet food commercialized on the EU market and claiming to contain high price ingredients was performed using Basic Local Alignment Search Tool (BLAST) and and Forensically Informative Nucleotide Sequencing (FINS). An impressive rate of mislabeling (100% for whole minnows and 40% for tuna fillets) was highlighted. DNA and mini-DNA barcoding technique was applied for the identification of commercial products sold with the Chinese term 鳕(Xue, Cod) in supermarkets (Nanjing and Shanghai) and in the online market in China. The mislabeling rate was assessed according to three increasingly stringent definitions due to the lack of harmonization around the definition of Cod in China. A very high mislabeling rate, which exceeded 60% even with the less stringent definition, was highlighted. The same technique applied for the identification of fish products sold on the Chinese e-commerce market as Anoplopoma fimbria highlighted an alarming overall misrepresentation rate of 85,7%. Finally, through the analysis of the commercial designations officially adopted by 5 European Member States (MSs) (Italy, France, United Kingdom (UK), Germany and Spain) and of those adopted on international lists of Chinese names such as the FAO Fisheries and Aquaculture Statistics and Information Service (ASFIS), FishBase and the Latin-Chinese Dictionary of Fish Names (LCDFN) for Cod, Salmon and Tuna species, possible implementations of the Chinese labeling system through the introduction of official denominations were suggested.

Norovirus in bivalve molluscs: development of a Real Time RT PCR protocol and its application for a viral contamination monitoring and for a bioaccumulation study
The Food Hygiene Package is a body of European Regulations laying down hygiene rules for foodstuffs produced in the EU and non-EU countries exporting to the EU. The pursuit of a high level of protection of human life and health is one of the fundamental objectives of this laws package. Some points of EC Regulations undergo to critical review by the Commission, and particular attention was given to fishery products. Commission Regulation (EC) No 2073/2005 of 15 November 2005 on microbiological criteria for foodstuffs, in Annex I, Chapter 1, lays down the food safety criteria for live bivalve molluscs and live echinoderms, tunicates and gastropods and sets sampling-plans, limits and analytical reference methods only concerning bacterial micro-organisms of Salmonella spp. and Escherichia coli genera. Epidemiological data of the last years reported the consumption of live bivalve shellfish infected by enteric viruses as common cause of human gastroenteritis. Norovirus resulted the leading cause of all human gastroenteritis outbreaks worldwide. For this reason, in the first part of Reg. (EC) No 2073/2005, in the whereas at point 12, it's written that the Scientific Committee on Veterinary Measures relating to Public Health (SCVPH) issued an opinion on Norwalk-like viruses (NLVs, noroviruses) on 30-31 January 2002. In that opinion it concluded that the conventional fecal indicators are unreliable for demonstrating the presence or absence of NLVs and that the reliance on fecal bacterial indicator removal for determining shellfish purification times is unsafe practice. So Salmonella and Escherichia coli can not be used as only indicators of safety criteria in live bivalve molluscs. In particular, at following points we can read that it may be necessary to set harmonized sampling frequencies at Community level, particularly in order to ensure the same level of controls to be performed throughout the Community. Finally it's confirmed that criteria for pathogenic viruses in live bivalve molluscs should be established when the analytical methods are developed sufficiently (at point 27). In response to concerns expressed by EU Regulations and taking into account the latest epidemiological data, the international networks dealing prevention, communication and control of human food borne illness diseases caused by enteric viruses have intensified their researches on norovirus. In the past few years, numerous research projects financed by government departments and international organizations have been implemented in order to get more information on these viral pathogens and check most hazardous areas. Among the areas traditionally known for a high fish consumption, there are the Southern Italy regions, where mussels are highly appreciated. Mytilus galloprovincialis accounts for over 22.4% of annual consumption of fish (data by E.U., Unimar, Ismea e Uniprom), particularly in Campania the mussels consumption was calculated to be about 41 000 tonnes each year. Since ancient times, in most coastal areas the custom of eating raw or undercooked bivalve