Regulation of the transcriptional program by DNA methylation during human αβ T-cell development

Thymocyte differentiation is a complex process involving well-defined sequential developmental stages that ultimately result in the generation of mature T-cells. In this study, we analyzed DNA methylation and gene expression profiles at successive human thymus developmental stages. Gain and loss of methylation occurred during thymocyte differentiation, but DNA demethylation was much more frequent than de novo methylation and more strongly correlated with gene expression. These changes took place in CpG-poor regions and were closely associated with T-cell differentiation and TCR function. Up to 88 genes that encode transcriptional regulators, some of whose functions in T-cell development are as yet unknown, were differentially methylated during differentiation. Interestingly, no reversion of accumulated DNA methylation changes was observed as differentiation progressed, except in a very small subset of key genes (RAG1, RAG2, CD8A, PTCRA, etc.), indicating that methylation changes are mostly unique and irreversible events. Our study explores the contribution of DNA methylation to T-cell lymphopoiesis and provides a fine-scale map of differentially methylated regions associated with gene expression changes. Signal integration and transcriptional regulation of the inflammatory response mediated by the GM-/MCSF signaling axis in human monocytes
In recent years, the macrophage colony-stimulating factor (M-CSF) and granulocyte-macrophage CSF (GM-CSF) cytokines have been identified as opposing regulators of the inflammatory program. However, the two cytokines are simultaneously present in the inflammatory milieu, and it is not clear how cells integrate these signals. In order to understand the regulatory networks associated with the GM/M-CSF signaling axis, we analyzed DNA methylation in human monocytes. Our results indicate that GM-CSF induces activation of the inflammatory program and extensive DNA methylation changes, while M-CSF-polarized cells are in a less differentiated state. This inflammatory program is mediated via JAK2 associated with the GM-CSF receptor and the downstream extracellular signal-regulated (ERK) signaling. However, PI3K signaling is associated with a negative regulatory loop of the inflammatory program and M-CSF autocrine signaling in GM-CSF-polarized monocytes. En este libro digital el Banco de la República pone a disposición de los investigadores y bibliotecas del país la totalidad de las memorias de Hacienda y Tesoro colombianas que se publicaron en el siglo XIX. Es una herramienta bibliográfica que esperamos sea de enorme utilidad para los profesionales, estudiantes y amantes de la historia de Colombia. El célebre economista austriaco Joseph Schumpeter sostenía que las finanzas públicas son uno de los mejores puntos de partida para investigar una sociedad: allí se reflejan la riqueza, su distribución territorial y sectorial, la marcha de la actividad económica, las prioridades en el gasto y sus beneficiarios. Por su relevancia, y por cuanto los ciudadanos quieren saber en qué se gastan los impuestos que han pagado, los gobiernos democráticos han sido cuidadosos en la rendición de informes periódicos sobre los recaudos, los gastos, los excedentes o déficits en las finanzas gubernamentales y las formas de cubrir estos últimos. Para ello, con cierta regularidad se presentan informes a los órganos de control político o administrativo. Por esa razón, en el Banco de la República hemos considerado que un aporte bibliográfico de gran relevancia para el estudio de la historia económica nacional es poner a disposición de los investigadores y bibliotecas del país esta edición digital de las memorias de Hacienda y Tesoro del siglo XIX.
