Bovine tuberculosis (bTB) caused by Mycobacterium bovis is a transmissible disease notifiable to the World Organization for Animal Health and to European Union, with ongoing efforts of surveillance and eradication in every EU member state. In Germany, a country which has been declared officially free from bovine tuberculosis since 1997 by the EU, M. bovis infections still occur sporadically in cattle and other mammals, including humans. Here, the transmission routes of a bovine TB outbreak in a wildlife park in Germany affecting different cervid species, bison, lynx, and pot-bellied pig were followed employing whole genome sequencing (WGS) combined with spoligotyping and MIRU-VNTR-typing. One single M. bovis strain persisted from 2002 – 2015, and transmission between the park and a distantly located captive cervid farm was verified. The spoligotyping pattern remained identical while MIRU-VNTR-typing of 24 loci of the standardized panel and locus 2163a as additional locus revealed one change at locus 2165 in a strain from a fallow deer, and one at locus 2461 in isolates from red deer over the whole time period. WGS analysis confirmed close relatedness of the isolates, with a maximum of 12 SNPs detected between any two sequenced isolates. In conclusion, our data confirm a longitudinal outbreak of M. bovis in a German wildlife park and provide first insights into the dynamics of different genotyping markers in M. bovis.

Die Rindertuberkulose ist eine weltweit bedeutsame Zoonose, welche vornehmlich durch Mycobacterium (M.) bovis und M. caprae verursacht wird. In Deutschland konnte unlängst der nunmehr seit zwei Jahrzehnten bestehende "anerkannt Rindertuberkulose-freie Status" durch die Ergebnisse eines landesweiten Monitorings untermauert werden. Labordiagnostische Weiterentwicklungen haben in den letzten Jahren zur Aufnahme einer differenzierten diagnostischen Kaskade in die Verordnung zum Schutz gegen die Tuberkulose des Rindes (RindTbV) geführt. Die vorliegende Arbeit beschreibt jedoch zwei Fälle von M. tuberculosis-Einträgen in Rinderbestände sowie die damit verbundenen diagnostischen Schwierigkeiten und Probleme für den Vollzug.

Using 894 phylogenetically diverse genomes of the Mycobacterium tuberculosis complex (MTBC), we simulated in silico the ability of the Hain Lifescience GenoType MTBC to differentiate the causative agents of tuberculosis. We propose a revised interpretation of this assay to reflect its strengths (e.g. it can distinguish some strains of M. canettii and variants of M. bovis that are not intrinsically resistant to pyrazinamide) and limitations (e.g. M. orygis cannot be differentiated from M. africanum). Germany has been an officially bovine tuberculosis (bTB)-free (OTF) country since 1996. Gradually rising numbers of bTB herd incidents due to Mycobacterium bovis and M. caprae in North-Western and Southern Germany during the last few years prompted the competent authorities to conduct a nationwide bTB survey in 2013/2014. This led to the detection of a dairy herd in which as many as 55 cattle reacted positively to consecutive intra vitam testing. Test-positive animals lacked visible lesions indicative of bTB at necropsy. Extensive mycobacterial culturing as well as molecular testing of samples from 11 tissues for members of the M. tuberculosis complex (MTC) yielded negative results throughout. However, caseous lymphadenitis of Ln. mandibularis accessorius was observed during meat inspection of a fattening pig from the same farm at regular slaughter at that time. Respective tissue samples tested MTC positive by polymerase chain reaction, and M. tuberculosis T1 family were identified by spoligotyping. Four human reactors within the farmer's family were also found to be immunoreactive. As exposure of livestock to M. tuberculosis is not generally considered, its impact may result in regulatory and practical difficulties when using protocols designed to detect classical bTB, particularly in OTF countries.

Die Überwachung und Bekämpfung vom Tier auf den Menschen übertragbarer Krankheiten (Zoonosen) wurde in der Europäischen Union durch das In-Kraft-Treten einer neuen Zoonosenrichtlinie kürzlich auf eine neue Grundlage gestellt. Eine Überwachungspflicht für Brucellose, Campylobacteriose, Echinokokkose, Listeriose, Salmonellose, Trichinellose und die jeweiligen Erreger, Tuberkulose (soweit sie durch Mycobacterium bovis verursacht ist) sowie Verotoxin bildende Escherichia coli wird festgelegt. Für weitere Zoonosen und Zoonosenerreger gilt die Überwachungspflicht je nach der epidemiologischen Situation. Auf diesem Hintergrund werden der aktuelle Kenntnisstand und die epidemiologische Situation wichtiger von Nutz- und einigen Wildtieren auf den Menschen übertragbarer Infektionskrankheiten in Deutschland dargestellt. ; The control of infectious diseases transmitted from animals to humans (zoonoses) was recently put on a new basis in the European Union when a new Zoonoses Directive entered into force. Brucellosis, campylobacteriosis, echinococcosis, listeriosis, salmonellosis, trichinosis, and the respective causative agents, tuberculosis due to Mycobacterium bovis, and verotoxigenic Escherichia coli must be included in monitoring. Additional zoonoses and zoonotic agents are to be monitored according to the epidemiological situation. Against this background, the current knowledge on important zoonoses transmitted from livestock and some wildlife animals to humans as well as the epidemiological situation in Germany with regard to these diseases is summarized.