shellfish remains common. In this way, however, the assumption of norovirus eventually present and bioaccumulated in shellfish makes easier. Noroviruses are highly resistant to adverse environmental conditions and a simple shellfish cleaning or the application of so-called "mild technologies", such as steaming, are unable to eliminate viral contamination. Among filter feeders bivalve, oysters are the species most common in France. In fact the French coastal areas produce more than 90% of oysters in the EU and France has the historic first for production and consumption of these molluscs. Oysters are traditionally eaten raw, still alive, mostly with a few drops of lemon juice. As for Mytilus galloprovincialis, oysters can represent an hazard for the occurrence of human food borne illness from enteric viruses. In the course of my PhD studies, I have dealt with norovirus in bivalve molluscs. During my first year of PhD study, I concurred to the development of a method for detection of norovirus in live bivalve molluscs working together with researchers of the ISS. The method is based on the use of Real Time RT PCR. During our researches it has been subjected to inner validation by the ISS and it resulted provided of efficiency to research norovirus in live bivalve molluscs. So the Italian Ministry of Health with Note of 24/11/2009 established the one-step Real Time RT-PCR protocol as the official method to research norovirus in live bivalve molluscs. After this period, during the second year of my PhD studies, I worked for a national monitoring to test norovirus presence in bivalve molluscs. The one-step Real Time RT PCR protocol was used again. Studies have been carried out within a project financed by Campania Region. Shellfish collected from harvesting areas and bought at retailers located on the three coastal districts of Naples, Caserta and Salerno have been tested for norovirus presence. During the last year, my scientific experience has been enriched by a collaboration with IFREMER researchers. IFREMER is one of the French national research centers taking part in FBVE-network. I have worked at the section of the Virology of Laboratory of Microbiology (MIC) sited in Nantes, that was indicated National Reference Laboratory (NRL) for the control of bacterial and viral contamination of bivalve molluscs in France by the French Ministry of Agriculture and Fisheries. IFREMER has the same functions of the Italian NRL concerning norvirus research and control. During my stage at IFREMER, bioaccumulation physiological activity was analyzed in oysters living in seawater contaminated by Norovirus spp. The research was carried out on samples of Crassostea gigas coming from different harvesting areas located along the Atlantic coast of Brittany, area known for oysters production worldwide. For bioaccumulation studies classified and quantized strains of norovirus GI.1 and GII.3 were used. At the IFREMER laboratory, the one-step RT Real Time PCR protocol was used and further validated.

Analisi del Sanitary Survey 2015-2017 condotto nel Golfo della Spezia: riclassificazione delle zone di produzione di molluschi bivalvi
Introduzione Il consumo di molluschi bivalvi (MB) crudi o poco cotti può rappresentare un rischio per la presenza di microrganismi patogeni derivanti da contaminazione fecale. La normativa comunitaria prevede che i produttori possano raccoglierli soltanto nelle zone di produzione classificate dall'autorità competente (AC) come A, B o C in relazione al livello di contaminazione fecale, con Escherichia coli come indicatore (Reg. CE n. 853/2004 e Reg. CE n. 854/2004). Per classificare le zone di produzione l'AC deve effettuare un "sanitary survey" per valutare le fonti di inquinamento e istituire un programma rappresentativo di campionamento dei MB nella zona considerata. Oltre alla valutazione di parametri biologici, chimici e fisici cogenti, l'AC può altresì valutare parametri aggiuntivi ai fini di una completa sorveglianza sanitaria. L'AC deve inoltre programmare attività di monitoraggio, generalmente a cadenza trimestrale, per verificare se il livello di rischio sia cambiato e la zona debba essere riclassificata. Il "Centro per le scienze dell'ambiente, della pesca e dell'acquacoltura" (CEFAS) ha pubblicato nel 2017 delle Linee Guida aggiornate per l'esecuzione dei sanitary surveys. Una serie di audit effettuati su undici Stati Membri nel periodo 2011-2013 dal Food and Veterinary Office (FVO) ha tuttavia evidenziato sostanziali carenze nei sistemi di riclassificazione. Nel 2011, nessuna zona di produzione era stata ancora riclassificata in Italia. La regione Liguria ha quindi intrapreso il primo sanitary survey relativo al triennio 2012-2014 per le zone di produzione di MB del golfo