Crystallization and melting events of water were studied by DSC and the amount of uncrystallized water was calculated. The evolution of dynamics with hydration level was followed for various dielectric relaxation processes, the emphasis being given to relaxation processes of polar groups on the surface of the proteins and of uncrystallized water molecules. [EN] The conformational mobility in poly(ethyl methacrylate-co-hydrxyethyl acrylate) co-polymers was studied by dielectric relaxation spectroscopy, thermally stimulated depolarization currents and dynamic-mechanical experiments. The relaxation strength and the shape of the dielectric relaxation spectra of the homopolymer and co-polymer networks were analyzed using the Havriliak-Negami equation. The dependence of the relaxation strength on co-polymer composition and temperature was analyzed taking into account the merging of the main and the secondary relaxation processes. The shape of the epsilon" versus frequency plots led to the conclusion that the distribution of relaxation times was broader in the co-polymers with intermediate composition than in the homopolymers, a feature that can be explained by the inhomogeneity produced at molecular scale by the sequence distribution of the monomeric units along the chain. Master curves were built both for the elastic modulus and the mechanical loss tangent, and the temperature dependence of the relaxation times was deduced from the shift factors. [EN] Poly(L-lactide) electrospun mats have been produced with random and aligned fiber orientation and degrees of crystallinity from 0 up to nearly 50%. These two factors, fiber alignment and degree of crystallinity, strongly affect the hydrophobicity of the samples, this being larger for the aligned fiber mats and for the fibers with a higher degree of crystallinity. Whereas, the first effect can be associated with a decrease in the degree of porosity, the second should be related to an increase in fiber stiffness as the observed fiber roughness variations do not show strong differences between the samples. Proliferation of human chondrocytes cultured in a monolayer on these substrates is similar in both aligned and non-aligned amorphous mats. [EN] The clinical management of large-size cartilage lesions is difficult due to the limited regenerative ability of the cartilage. Different biomaterials have been used to develop tissue engineering substitutes for cartilage repair, including chitosan alone or in combination with growth factors to improve its chondrogenic properties. The main objective of this investigation was to evaluate the benefits of combining activated platelet-rich plasma with a stabilized porous chitosan scaffold for cartilage regeneration. To achieve this purpose, stabilized porous chitosan scaffolds were prepared using freeze gelation and combined with activated platelet-rich plasma. Human primary articular chondrocytes were isolated and cultured in stabilized porous chitosan scaffolds with and without combination to activated platelet-rich plasma. Scanning electron microscopy was used for the morphological characterization of the resulting scaffolds. Cell counts were performed in hematoxylin and eosin-stained sections, and type I and II collagen expression was evaluated using immunohistochemistry. Significant increase in cell number in activated platelet-rich plasma/stabilized porous chitosan was found compared with stabilized porous chitosan scaffolds. Chondrocytes grown on stabilized porous chitosan expressed high levels of type I collagen but type II was not detectable, whereas cells grown on activated platelet rich plasma/stabilized porous chitosan scaffolds expressed high levels of type II collagen and type I was almost undetectable. [EN] The increasing use of electronic portable systems and the consequent energy demand, leads to the need to improve energy storage systems. According to that and due to safety issues, high-performance non-flammable electrolytes and solid polymer electrolytes (SPE) are needed. SPE containing different amounts of lithium bis(trifluoromethanesulfonyl) imide (LiTFSI) into a poly(vinylidene fluoride-co-hexafluoropropylene), PVDF-HFP, polymer matrix have been prepared by solvent casting. The addition of LiTFSI into PVDF-HFP allows to tailor thermal, mechanical and electrical properties of the composite. In particular, the ionic conductivity of the composites increases with LiTFSI content, the best ionic conductivities of 0.0011 mS/cmat 25 degrees C and 0.23 mS/cmat 90 degrees C were obtained for the PVDF-HFP/LiTFSI composites with 80 wt % of LiTFSI. This solid electrolyte allows the fabrication of Li metallic/SPE/C-LiFePO4 half-cells with a discharge capacity of 51.2 mAh/ g at C/20. Further, theoretical simulations show that the discharge capacity value depends on the lithium concentration and percentage of free ions and is independent of the solid polymer electrolyte thickness. On the other hand, the voltage plateau depends on the SPE thickness. [EN] The development of three-dimensional environments to mimic the in vivo cellular response is a problem in the building of disease models. This study aimed to synthesize and validate three-dimensional support for culturing monoclonal plasma cells (mPCs) as a disease model for multiple myeloma. The three-dimensional environment is a biomimetic microgel formed by alginate microspheres and produced on a microfluidic device whose surface has been functionalized by a layer-by-layer process with components of the bone marrow's extracellular matrix, which will interact with mPC. As a proof of concept, RPMI 8226 cell line cells were cultured in our 3D culture platform. We proved that hyaluronic acid significantly increased cell proliferation and corroborated its role in inducing resistance to dexamethasone. Despite collagen type I having no effect on proliferation, it generated significant resistance to dexamethasone. Additionally, it was evidenced that both biomolecules were unable to induce resistance to bortezomib. These results validate the functionalized microgels as a 3D culture system that emulates the interaction between tumoral cells and the bone marrow extracellular matrix. [EN] The aim of this work was to determine the influence of the biomaterial environment on human mesenchymal stem cell (hMSC) fate when cultured in supports with varying topography. Poly(vinylidene fluoride) (PVDF) culture supports were prepared with structures ranging between 2D and 3D, based on PVDF films on which PVDF microspheres were deposited with varying surface density. Maintenance of multipotentiality when cultured in expansion medium was studied by flow cytometry monitoring the expression of characteristic hMSCs markers, and revealed that cells were losing their characteristic surface markers on these supports. Cell morphology was assessed by scanning electron microscopy (SEM). Alkaline phosphatase activity was also assessed after seven days of culture on expansion medium. On the other hand, osteoblastic differentiation was monitored while culturing in osteogenic medium after cells reached confluence. Osteocalcin immunocytochemistry and alizarin red assays were performed. We show that flow cytometry is a suitable technique for the study of the differentiation of hMSC seeded onto biomaterials, giving a quantitative reliable analysis of hMSC-associated markers.
