Background/Aim. Due to their low tolerance to pollutants and hand-to-mouth pathways the health risk is very high in children's population. The aim of this study was to evaluate risk to children's health based on the content of heavy metals in urban soil samples from Podgorica, Montenegro. This study included the investigation of several toxic metals such as Pb, Cd, Cu and Zn in soil samples from public parks and playgrounds. Methods. Sampling was conducted in a period October-November, 2012. Based on cluster analysis, soil samples were divided into two groups related to similarity of metal content at examinated locations: the group I - near by recreational or residential areas of the city, and the group II - near traffic roads. Concentration of toxic metals, in urban soil samples were determined by a graphite furnace atomic absorption spectrometry (Pb and Cd) and by inductively coupled plasma optical emission spectrometry technique after microwave digestion. Due to exposure to urban soil, non-cancerogenic index hazardous index (HI) for children was estimated using 95th percentile values of total metal concentration. The value of the total (ingestion, dermal and inhalation) HI is calculated for maximum, minimum and the average concentration of metals for children. Results. Mean concentrations of Pb, Cd, Cu and Zn in the surface layer of the studied urban soils were 85.91 mg/kg, 2.8 mg/kg and 52.9 mg/kg and 112.5 mg/kg, respectively. Samples from group II showed higher metal content compared to group I. Urbanization and traffic are the main sources of pollution of the urban soils of Podgorica. Most of the samples (93.5%) had a high Pb content, 12.9% of the samples had a higher content of Cd, while Cu and Zn were within the limits prescribed by national legislation. At one location the level of security for lead is HI = 0.8 and very closed to maximum acceptable value of 1. It is probably the result of intensive traffic near by. Conclusion. All metals investigated showed relatively higher concentrations at sites that were close to industrial places and high ways. The mean concentrations of Pb and Zn and maximum concentrations of Pb, Cd, and Zn were higher than presented values in the National Regulation. ; Uvod/Cilj. Zbog niske tolerancije na zagađivače i puta prenošenja ruke-usta, rizik od narušenja zdravlja je veoma visok u dečjoj populaciji. Cilj ovog rada bio je da se izvrši procena rizika po zdravlje dece na osnovu sadržaja teških metala u uzorcima gradskog zemljišta u Podgorici, Crna Gora. Ovo istraživanje je obuhvatilo određivanje koncentracija nekoliko toksičnih metala kao što su Pb, Cd, Cu i Zn u uzorcima zemljišta javnih parkova i dečijih igrališta. Metode. Uzorkovanje je sprovedeno tokom oktobra i novembra 2012. godine. Klaster analizom uzorci zemljišta podeljeni su u dve grupe na osnovu sličnosti sadržaja metala na ispitivanim lokacijama: grupa I - uzorci sa mesta u rekreativnim ili stambenim delovima grada i grupa II - uzorci iz parkova i igralište blizu saobraćajnice. Koncentracija metala u uzorcima urbanog zemljišta određena je primenom atomske apsorpcione spektrometrije pomoću grafitne kivete (Pb i Cd) i tehnike optičke emisione spektroskopije sa induktivno kuplovanom plazmom (Cu i Zn) nakon mikrotalasne digestije. Na osnovu stepena izloženosti uticaju gradskog zemljišta, nekancerogeni indeks opasnosti (hazard index - HI) za decu izračunat je na 95. procentu vrednosti ukupne koncentracije metala. Vrednost za ukupni (ingestioni, dermalni i inhalacioni) HI izračunata je za maksimalne, minimalne i srednje koncentracije ispitivanih metala za decu. Rezultati. Srednja koncentracija toksičnih metala u uzorcima površinskog sloja zemljišta iznosila je 85,91 mg Pb/kg tla, 2,8 mg Cd/kg tla, 52,9 mg Cu/kg tla i 112,5 mg Zn/kg tla. Sadržaj metala bio je značajno veći u uzorcima zemljišta iz grupe II nego u uzorcima grupe I. Ovo jasno ukazuje na izražen i uočljiv uticaj urbanizacije, a naročito saobraćaja, na zagađenje zemljišta. Većina uzoraka (93,5%) imala je povećan sadržaj Pb, 12,9% uzoraka imalo je povećan sadržaj Cd, dok je sadržaj Cu i Zn bio u granicama propisanim nacionalnom regulativom. Na jednoj lokaciji vrednost za nivo bezbednosti za olovo bio je HI = 0,8, vrlo blizu maksimalne prihvatljive vrednosti koja iznosi 1, što je verovatno posledica intenzivnog saobraćaja u neposrednoj blizini ispitivane lokacije. Zaključak. Ispitivani metali imali su više koncentracije na mestima u blizini industrijske zone i autoputa. Prosečne koncentracije Pb i Zn, kao i maksimalne koncentracije Pb, Cd i Zn bile su više od vrednosti propisanih nacionalnom regulativom.

