A major historical event, the December 14, 1988 strike, reflects not only labor unions' efforts to redefine their position in post-Franco Spain. It is also evidence of increasing social differentiation and a fundamental conflict among Spanish social democrats.
This issue of Toxicological Sciences features several exciting changes: a redesigned cover, revised category subheadings, and this "Look Inside ToxSci" feature. Together with my esteemed colleagues in the field of green chemistry, we have outlined some exciting opportunities for the field of toxicology in the editorial on green chemistry and toxicology. There are insightful articles on the regulatory challenges regarding mixtures that are being addressed by the European Union and on risk assessment of carbon nanotubes. In the past, we have highlighted a single article from each issue. It has become increasingly difficult to select just one article from the volume of high quality work, so beginning this month we will highlight multiple articles in each issue here in "Look Inside ToxSci." From solvents, pesticides, and nanoparticles to the microbiome, the highlighted articles span the breadth of our field. Of course, this issue contains something that will never change; our raison d'etre …the best original research in the field of toxicology. -Gary W. Miller.

; This article analyses the recommendations for improvement of elections made in the final reports of observation missions deployed by the European Union to Latin America over a 15-year period (2000-2014), taking the implementation of recommendations as an empirical indicator of the observation missions' impact. The analysis reveals that it is not infrequent that eu missions reiterate recommendations when they return to a country to observe another election, and that persistent problems turn out to be more numerous than shortcomings being satisfactorily resolved. This circumstance asks for a review of the mechanisms to formulate recommendations as well as for the search of the most adequate means to promote their implementation during the post-election period.

Ion channels have recently attracted attention as potential mediators of skin disease. Here, we explored the consequences of genetically encoded induction of the cell volume-regulating Ca2+-activated KCa3.1 channel (Kcnn4) for murine epidermal homeostasis. Doxycycline-treated mice harboring the KCa3.1+-transgene under the control of the reverse tetracycline-sensitive transactivator (rtTA) showed 800-fold channel overexpression above basal levels in the skin and solid KCa3.1-currents in keratinocytes. This overexpression resulted in epidermal spongiosis, progressive epidermal hyperplasia and hyperkeratosis, itch and ulcers. The condition was accompanied by production of the pro-proliferative and pro-inflammatory cytokines, IL-β1 (60-fold), IL-6 (33-fold), and TNFα (26-fold) in the skin. Treatment of mice with the KCa3.1-selective blocker, Senicapoc, significantly suppressed spongiosis and hyperplasia, as well as induction of IL-β1 (-88%) and IL-6 (-90%). In conclusion, KCa3.1-induction in the epidermis caused expression of pro-proliferative cytokines leading to spongiosis, hyperplasia and hyperkeratosis. This skin condition resembles pathological features of eczematous dermatitis and identifies KCa3.1 as a regulator of epidermal homeostasis and spongiosis, and as a potential therapeutic target ; This work was supported by the Government of Aragón (DGA-METIC–consortium; B04_17R), FP7-PEOPLE-MC-CIG to RK, FIS-CB06/07/1036 to PG and RK, ERC-2014-StG-638284, DPI2016-75458-R, T39_17R to EP; FIS-PI16/02112 to ALGO; NHLBI-R01-HL080173, NCRRP20-RR018751 to HM.