della Spezia. Successivamente, è stato effettuato il sanitary survey, relativo al triennio 2015-2017. Questo studio è volto ad analizzare i dati provenienti dal report "sanitary survey 2018: riclassificazione delle zone del golfo adibite alla molluschicoltura" relativo al triennio 2015-2017, elaborato dalla Struttura Igiene degli Alimenti di Origine Animale dell'ASL 5 Spezzino. Summary Consumption of raw or insufficiently cooked bivalve molluscs can result in illness due to the presence of microorganisms, notably algal biotoxins and viral contamination arising from human faecal pollution (EFSA, 2015). As established by EU legislation, gatherers may only harvest LBM from production areas with fixed locations and boundaries that the competent authority (CA) has classified as being of class A, B or C according to the level of faecal contamination and using Escherichia coli as indicator organism (Regulation CE n. 853/2004 and Regulation CE n. 854/2004). In order to classify a production area, the CA implements a sanitary survey aimed at evaluating the sources of contamination in the catchment area and establishing a sampling programme of LBM ensuring that the results of the analysis are as representative as possible for the considered area. Ongoing monitoring (generally of a three-annual basis) are also needed to determine whether the level of risk has changed, and the classification status should be therefore modified. In 2017, the Centre for Environment Fisheries and Aquaculture Science (CEFAS) published an updated Community guide providing principles and operational guidelines that should be applied by the CA to practically perform the sanitary survey. Despite of this legislative background, a series of audits undertaken between 2011 and 2013 by the European Commission's Food and Veterinary Office (FVO) on eleven Member States highlighted consistent gaps in the classification systems of LBM production areas. According to the official data, no re-classification procedures had been performed yet in 2011 within the entire Italian territory. Given the necessity to fulfil EU requirements, the Liguria Region implemented its first sanitary survey on the production areas of the gulf of La Spezia, which covered the three years 2012-2014 and whose results were reported in a final report published in 2015. The procedures for the sanitary survey concerning the subsequent three-year period were initiated in 2015 and the relative final report was published in 2018. The aim of this study was to analyse the data from the final report of the sanitary survey concerning the three years 2015-2017 developed by ASL 5 Spezzino and published in 2018. Materiali e Metodi Sono stati estrapolati dati relativi al numero e alla tipologia di analisi effettuate sulle specie Mytilus galloprovincialis, Crassostrea gigas e Venus verrucosa nei punti di campionamento previsti dal piano di monitoraggio e al numero e alla tipologia di non conformità (NC) riscontrate sulla base di parametri cogenti e parametri aggiuntivi di sorveglianza sanitaria. I dati sono stati confrontati con quelli del sanitary survey relativo al triennio precedente (2012-2014). Opportuni test statistici sono stati utilizzati per valutare i dati provenienti dal monitoraggio di E. coli e Norovirus. Risultati e Discussione Sono state effettuate 4306 analisi, soprattutto su M. galloprovincialis (89%) e per lo più su agenti biologici e biotossine marine. Sono state rilevate 160 NC, la maggior parte delle quali (93.7%) riferibili a positività per Norovirus in M. galloprovincialis e C. gigas, con una forte prevalenza del genogruppo GII. La prevalenza di Norovirus nel golfo della Spezia è risultata essere tra le più elevate a livello nazionale. Al contrario, nonostante alcune NC riscontrate per le biotossine marine, lo stato sanitario delle zone di produzione del golfo è risultato relativamente sicuro rispetto ad altre realtà nazionali. È stato dimostrato che i livelli di E. coli sono correlati sia con la piovosità che con la stagionalità (maggiori nel periodo più freddo). In entrambi i casi sono principalmente coinvolti i punti di campionamento interni alla diga e della Baia di Portovenere, da attribuire principalmente alla direzione delle correnti marine ed al loro rallentamento nei suddetti punti. Rispetto al confronto con il triennio precedente, è stata riconfermata la classe B per M. galloprovincialis, le zone di produzione di C. gigas sono state riclassificate A e quelle di V. verrucosa sono invece state definitivamente chiuse per le difficoltà riscontrate nel reperimento di un numero di campioni rappresentativo. Il sanitary survey si è confermato uno strumento utile nella riclassificazione e nel monitoraggio sanitario delle zone di produzione di MB.

