Paratuberculosis (PTB) is an enteric granulomatous disease caused by Mycobacterium avium subsp. paratuberculosis (MAP) that mainly aects ruminants. Current vaccines have shown to be cost-eective control reagents, although they are restricted due to cross-interference with bovine tuberculosis (bTB). Therefore, novel vaccination strategies are needed and this study is focused on evaluating alternative vaccination routes and their eect on the local immune response. The MAP oral challenge rabbit model was used to evaluate and compare an experimental inactivated MAP vaccine through oral (VOR) and intradermal (VID) routes. The VID group presented the highest proportion of animals with no visible lesions and the lowest proportion of animals with MAP positive tissues. Immunohistochemistry analysis revealed that the VID group presented a dominantly M1 polarized response indicating an ability to control MAP infection. In general, all vaccinated groups showed lower calprotectin levels compared to the non-vaccinated challenged group suggesting less active granulomatous lesions. The VID group showed some degree of skin test reactivity, whereas the same vaccine through oral administration was completely negative. These data show that PTB vaccination has an eect on macrophage polarization and that the route influences infection outcome and can also have an impact on bTB diagnosis. Fibrinous polyserositis in swine farming is a common pathological finding in nursery animals. The differential diagnosis of this finding should include Glaesserella parasuis (aetiological agent of Glässer's disease) and Mycoplasma hyorhinis, among others. These microorganisms are early colonizers of the upper respiratory tract of piglets. The composition of the nasal microbiota at weaning was shown to constitute a predisposing factor for the development of Glässer's disease. Here, we unravel the role of the nasal microbiota in the subsequent systemic infection by M. hyorhinis, and the similarities and differences with Glässer's disease. Nasal samples from farms with recurrent problems with polyserositis associated with M. hyorhinis (MH) or Glässer's disease (GD) were included in this study, together with healthy control farms (HC). Nasal swabs were taken from piglets in MH farms at weaning, before the onset of the clinical outbreaks, and were submitted to 16S rRNA gene amplicon sequencing (V3-V4 region). These sequences were analyzed together with sequences from similar samples previously obtained in GD and HC farms. Animals from farms with disease (MH and GD) had a nasal microbiota with lower diversity than those from the HC farms. However, the composition of the nasal microbiota of the piglets from these disease farms was different, suggesting that divergent microbiota imbalances may predispose the animals to the two systemic infections. Infection by human immunodeficiency virus (HIV) depends on the function, in virion morphogenesis and other stages of the viral cycle, of a highly conserved structural element, the major homology region (MHR), within the carboxyterminal domain (CTD) of the capsid protein. In a modified CTD dimer, MHR is swapped between monomers. While no evidence for MHR swapping has been provided by structural models of retroviral capsids, it is unknown whether it may occur transiently along the virus assembly pathway. Whatever the case, the MHR-swapped dimer does provide a novel target for the development of anti-HIV drugs based on the concept of trapping a nonnative capsid protein conformation. We have carried out a thermodynamic and kinetic characterization of the domain-swapped CTD dimer in solution. The analysis includes a dissection of the role of conserved MHR residues and other amino acids at the dimerization interface in CTD folding, stability, and dimerization by domain swapping. The results revealed some energetic hotspots at the domain-swapped interface. In addition, many MHR residues that are not in the protein hydrophobic core were nevertheless found to be critical for folding and stability of the CTD monomer, which may dramatically slow down the swapping reaction. Conservation of MHR residues in retroviruses did not correlate with their contribution to domain swapping, but it did correlate with their importance for stable CTD folding. Because folding is required for capsid protein function, this remarkable MHR-mediated conformational stabilization of CTD may help to explain the functional roles of MHR not only during immature capsid assembly but in other processes associated with retrovirus infection. The rate of warming of the Antarctic Circumpolar Current exceeds that of the global ocean, and the enhanced uptake of carbon dioxide is causing basin-wide ocean acidification. Observational data suggest that these changes are influencing the distribution and composition of pelagic plankton communities. Long-term and annual field observations on key environmental variables and organisms are a critical basis for predicting changes in Southern Ocean ecosystems. These observations are particularly needed, since high-latitude systems have been projected to experience the most severe impacts of ocean acidification and invasions of allochthonous species. Coccolithophores are the most prolific calcium-carbonate-producing phytoplankton group playing an important role in Southern Ocean biogeochemical cycles. Satellite imagery has revealed elevated particulate inorganic carbon concentrations near the major circumpolar fronts of the Southern Ocean that can be attributed to the coccolithophore Emiliania huxleyi. Recent studies have