Technological applications have caused a revolution in the way individual transportation rides are offered and taken in cities all over the world. The adoption and regulation of ride-hailing has been the subject of heated discussion involving elected officials, bureaucrats, industry proponents, the traditional taxicab industry, and civil society. To implement ride-hailing, proponents and platform operators confront an intricate web of decision-making processes and institutional politics. In this way, existing normative processes shape the emergent regulation of such transportation network companies. This article analyzes the case of Vancouver, Canada, one of the largest cities in North America where ride-hailing companies belatedly secured authorization to operate from the provincial government in 2019. Focusing on the policy debate since 2012, the research identifies the interactions and processes of interest representation among various actors regarding this new transportation technology. The analysis shows how a variety of political, economic and regulatory strategies contributed to the delayed adoption. ; Les applications technologiques ont révolutionné la manière dont les trajets de transports individuels sont proposés et empruntés dans les villes du monde entier. L'adoption et la réglementation du covoiturage a fait l'objet de discussions animées impliquant des élues, des bureaucrates, des promoteurs de l'industrie, l'industrie traditionnelle des taxis et la société civile. Pour mettre en oeuvre le covoiturage, les promoteurs et les opérateurs de plateformes sont confrontés à un réseau complexe de processus décisionnels et de politiques institutionnelles. Cela faisant, les processus normatifs existants façonnent la réglementation émergente de ces sociétés de réseau de transport. Cet article analyse le cas de Vancouver, au Canada, l'une des plus grandes villes d'Amérique du Nord où les entreprises de covoiturage ont tardivement obtenu l'autorisation d'opérer du gouvernement provincial en 2019. En se concentrant sur le débat politique depuis 2012, la recherche identifie les interactions et les processus de représentation des intérêts entre les différents acteurs concernant cette nouvelle technologie de transport. L'analyse montre comment diverses stratégies politiques, économiques et réglementaires ont contribué au retard de l'adoption.

Cordyceps militaris (L.) Link, an edible entomopathogenic fungus widely used in traditional Chinese medicine, has numerous potential medicinal properties including antitumor activity. The methanolic extract of C. militaris fruiting body was recently shown to have tumor cell growth inhibitory activity in several human tumor cell lines. Nonetheless, the mechanism of action involved is still not known. This work aimed at further studying the effect of the methanolic extract of C. militaris regarding its antitumor mechanism of action, using the non-small cell lung cancer cell line (NCI-H460) as a model. Results showed that treatment with the extract decreased cellular proliferation, induced cell cycle arrest at G0/G1 and increased apoptosis. In addition, the extract increased the levels of p53 and p21. Moreover, an increase in p-H2A.X and 53BP1 levels, together with an increase in the number of 53BP1 foci/cell (all indicative of DNA damage), were also observed after treatment with the extract. This work suggests that this extract affected NCI-H460 cellular viability through a mechanism involving DNA damage and p53 activation. This further supports the potential of this extract as a source of bioactive compounds, which may be used in anticancer strategies.

The paper describes the on-line dynamic security assessment functions developed within the European Union, DGXII programme, CARE. These functions are based exclusively on the application of machine learning techniques. A description of the problem and the data set generation procedure for the Crete island power system are included. This article belongs to the Special Issue Advances in the Toxicity Mechanisms of Microcystins Revealed by Omics Approaches ; Microcystins (MCs) are hepatotoxins produced by some cyanobacteria. They are cyclic peptides that inhibit the serine/threonine protein phosphatases (PPs) PP1 and PP2A, especially PP2A. The inhibition of PP2A triggers a series of molecular events, which are responsible for most MC cytotoxic and genotoxic effects on animal cells. It is also known that MCs induce oxidative stress in cells due to the production of reactive oxygen species (ROS). However, a complete characterization of the toxic effects of MCs is still not accomplished. This study aimed to clarify additional molecular mechanisms involved in MC-LR toxicity, using Saccharomyces cerevisiae as eukaryotic model organism. First, a shotgun proteomic analysis of S. cerevisiae VL3 cells response to 1 nM, 10 nM, 100 nM, and 1 µM MC-LR was undertaken and compared to the control (cells not exposed to MC-LR). This analysis revealed a high number of proteins differentially expressed related with gene translation and DNA replication stress; oxidative stress; cell cycle regulation and carbohydrate metabolism. Inference of genotoxic effects of S. cerevisiae VL3 cells exposed to different concentrations of MC-LR were evaluated by analyzing the expression of genes Apn1, Apn2, Rad27, Ntg1, and Ntg2 (from the Base Excision Repair (BER) DNA repair system) using the Real-Time RT-qPCR technique. These genes displayed alterations after exposure to MC-LR, particularly the Apn1/Apn2/Rad27, pointing out effects of MC-LR in the Base Excision Repair system (BER). Overall, this study supports the role of oxidative stress and DNA replication stress as important molecular mechanisms of MC-LR toxicity. Despite efforts to reduce deforestation in the Brazilian Amazon, there has been an up-turn in clearing rates since 2012. These increases are in part due to failures in deforestation control. Soybean planters, cattle ranchers, and timber merchants find ways to circumvent agreements and legislation. Here we explain some of the key problems with the implementation of the principal agreements and Brazilian laws that should be keeping clearing rates under control. To combat increased clearing in the Amazon, we suggest an urgent need to strengthen Brazilian environmental agencies, improve technologies used to monitor the effectiveness of clearing-reduction programmes, better integrate agrarian and environmental policies and integrate environmental enforcement across federal, state and municipal governments, as well as improve transparency along global supply chains and raise awareness among consumers to put market pressure on producers to avoid new deforestation. © 2019 Associação Brasileira de Ciência Ecológica e Conservação

An Amazonian savanna in northern Brazil known as the Cerrado of Amapá is under imminent threat from poor land-use planning, the expansion of large-scale agriculture and other anthropogenic pressures. These savannas house a rich and unique flora and fauna, including endemic plants and animals. However, the area remains under-sampled for most taxa, and better sampling may uncover new species. We estimate that only ~9.16% of these habitats have any kind of protection, and legislative changes threaten to further weaken or remove this protection. Here we present the status of knowledge concerning the biodiversity of the Cerrado of Amapá, its conservation status, and the main threats to the conservation of this Amazonian savanna. To secure the future of these unique and imperilled habitats, we suggest urgent expansion of protected areas, as well as measures that would promote less-damaging land uses to support the local population.

Although Amapa´ is the most protected Brazilian state, the same level of protection does not extend to its savannas. These are currently suffering increased pressure from threats including large-scale agriculture, particularly the expansion of soybean plantations. In September 2016, the Government of Amapa´ presented a zoning proposal (Zoneamento Socioambiental do Cerrado [ZSC]) that reserves most of the savannas for agricultural activities. Here, we outline how the methodology employed is flawed because it does not include fauna surveys, evaluations of ecosystem services or an assessment of the social importance of the savannas. The ZSC authors admit that, contrary to Brazilian legislation, the zoning was carried out with the single intention of increasing agriculture production. Current knowledge indicates that Amapa´'s savannas are rich in biodiversity, including endemic and threatened species, and are also home to a rich culture of traditional populations. These savannas are important providers of ecosystem services that, if intact, could represent around US$ 1.52 billion annually. We hold that the ZSC should be reformulated, with fair participation of stakeholders, in accordance with Brazil's legal requirements. At least 30% of the savannas should be protected, local family farming should be supported, and the rights of traditional peoples must now be assured through recognition of their land rights.
