Besides anthropogenic contamination, freshwater environments can also be affected by the presence of natural toxins. Mycotoxins, plant toxins and cyanotoxins are the most relevant groups that can be found in the aquatic system. However, until now, only cyanotoxins have been more carefully studied. In the present work, single workflow for the assessment of natural toxins in waters, based on suspect screening and target screening of a selected group of toxins is presented. The approach is based on a triple-stage solid-phase extraction (SPE) able to isolate a wide range of natural toxins of different polarities, followed by liquid chromatography coupled to high-resolution mass spectrometry (HPLC-ddHRMS2) using a Q-Exactive Orbitrap analyser. The acquisition was performed in full-scan (FS) and data-dependant acquisition (ddMS2) mode, working under positive and negative mode. For the tentative identification, different on-line databases such as ChemSpider and MzCloud and an in-house natural toxins list with 2384 structures, that includes cyanotoxins, plant toxins and mycotoxins, were used. Also, thanks to the MS2 data, it was possible to achieve a high level of tentative identification confidence, but confirmation was only possible comparing the standards of the suspected compounds. For those, the analytical parameters of the developed method were also validated, and the quantification was possible by external calibration. Validation showed recoveries in the range between 53 and 95%, and method limits of detection (MDL) between 0.02 and 1.22 μg/L. This approach was applied to study natural toxins in 4 sampling sites along the Ter River in Catalonia (NE Spain). In this preliminary study 23 natural toxins were tentatively identified, and 9 of them confirmed (aflatoxin B1, anatoxin-a, nodularin, microcystin-LR, baicalein, kojic acid, cinchonine, B-asarone and atropine). The results of the quantification of these compounds showed concentrations below 1 μg/L in all cases, that is considered safe according to the actual legislation. Among natural freshwater pollutants, cyanotoxins, mycotoxins, and phytotoxins are the most important and less studied. Their identification in surface water is challenging especially cause of the lack of standards and established analytical parameters. Most target methods focus one or a single group of compounds with similar characteristics. Here we present an AIF fast method for the tentative identification of natural toxins in water. Respect to the previous method [1], it offers higher performances for the acquisition of unknown compounds at low levels for higher number of analytes.The key aspects of the method are: -The qualitative screening DIA-AIF workflow using Q Exactive Orbitrap. Both targeted and suspect screening bases have been combined with online databases and suspect list to retrieve candidates as suspect natural toxins and their metabolites or degradation products. -The in-silico analysis of mass spectrums allowed a fast structural characterization. -The workflow has been finally applied to real samples coming from the Czech Republic, Italy, and Spain allowing the determination of 17 suspect natural toxins, 4 of them confirmed. None toxin passed the limit of 1 µg/L taken from the legislation applied for microcystin LR and arbitrarily extended to all toxins.

Abstract
Background
Bloom-forming cyanobacteria occur globally in aquatic environments. They produce diverse bioactive metabolites, some of which are known to be toxic. The most studied cyanobacterial toxins are microcystins, anatoxin, and cylindrospermopsin, yet more than 2000 bioactive metabolites have been identified to date. Data on the occurrence of cyanopeptides other than microcystins in surface waters are sparse.

Results
We used a high-performance liquid chromatography–high-resolution tandem mass spectrometry/tandem mass spectrometry (HPLC–HRMS/MS) method to analyse cyanotoxin and cyanopeptide profiles in raw drinking water collected from three freshwater reservoirs in the United Kingdom. A total of 8 cyanopeptides were identified and quantified using reference standards. A further 20 cyanopeptides were identified based on a suspect-screening procedure, with class-equivalent quantification. Samples from Ingbirchworth reservoir showed the highest total cyanopeptide concentrations, reaching 5.8, 61, and 0.8 µg/L in August, September, and October, respectively. Several classes of cyanopeptides were identified with anabaenopeptins, cyanopeptolins, and microcystins dominating in September with 37%, 36%, and 26%, respectively. Samples from Tophill Low reservoir reached 2.4 µg/L in September, but remained below 0.2 µg/L in other months. Samples from Embsay reservoir did not exceed 0.1 µg/L. At Ingbirchworth and Tophill Low, the maximum chlorophyll-a concentrations of 37 µg/L and 22 µg/L, respectively, and cyanobacterial count of 6 × 104 cells/mL were observed at, or a few days after, peak cyanopeptide concentrations. These values exceed the World Health Organization's guideline levels for relatively low probability of adverse health effects, which are defined as 10 µg/L chlorophyll-a and 2 × 104 cells/mL.