Background/Aim. Cyanobacterial toxins befall a group of various compounds according to chemical structure and health effects on people and animals. The most significant in this large group of compounds are microcystins. Their presence in water used for human consumption causes serious health problems, liver beeing the target organ. Microcystins are spread all over the world. Waterblooms of cyanobacterias and their cyanotoxins are also common in the majority of surface waters in Serbia. The aim of this study was to propose HPLC method for determination of mikrocystin-LR, to validate the method and to use it for determination of microcystin-LR in the surface water of the river Ponjavica. The Ponjavica is very eutrophic water and has ideal conditions for the cyanobacterial growth. Methods. Sample of water form the Ponjavica river were collected during the summer 2008. Coupled columns (HLB, Sep-Pak), were used for sample preparation and HPLC/PDA method was used for quantification of microcystin- LR. Results. Parameters of validation show that the proposed method is simple, fast, sensitive (0.1 mg/L) and selective with the yield of 89%-92%. The measuring uncertainty of 􀁲 5% was obtained. The obtained results for surface water show that microcystin concentration reached the maximum level during August and September (1.5 μg/L). The value is higher than maximum allowable concentration of microcystin in drinking water (1 μg/L) proposed by WHO. Conclusion. This study contributes to the issue of pollution of the National Park Ponjavica. Besides, literature data and WHO clearly point out harmfulness of cyanobasterias and their toxins and implicate the necessity of legislation concerning determination and monitoring of these toxins in our country. Method used for quentification of mycrocystin-LR was shown to be sensitive, selective, rapid and simple and could be recommended for routine determination of this toxin. ; Uvod/Cilj. Cijanotoksini, odnosno toksini cijanobakterija, spadaju u grupu raznovrsnih jedinjenja kako po hemijskoj strukturi, tako i po dejstvu na ljude i životinje. Najveći značaj u ovoj velikoj grupi jedinjenja imaju mikrocistini. Njihovo prisustvo u vodi može da izazove toksične efekte, posebno na jetru. Mikrocistini su rasprostranjeni širom sveta. Cvetanje toksičnih cijanobakterija i njihovi cijanotoksini karakteristični su i za neke površinske vode u Srbiji. Cilj ovog rada bio je da se uvede HPLC metoda za određivanje mikrocistina-LR, da se izvrši validacija metode i odredi sadržaj mikrocistina-LR u površinskoj vodi reke Ponjavice, veoma eutrofne reke, koja poseduje idealne uslove za razvoj cijanobakterija. Metode. Za pripremu uzoraka korišćeni su HLB i C8 ulošci, a za dodatno prečišćavanje upotrebljeni su Sep-Pak silika ulošci. Kvantifikacija mikrocistina-LR vršena je metodom HPLC pomoću detektora na principu fotodiode (HPLC/PDA) korišćenjem kolone RP C18 Zorbax Eclips (4,6x150,5 μm), upotrebom mobilne faze sastava 0,1% rastvor trifluorosirćetne kiseline (TFA) u metanolu: 0,1% vodeni rastvor TFA u odnosu 60 : 40 (v/v), na 238 nm. Retenciono vreme mikrocistina-LR iznosilo je 10 min. Uzorci vode reke Ponjavice za analizu mikrocistina-LR sakupljeni su tokom perioda jun-novembar 2008. godine. Rezultati. Validacijom metode utvrđena je osetljivost 0,1 μg/L, prinos 89-92% i merna nesigurnost od ± 5%. Rezultati uzoraka vode reke Ponjavice pokazali su da je vrednost mikrocistina-LR dostigla maksimum tokom avgusta i septembra (1,5 μg/L), i da je iznad maksimalno dozvoljene koncentracije za pijaću vodu (1 μg/L) prema preporuci Svetske zdravstvene organizacije. Zaključak. Parametri validacije pokazuju da je predložena metoda osetljiva i selektivna, što je, uz činjenicu da je jednostavna i brza, čini pogodnom za rutinsko određivanje mikrocistina-LR. Predloženom metodom određene su povišene koncentracije ovog toksina cijanobakterija u uzorcima vode reke Ponjavice, koje ukazuju na zagađenost Parka prirode 'Ponjavica'.

