A Test System for Tick-Borne Encephalitis Virus Detection Based on Bioluminescent Immunoassay

The tick-borne encephalitis virus (TBEV) is the causative agent of one of the most severe human neuroinfections. The infection transmitted by ixodid ticks is spread throughout the forest and foreststeppe zones of the temperate climatic belt of the Eurasian continent, including the Siberian region of the Russian Federation. Despite the availability of commercial analytical systems for the detection of TBEV, the task of developing approaches to a quick and reliable analysis that can be performed routinely, particularly in environmental studies, remains topical. A solid-phase bioluminescent immunoassay for determining the tick-borne encephalitis virus (TBEV) in ticks was developed. The assay is based on the hybrid protein consisting of a modified thermostable version of Renilla muelleri luciferase and a single-chain mini-antibody to protein E. This unique protein had been obtained and investigated by the authors earlier. The current study describes the expression of the hybrid protein in two different strains of recombinant E. coli cells. The optimal conditions for obtaining a highly purified protein were found. The bioluminescent reaction of the luciferase domain was triggered with the help of the stable natural form of the substrate, a Ca2+-dependent coelenterazine-binding protein, the recombinant variant of which was obtained by the authors. The conditions for production and storage of the immunoassay components (the hybrid protein, the stable form of the luciferase substrate, and activated microplates) were determined. Using the developed test system, more than 900 tick samples were analyzed for TBEV. In terms of sensitivity (89.5%) and specificity (98.9%), the proposed method is not inferior to colorimetric detection and is much simpler and faster than the latter
Interdisciplinary integration in teaching Mathematics as a means of forming pupils' mathematical literacy

This article discusses the problem of implementing interdisciplinary connections between a school course in Mathematics and other natural science disciplines at a secondary school as topical means of developing functional literacy among pupils. The mechanisms of interdisciplinary connection of Mathematics with other disciplines are listed. As a means of interdisciplinary communication of Mathematics with other disciplines, there can be the interaction of pupils in the information educational environment to search for new knowledge, to identify properties of a generalised character, to solve practice-oriented problems, to combine knowledge from various general education subjects, to teach Computer Science based on the transfer and generalisation of knowledge from related subjects. The article provides examples of practice-oriented tasks that illustrate the relationship of Mathematics with such secondary school disciplines as Computer Science, Biology, Geography, and Physics. It is noted that when solving such problems, schoolchildren learn various aspects of multi-level practice-oriented activities, including the use of computer capabilities and the global Internet. It is shown that mathematical constructions and their analogues are found even in areas of knowledge quite distant from exact sciences, such as literature, for example. It is substantiated that it is the solution of interdisciplinary problems that forms the meta-skills necessary for everyday life in pupils.

Climate change mitigation: Hypothesis-formulation and analysis of interventions
Nowadays the world energy consumption continuously increases due to various human-related activities. Increasing energy efficiency and decarbonizing the economic development, especially with measures aimed at the most energy-intensive sectors (industry, buildings, and mobility) is therefore a way towards the "eco-sustainable" community. The energy efficiency improving has environmental implications as it results in a reduction of the atmosphere polluting emissions. For the GHG (and in particular for the carbon dioxide) it is necessary to study and to analyze the so-called "negative technologies" - technologies that allow a carbon dioxide concentrations reduction. Considering the strategies in order to limit the climate change and to improve the air quality, it is necessary to assess the impacts of both these phenomena and therefore to identify synergies (win - win) and avoiding solutions that improve one of the two phenomena and worsen the other one (win - lose). The aim of this work is to analyze the opportunities of the "negative technologies" application for the climate change mitigation and to formulate the hypothesis of mobility modification using methods for the fuel consumption reduction. The negative technologies were studied from a technological point of view analyzing the potential advantages and disadvantages and stressing the technical challenge. The new approach for reduction of environmental and climatic impacts of vehicle operation on the environment by changes in the albedo of a vehicle roof is suggested. The proposed method allows to reduce the fuel consumption and greenhouse gases emission while idle with the engine running. The cost-effective modification in fuel quality using the microdose of the developed surface-active additive enables to reduce the specific fuel consumption dramatically, and substantially improves the environmental characteristics of vehicles operation.
