Este artigo apresenta e discute um conjunto de estampas que foram concebidas a partir dos arquétipos menstruais propostos por Lara Owen, em seu livro Seu Sangue é Ouro. Os desenhos foram pensados como linguagem de resistência e ressignificação das emoções em torno do ciclo menstrual associado, pelas pessoas que participaram da pesquisa, à vergonha, ao medo e, por vezes, ao desejo de ter nascido com outro corpo. O ciclo menstrual é discutido a partir de aportes críticos dos estudos feministas pós-estruturalistas, da antropologia das emoções contextualista e do design emocional. Tencionamos as abordagens biologizantes em torno da menstruação e das emoções propondo-nos, por meio dos desenhos estampados em camisetas, provocar outros olhares e politizar as emoções em torno da menstruação.

Fluids. 3(1):1-21. https://doi.org/10.3390/fluids3010003 ; [EN] Velocity profiles originated by a pump as turbine (PAT) were measured using an ultrasonic doppler velocimetry (UDV). PAT behavior is influenced by the velocity data. The effect of the rotational speed and the associated flow velocity variations were investigated. This research focuses, particularly, on the velocity profiles achieved for different rotational speeds and discharge values along the impeller since that is where the available hydraulic power is transformed into the mechanical power. Comparisons were made between experimental test results and computational fluid dynamics (CFD) simulations. The used CFD model was calibrated and validated using the same conditions as the experimental facility. The numerical simulations showed good approximation with the velocity measurements for different cross-sections along the PAT system. The application of this CFD numerical model and experimental tests contributed to better understanding the system behavior and to reach the best efficiency operating conditions. Improvements in the knowledge about the hydrodynamic flow behavior associated with the velocity triangles contribute to improvements in the PAT concept and operation. 302 311 208 ; S ; [EN] The use of pumps working as turbines (PATs) connected to the electric system, in the replacement of pressure reduction valves to reduce the excessive pressure in water distribution networks, have been studied for the last years. The introduction of PATs is very important in the water-energy nexus to promote the increase of the energy savings. As consequence, the majority of the water systems does not have access to the electrical grid and, therefore, the need to study the PATS operation off-grid is necessary. In this line, the novelty of this research is the application and optimization of a PAT in water systems when the recovery solution is off-grid type. To operate correctly, the induction machine requires an external source of reactive power, which is typically provided by the electrical grid. To supply the required reactive power, a bank of capacitors is installed at the machine terminals, so-called self-excited induction generator (SEIG). The analytical model, simulation and experimental works were performed, to analyse the SEIG behaviour. The results were applied in a SEIG-PAT system obtaining the global efficiency of the system for different speeds and loads. The global efficiency decreases 47% when off-grid operation, showing the need to optimize the electrical parameters of the generator to operate as off- grid with acceptable efficiency levels. In this framework, a tuning methodology for the SEIG capacitor bank values was developed to be automatically adjusted according to the operating point of the PAT to maximize its efficiency. Studies have shown micro-hydropower (MHP) opportunities for energy recovery and CO2 reductions in the water sector. This paper conducts a large-scale assessment of this potential using a dataset amassed across six EU countries (Ireland, Northern Ireland, Scotland, Wales, Spain, and Portugal) for the drinking water, irrigation, and wastewater sectors. Extrapolating the collected data, the total annual MHP potential was estimated between 482.3 and 821.6 GWh, depending on the assumptions, divided among Ireland (15.5–32.2 GWh), Scotland (17.8–139.7 GWh), Northern Ireland (5.9–8.2 GWh), Wales (10.2–8.1 GWh), Spain (375.3–539.9 GWh), and Portugal (57.6–93.5 GWh) and distributed across the drinking water (43–67%), irrigation (51–30%), and wastewater (6–3%) sectors. The findings demonstrated reductions in energy consumption in water networks between 1.7 and 13.0%. Forty-five percent of the energy estimated from the analysed sites was associated with just 3% of their number, having a power output capacity >15 kW. This demonstrated that a significant proportion of energy could be exploited at a small number of sites, with a valuable contribution to net energy efficiency gains and CO2 emission reductions. This also demonstrates cost-effective, value-added, multi-country benefits to policy makers, establishing the case to incentivise MHP in water networks to help achieve the desired CO2 emissions reductions targets.

Background Half of human cancers harbour TP53 mutations that render p53 inactive as a tumor suppressor. As such, reactivation of mutant (mut)p53 through restoration of wild-type (wt)-like function represents one of the most promising therapeutic strategies in cancer treatment. Recently, we have reported the (S)-tryptophanol-derived oxazoloisoindolinone SLMP53-1 as a new reactivator of wt and mutp53 R280K with in vitro and in vivo p53-dependent antitumor activity. The present work aimed a mechanistic elucidation of mutp53 reactivation by SLMP53-1. Methods and results By cellular thermal shift assay (CETSA), it is shown that SLMP53-1 induces wt and mutp53 R280K thermal stabilization, which is indicative of intermolecular interactions with these proteins. Accordingly, in silico studies of wt and mutp53 R280K DNA-binding domain with SLMP53-1 unveiled that the compound binds at the interface of the p53 homodimer with the DNA minor groove. Additionally, using yeast and p53-null tumor cells ectopically expressing distinct highly prevalent mutp53, the ability of SLMP53-1 to reactivate multiple mutp53 is evidenced. Conclusions SLMP53-1 is a p53-activating agent with the ability to directly target wt and a set of hotspot mutp53. Half of human cancers harbor TP53 mutations that render p53 inactive as a tumor suppressor. In these cancers, reactivation of mutant p53 (mutp53) through restoration of wild-type-like function constitutes a valuable anticancer therapeutic strategy. In order to search for mutp53 reactivators, a small library of tryptophanol-derived oxazoloisoindolinones was synthesized and the potential of these compounds as mutp53 reactivators and anticancer agents was investigated in human tumor cells and xenograft mouse models. By analysis of their anti-proliferative effect on a panel of p53-null NCI-H1299 tumor cells ectopically expressing highly prevalent mutp53, the compound SLMP53-2 was selected based on its potential reactivation of multiple structural mutp53. In mutp53-Y220C-expressing hepatocellular carcinoma (HCC) cells, SLMP53-2-induced growth inhibition was mediated by cell cycle arrest, apoptosis, and endoplasmic reticulum stress response. In these cells, SLMP53-2 restored wild-type-like conformation and DNA-binding ability of mutp53-Y220C by enhancing its interaction with the heat shock protein 70 (Hsp70), leading to the reestablishment of p53 transcriptional activity. Additionally, SLMP53-2 displayed synergistic effect with sorafenib, the only approved therapy for advanced HCC. Notably, it exhibited potent antitumor activity in human HCC xenograft mouse models with a favorable toxicological profile.