This study presents the case of opiate analysis in urine of two female participants, aged between 20 and 25 who consumed commonly available bakery products with poppy seeds. One of the participants consumed strudel, the product with larger amount of poppy seeds, while the other consumed braid pastry. Urine samples of both participants (n=8) were collected 5, 12, 24, 30, 36, 48, 60 and 72 h after the consummation. Immunochromatographic screening test (cuttof 300 ng/mL) and LC-MS analysis were preformed on the presence of opiates. The screening test for opiates was positive for a very long period of time, 48 h after the consumption of braid pastry and even 72 h after the consumption of strudel. LC-MS analysis verified the results of screening test and the presence of the main opioide alcaloides, morphine and codeine, was confirmed in all positive samples. Determined morphine level was higher than the level of codeine in all samples. The concentration of morphine (0.25 mg/L) was higher in urine of the participant who consumed strudel, compared to the concentration of morphine (0.06 mg/L) after the consumption of braid pastry. The results of this research show that opiates can be found in urine after the consumption of products which contain poppy seed. Morphine and codeine can be detected even during the same time period as in the samples of users of psychoactive substances (3 days and longer). According to the results, retention time of opiates in urine is longer after the consumption of products with higher amounts of poppy. This finding should be taken into account in routine testing of employees in the workplace, members of the military, students in schools, as well as athletes. ; U ovom radu je prikazan slučaj dve osobe ženskog pola starosti između 20 i 25 godina, koje su konzumirale široko dostupne pekarske proizvode sa makom, sa ciljem da se izvrši analiza opijata u urinu. Jedna osoba je konzumirala štrudlu a druga pletenicu, s tim da je štrudla vizuelno sadržala veću količinu maka. Uzorci urina od obe osobe (po 8 uzoraka) su sakupljeni nakon 5, 12, 24, 30, 36, 48, 60 i 72 sata od konzumacije. Urađena je imunohromatografska i LC/MS analiza na prisustvo opijata. Rezultati imunohromatografske analize su pokazali da je test na opijate (cuttof 300 ng/mL) bio pozitivan u veoma dugom vremenskom periodu, 48 sati nakon konzumacije pletenice i čak 72 sata nakon konzumacije štrudle. LC-MS analizom su potvrđeni rezultati dobijeni preliminarnim testom i u svim pozitivnim uzorcima je dokazano prisustvo glavnih opijatnih alkaloida, morfina i kodeina. Koncentracija morfina je bila veća od koncentracije kodeina u svim uzorcima. Uočena je veća koncentracija morfina (0,25 mg/L) u urinu osobe koja je konzumirala štrudlu u odnosu na koncentraciju morfina (0,06 mg/L) nakon konzumacije pletenice. Rezultati ovog rada potvrđuju da se opijati nalaze u urinu nakon konzumacije proizvoda sa makom i da ih je moguće detektovati čak tokom istog vremenskog perioda kao i kod korisnika psihoaktivnih supstanci, tri dana, i više. Takođe, prema dobijenim rezultatima vreme zadržavanja opijata u urinu je duže nakon konzumacije proizvoda sa većom količinom maka. Navedene rezultate treba imati u vidu prilikom rutinskih testiranja zaposlenih na radnom mestu, pripadnika vojske, učenika u školama i sportista.