Conclusions
This data is the first to present concentrations of anabaenopeptins, cyanopeptolins, aeruginosins, and microginins, along with microcystins, in U.K. reservoirs. A better understanding of those cyanopeptides that are abundant in drinking water reservoirs can inform future monitoring and studies on abatement efficiency during water treatment.

In this work a suspect-screening approach was employed to assess the polymers and plastic additives of micro(nano)plastics (NPL/MPLs) of size ranges from the nm range to 20 μm present in seawater from the top 5 cm of the Mar Menor lagoon during two sampling campaigns (summer and winter), as well of other potentially adsorbed compounds onto the plastic particles surfaces and suspended material. The identification of NPL/MPLs has been based on characteristic Kendrick Mass Defect analysis for each polymer type in mass spectra. The applied methodology allowed to identify NPLs/MPLs of polystyrene (PS), polyethylene (PE), polyisoprene (PI), polybutadiene (PBD), polypropylene (PP), polyamides (PA), polyvinylchloride (PVC), n-isopropylacrylamide (PNIPAm), and polydimethylsiloxanes. In addition, PS, PE, PI, PBD, PP, PA, and PVC were confirmed with standards, and the equivalent concentrations were quantified. The results of this study showed that most frequently found compounds were PP, PE, PA and PNIPAm, while the compound found at higher concentrations was by far PP reaching the 9,303 ± 366 ng/mL in one of the samples. A total number of 135 chemical compounds were tentatively identified, 74 of them plastic additives and compounds used in the polymers manufacture or coming from the polymer's decomposition. In relation to plastic additives, the more frequently tentatively identified compounds were plasticizers such as phthalates group; stabilizers such as antioxidants (e.g., distearyl 3,3′-thiodipropionate, 2,5-di-tert-butylhydroquinone), and UV filters as benzotriazoles. Several flame retardants of the group of phosphates were as well detected. The other compounds tentatively identified in the samples were pharmaceuticals, pesticides, food additives, flavors and natural products that were attached onto the plastic particles and particulate matter from surrounding waters. In regards to the seasonal variation, during the summer a major number of compounds were tentatively detected, while de concentrations of polymers were slightly higher in winter. A huge variety of organic microcontaminants are presently detected in freshwater ecosystems, but there is still a lack of knowledge about their interactions, either with living organisms or with other contaminants. Actually, carbon nanomaterials like fullerenes (C 60 ) can act as carriers of organic microcontaminants, but their relevance in processes like bioaccumulation and biotransformation of organic microcontaminants by organisms is unknown. In this study, mesocosm experiments were used to assess the bioaccumulation and biotransformation of three organic microcontaminants (venlafaxine, diuron and triclosan) in river biofilms, and to understand how much the concomitant presence of C 60 at environmental relevant concentrations could impact these processes. Results indicated that venlafaxine exhibited the highest bioaccumulation (13% of the initial concentration of venlafaxine in water), while biotransformation was more evident for triclosan (5% of the initial concentration of triclosan in water). Furthermore, biotransformation products such as methyl-triclosan were also present in the biofilm, with levels up to 42% of the concentration of accumulated triclosan. The presence of C 60 did not involve relevant changes in the bioaccumulation and biotransformation of microcontaminants in biofilms, which showed similar patterns. The contamination of marine ecosystems by contaminants of emerging concern such as personal care products or per- and polyfluoroalkyl substances is of increasing concern. This work assessed the concentrations of selected contaminants of emerging concern in water and sediment of European aquaculture areas, to evaluate their co-variation with legacy contaminants (polycyclic aromatic hydrocarbons) and faecal biomarkers, and estimate the risks associated with their occurrence. The 9 study sites were selected in 7 European countries to be representative of the aquaculture activities of their region: 4 sites in the Atlantic Ocean and 5 in the Mediterranean Sea. Musks, UV filters, preservatives, per- and polyfluoroalkyl substances and polycyclic aromatic hydrocarbons were detected in at least one of the sites with regional differences. While personal care products appear to be the main component of the water contamination, polycyclic aromatic hydrocarbons were mostly found in sediments. As expected, generally higher levels of personal care products were found in sewage impacted sites, urbanised coasts and estuaries. The risk assessment for water and sediment revealed a potential risk for the local aquatic environment from contaminants of both legacy and emerging concern, with a significant contribution of the UV filter octocrylene. Despite marginal contributions of per- and polyfluoroalkyl substances to the total concentrations, PFOS (perfluorooctane sulfonate) aqueous concentrations combined to its low ecotoxicity thresholds produced significant hazard quotients indicating a potential risk to the ecosystems. ; This study was financially supported by the SEA-on-a-CHIP project, funded from European Union Seventh Framework Programme (FP7-OCEAN-2013) under grant agreement No. 614168.