suggested changes during the last decades in the distribution and abundance of Southern Ocean coccolithophores. However, due to limited field observations, the distribution, diversity and state of coccolithophore populations in the Southern Ocean remain poorly characterised. We report here on seasonal variations in the abundance and composition of coccolithophore assemblages collected by two moored sediment traps deployed at the Antarctic zone south of Australia (2000 and 3700 m of depth) for 1 year in 2001–2002. Additionally, seasonal changes in coccolith weights of E. huxleyi populations were estimated using circularly polarised micrographs analysed with C-Calcita software. Our findings indicate that (1) coccolithophore sinking assemblages were nearly monospecific for E. huxleyi morphotype B/C in the Antarctic zone waters in 2001–2002; (2) coccoliths captured by the traps experienced weight and length reduction during summer (December–February); (3) the estimated annual coccolith weight of E. huxleyi at both sediment traps (2.11 ± 0.96 and 2.13 ± 0.91 pg at 2000 and 3700 m) was consistent with previous studies for morphotype B/C in other Southern Ocean settings (Scotia Sea and Patagonian shelf); and (4) coccolithophores accounted for approximately 2–5 % of the annual deep-ocean CaCO3 flux. Our results are the first annual record of coccolithophore abundance, composition and degree of calcification in the Antarctic zone. Polymerization of the intact capsid protein (CA) of HIV-1 into mature capsidlike particles at physiological ionic strength in vitro requires macromolecularly crowded conditions that approach those inside the virion, where the mature capsid is assembled in vivo. The capsid is organized as a hexameric lattice. CA subunits in each hexamer are connected through interfaces that involve the CA N-terminal domain (NTD); pairs of CA subunits belonging to different hexamers are connected through a different interface that involves the C-terminal domain (CTD). At physiological ionic strength in noncrowded conditions, CA subunits homodimerize through this CTD-CTD interface, but do not hexamerize through the other interfaces (those involving the NTD). Here we have investigated whether macromolecular crowding conditions are able to promote hexamerization of the isolated NTD and/or full-length CA (with an inactive CTD-CTD interface to prevent polymerization). The oligomerization state of the proteins was determined using analytical ultracentrifugation in the absence or presence of high concentrations of an inert macromolecular crowding agent. Under the same conditions that promoted efficient assembly of intact CA dimers, neither NTD nor CA with an inactive CTD-CTD interface showed any tendency to form hexamers or any other oligomer. This inability to hexamerize was observed even in macromolecularly crowded conditions. The results indicate that a functional CTD-CTD interface is strictly required for hexamerization of HIV-1 CA through the other interfaces. AbstractEducation needs to be at the forefront of the efforts to mitigate and adapt to Climate Change (CC) effects. We have introduced the Climate Change Competence (C3) to provide a comprehensive route to include the topics of sustainability and CC into the curriculum. This paper analyses how different primary and secondary teachers can improve this competence using a Massive Online Open Course (MOOC) on the basic science of CC. While the improvement in knowledge is expected, we have observed important advances on abilities, and attitudes on CC which are also essential to teach about CC mitigation and adaptation. The present study used a pre-experimental design, with pre-and post-test measurements with a sample of 530 students. While the MOOC focuses on the Science of CC, the participants also advance in attitudes and abilities, revealing an important correlation between these three dimensions of competence. Also, the study reveals that the MOOC improves the C3 in all the participants, but it does in diverse ways for diverse groups.

Animal tuberculosis (TB) remains a major problem in some countries despite the existence of control programmes focused mainly on cattle. In this species, aerogenous transmission is accepted as the most frequent infection route, affecting mainly the respiratory system. Under the hypothesis that the oral route could be playing a more relevant role in transmission, diagnosis and disease persistence than previously thought, this study was performed to assess the course of TB infection in cattle and its effects on diagnosis depending on the route of entry of Mycobacterium bovis. Two groups of five calves each were either endotracheally (EC) or orally (OC) challenged. Necropsies were carried out 12 weeks after challenge except for three OC calves slaughtered 8 weeks later. All animals reacted to the tuberculin skin test and the entire EC group was positive to the interferon-gamma release assay (IGRA) 2 weeks after challenge and thereafter. The first positive IGRA results for OC calves (3/5) were recorded 4 weeks after challenge. Group comparison revealed significant differences in lesion and positive culture location and scoring. TB-compatible gross lesions and positive cultures were more frequently found in the thorax (p < 0.001) and lung (p < 0.05) of EC animals, whereas OC animals presented lesions (p = 0.23) and positive cultures (p < 0.05) mainly located in the abdomen. These results indicate that the infection route seems to be a determining factor for both the distribution and the time needed for the development of visible lesions.