Background/Aim. The beneficial effects of medicinal plants are well-known from the ancient times. However, expansion of phytopharmacy and phytotherapy occured during the last decades. Medicinal plants can absorb environmental contaminants from the ground and consequently may cause harmful effects on human health. Quality control usually comprises standard methodology which includes macroscopic identification and examination of active ingredients. Additionaly, there is a permanent need to control the level of pollutants in herbs, with a particular attention to the level of toxic metals. In this study we estimated the level of contamination by determining the content of cadmium (Cd) in the herbs of Hypericum perforatum and Thymus serpyllum collected from the different localities of the mountains Rtanj and Ozren. Methods. Herbs of investigated plants were collected during July 2005 from various localities of Rtanj and Ozren mountains. After drying, homogenization and mineralization, Cd content was determined by atomic absorption spectrophotometry. Results. The obtained results show that Cd content varies significantly in Hypericum perforatum samples collected. The lowest Cd level was found in samples from the one of Rtanj localities (0.25 mg Cd/kg), while the highest was observed in Hypericum perforatum from Ozren locality (1.24 mg/kg). Levels of Cd in the three of four investigated localities were higher than pro posed by WHO (0.3 mg/kg dried herb material). In all investigated samples of Thymus serpyllum herbs Cd levels were below the limit of detection of analytical method (0.2 mg Cd/kg dried materials). Conclusion. This work contributes to the issue of Cd content in Hypericum perforatum and Thymus serpyllum grown in localities of Rtanj and Ozren, and implies the importance for systemic control of Cd content in Hypericum species in order to provide safety of their preparations. Furthermore, regarding Cd toxicity, maximal permissible level of Cd in plant material should be evaluated and established concerning national legislative frame. ; Uvod/Cilj. Lekovitost nekih biljnih vrsta bila je poznata čoveku još pre naše ere. Međutim, do ekspanzije fitofarmacije i fitoterapije dolazi tek poslednjih decenija. Lekovite biljke usled zagađenja životne sredine mogu da apsorbuju prisutne zagađivače. Kvalitet biljnih sirovina prati se standardnim metodama koje se prvenstveno odnose na identifikaciju biljne vrste i ispitivanja aktivnih principa. Ipak, i pored toga, uočena je potreba za kontrolisanjem i praćenjem nivoa zagađivača, među kojima značajno mesto zauzimaju toksični metali. U ovom radu ispitivan je sadržaj kadmijuma (Cd) u herbi dve biljne vrste, Hypericum perforatum L. i Thymus serpyllum L. koje su prikupljene sa različitih lokaliteta planina Rtanj i Ozren, sa ciljem procene nivoa kontaminacije. Metode. Herbe ispitivanih lekovitih biljaka sakupljene su tokom jula 2005. godine sa različitih lokaliteta planina Rtnja i Ozrena. Nakon sušenja, homogenizacije i mineralizacije, sadržaj Cd određivan je metodom atomske apsorpcione spektrofotometrije. Rezultati. Dobijeni rezultati ukazali su na značajne varijacije sadržaja Cd u sakupljenim uzorcima Hypericum perforatum. Najniži sadržaj Cd od 0,25 mg Cd/kg dobijen je u uzorku sa lokaliteta Rtnja, dok je najviši nivo Cd određen u uzorku Hypericum perforatum sa lokaliteta Ozrena (1,24 mg/kg). Dobijene vrednosti sadržaja Cd u herbi Hypericum perforatum sa tri od četiri ispitivana lokaliteta bile su iznad vrednosti koju preporučuje SZO (0,3 mg/kg osušene droge). U svim ispitivanim uzorcima Thymus serpyllum nivoi Cd bili su ispod nivoa detekcije primenjene analitičke tehnike (0,2 mg Cd/kg osušene herbe). Zaključak. Rezultati ovog rada doprineli su istraživanju sadržaja Cd u biljnim vrstama Hypericum perforatum i Thymus serpyllum koje rastu na lokalitetima Rtnja i Ozrena i ukazuju na potrebu uvođenja sistematske kontrole sadržaja Cd u Hypericum vrstama radi bezbedne upotrebe preparata izrađenih na bazi ovih biljnih vrsta. Imajući u vidu toksičnost Cd nameće se potreba da se nacionalnim zakonodavstvom odrede i ustanove maksimalno dozvoljene koncentracije sadržaja ovog metala u biljnim lekovitim sirovinama.