Dottorato di ricerca in Ecologia e gestione delle risorse biologiche ; La crescente richiesta di energia elettrica per il funzionamento delle apparecchiature elettroniche di uso quotidiano, sia in ambito civile che industriale, ha provocato negli ultimi decenni il proliferare di centrali termoelettriche, con conseguente aumento della potenza di produzione e delle dimensioni delle singole unità produttive. La necessità di raffreddare i condensatori e, quindi, l'esigenza di disporre di ingenti quantitativi d'acqua atti allo scopo, ha reso sempre più pressante il problema di reperire siti idonei per l'istallazione delle centrali, motivo per cui la maggior parte di esse sono state collocate presso la costa, con evidenti vantaggi di presa di acqua e smaltimento del calore residuo direttamente a mare. Spesso non si è tenuto in debito conto che, l'utilizzo di notevoli masse di acqua, potesse provocare problemi ambientali, non solo come impatto sulle biocenosi nel bacino ricevente il refluo termico, ma anche con una serie di effetti nocivi sugli organismi associati al volume d'acqua prelevata, quasi sempre con conseguenze mortali. Studi sugli impatti degli scarichi di reflui caldi sulle biocenosi, hanno dimostrato che gli organismi viventi sottoposti all'effetto del "pennacchio caldo" possono subire shock termici dovuti a repentini cambiamenti di temperatura in un tempo così breve da non consentirne l'acclimatazione. Questi impatti, sono in grado di provocare cambiamenti sulle risorse trofiche dei pesci, anticipazione nel raggiungimento della maturità sessuale, aggregazione massiva di individui nelle aree di scarico, riduzione della ricchezza specifica della comunità. Per quanto riguarda i volumi di acqua prelevati dall'ambiente per i processi di raffreddamento delle centrali termoelettriche, l'impossibilità di resistere alla forza con cui l'acqua viene pompata nell'impianto, fa sì che un certo numero di organismi (pesci, molluschi e organismi planctonici) associati alla massa in ingresso, vengano trascinati verso l'interno della struttura. Gli organismi di taglia maggiore si arrestano contro le grate metalliche dei filtri, dove subiscono effetti dannosi dipendenti dall'esaurimento fisico, dal soffocamento e dall'impatto meccanico contro le barriere, tale fenomeno viene definito "impingement". Gli organismi di piccole dimensioni (fra cui larve e uova di pesci), vengono trascinati all'interno delle strutture di raffreddamento, fenomeno di "entrainment", dove subiscono abrasioni meccaniche, intossicazione per la presenza di biocidi e stress legati all'aumento di temperatura ed a cambiamenti di pressione. L'impatto delle centrali elettriche sulle comunità ittiche ha riscosso particolare attenzione nel Nord America ed in Canada, dove l'industria idroelettrica ha una lunga tradizione e l'entità delle ricadute economiche per il depauperamento di specie commerciali sono notevoli. In Italia la presa e restituzione di acqua per usi industriali è regolamentata dal Decreto Legislativo n.152 del 3 Aprile 2006 (Gazzetta Ufficiale n.88) anche se, nel detto decreto, non si fa cenno agli organismi viventi associati alla massa d'acqua utilizzata. Inoltre nonostante la maggior parte delle centrali Italiane siano collocate lungo la costa, non risultano studi sperimentali volti all'individuazione di un impatto sulle biocenosi, conseguente la presa di acqua marina. E' in tale ambito che è nata l'idea progettuale, indirizzata alla verifica di eventuali impatti sulla fauna ittica relativi alla presa e restituzione di acqua marina per il raffreddamento dei condensatori della centrale termoelettrica A. Volta nel Tirreno centrale. L'idea, seppur innovativa e mai sperimentata in Italia, non teneva in considerazione il sopraggiungere della crisi economica che ha, di fatto, dal 2008 ad oggi provocato la riduzione del quantitativo annuale di produzione elettrica della centrale, in conseguenza della chiusura in tutto il territorio nazionale di numerosi esercizi commerciali e fabbriche, con successiva minor richiesta energetica. L'altalenante funzionamento della centrale e quindi il ridotto prelievo di acqua marina, ha aumentato enormemente i livelli di variabilità del sistema, diminuendo al contempo la disponibilità programmatica per i campionamenti e la presenza costante di un refluo termico a mare consistente. Si è quindi deciso di concentrare le attività di ricerca nella valutazione del fenomeno dell'entrainment sugli stadi precoci dei teleostei, ricerca che presenta il maggior indice di innovazione, non essendo mai stata sperimentata in nessuna centrale Italiana e di cui non ci risulta esistano lavori analoghi in tutto il Mediterraneo. L'analisi ha riguardato la messa a punto di un sistema di campionamento particolare, dovendo operare all'interno delle strutture artificiali dell'impianto, con la scelta degli attrezzi da pesca più idonei, l'individuazione dell'area di prelievo, il lume della maglia più adatto ed i tempi di cala più redditizi, cioè in grado di armonizzare abbondanza di catture e rigurgito dovuto ad intasamento delle maglie. I campionamenti con reti da plancton per la componente ittioplanctonica e le uova sono state eseguite nei canali interni alla centrale nell'area antecedente i filtri. Le attività a mare invece, hanno riguardato il campionamento con sciabica da terra nell'area interessata dal refluo termico ed in due aree controllo a nord e sud dello stesso, per la cattura dei giovanili in fase d'insediamento. Le attività di campionamento interne alla centrale sono state condotte dal 2009 al 2011, con cadenza stagionale nei primi due anni. Durante le 384 ore di lavoro sono stati filtrati un totale di 235.574,5 m3 di acqua marina, suddiviso in 236 cale, la maggior parte delle quali nel 2009. Alcune cale sono state effettuate preliminarmente nel 2008, al fine di definire il disegno di campionamento per gli anni successivi ed individuare l'area di prelievo, la tipologia di rete adatta e standardizzare i tempi di cala più idonei. Le attività di smistamento e determinazione tassonomica hanno portato complessivamente alla raccolta di 668 individui suddivisi in 24 taxa e 2.459 uova di teleostei. Lo studio ha dimostrato, per il sistema di raffreddamento della centrale A. Volta, l'esistenza di un impatto sugli stadi precoci dei teleostei, in conseguenza del fenomeno di entrainment, con un prelievo stimato per l'anno 2009 di circa 12 x 106 larve e 24,5 x 106 uova, le sardine e le alici sono risultate le specie maggiormente impattate dalla centrale. Il peso stimato, circa 300 Kg, delle larve di sardina prelevate dalla centrale nel corso del 2009, è stato relazionato con i dati di cattura del bianchetto in Liguria. Il dato corrisponde a 50 giorni barca per il compartimento di Genova e 31 giorni barca per il compartimento di Savona, con una perdita economica che va dai 9.000,00 euro (vendita all'ingrosso) ai 15.000,00 euro (vendita al dettaglio). L'analisi statistica mediante PERMANOVA ha inoltre constatato differenze significative fra le comunità ittioplanctoniche nelle varie stagioni per i valori di abbondanza, mentre non sono risultati significativamente differenti i dati stagionali per numero di uova. L'estate si è confermata come il periodo con i valori di diversità, ricchezza specifica ed equitabilità più elevati, mentre l'inverno ha registrato il maggior numero di esemplari campionati. La relazione fra il numero di esemplari catturati e le fasi lunari ha mostrato una maggiore abbondanza di larve durante il I quarto lunare, fase in cui la differenza di marea manifesta un'escursione minore rispetto alla fase di luna piena o luna nuova. L'identificazione delle principali specie registrate nei diversi periodi, è stata confermata da dati di letteratura in aree limitrofe del Mediterraneo. Le catture con sciabica da terra non hanno mostrato differenze significative nelle comunità ittiche soggette al refluo caldo rispetto alle aree di controllo. ; Due to the large amount of natural water used in the cooling intake of electric power plants, resulting impact on marine organisms have been studied in different part of the world. Cooling water intake usually affect marine organisms in three different way: entrainment (where small aquatic organisms are carried by the cooling water into the power plant), impingement (where the cooling water intake traps larger organisms against the intake screens) and thermal outfall (hot water discharged on living organisms in the receiving water body). In this study (the first one carried out in Italy), the impact of the cooling water intake of the electric power plant of Montalto di Castro (VT) (Central Tyrrhenian Sea) on fish was determined by evaluating the effect of entrainment on eggs and larvae and that of thermal outfall on juveniles. A specific sampling design has been create to operate inside the artificial intake, with new methodology, different plankton nets and right time of haul. Sampling campaign have been conduct at sea, using a handed trawling net, to the catch of juveniles fish during settlement stage, in the thermal outfall area and in two control areas. Overall 668 fish larvae of 24 taxa and 2500 fish eggs were collected after filtering 235.574 m3 of marine water inside the intake. Impact of entrainment on early stage of fish was estimated to be of 12 x 106 larvae and 24,5 x 106 eggs per year. Sardina pilchardus was the most impacted species, with an estimated economic damage to the traditional fisheries, ranging between 9.000 and 15.000 euro per year. The highest values of diversity index, richness and evenness were recorded in summer, whereas the largest number of larvae were caught during winter.
