Regions rely not only on their own efforts and characteristics but also on their capacity to attract and assimilate knowledge produced elsewhere to innovate. In other words, interactions among individuals, firms and institutions produce the transmission of knowledge in the form of knowledge spillovers. In the case of knowledge-intensive business services (KIBS), one of their main features is their capacity to adapt and disseminate tacit knowledge. Despite a variety of recent studies have contributed to improve one´s understanding of the tacit knowledge diffuser role of KIBS, there has been little investigation on the spatial effects related to the presence of KIBS. This article represents an attempt to combine in one model knowledge spillovers and availability of a group of KIBS, those called high-tech services (HTS). The objective is to shed some light on the role of both geographical and technological distance in the knowledge diffusion process and to show how HTS account for a significant part of the regional innovation process using an extended knowledge production function (KPF) framework applied to 240 European regions from 23 countries. Two major findings of this analysis are following. For one part, inter-regional knowledge flows are key elements for explaining regional innovation performance, although technological proximity is far less important than geographical proximity. For the other part, regions with a higher specialization in HTS, or proximate to regions with a higher presence of HTS, tend to innovate more, mainly because HTS can improve their capacity to transform knowledge into innovation. In this article we develop a measure to approximate the diffusion of product-embodied R&D that takes place by means of the intermediate purchases of KIBS in 19 of the EU27 countries over the period 2000-2005. The results obtained confirm a rising trend in the use of KIBS as intermediate consumptions as well as the predominance of the domestic provision of this type of services with two exceptions: Ireland and, to a lesser extent, Finland. Leaving aside the two countries with the lowest intensities, Estonia and Hungary, four major groups of countries in terms of the R&D intensity of their KIBS sector were distinguished: very high (Finland, Sweden and Austria), high (Ireland, Czech Republic and Spain), medium (Greece, Belgium, Slovakia, Romania, Slovenia and Germany) and low (Portugal, Netherlands, Poland, Italy and Lithuania). Overall, a high correspondence between R&D intensity and product-embodied R&D diffusion was found, although Ireland, Romania, Belgium and the Netherlands displayed a disappointing diffusion per unit of value added in comparison with the R&D expenditures developed. The existence of weak linkages between the KIBS sector and the rest of industries can be identified as the main explanation for this "diffusion gap". In brief, the results obtained confirm that KIBS are essential industries for improving innovation performance as they carry out strong efforts in R&D that spill over to the rest of the production system. There is a rich debate in the innovation literature about to what extent innovation has become an international (or globalised) phenomenon, or, on the contrary, it maintains its local/regional character. As Koschatzky (2001) notes, given the fact that knowledge is commonly tied to personal capabilities; it has a clear geographical component. In the case of knowledge-intensive services (KIS) most of analyses come to the same conclusion: distance is particularly relevant when knowledge (mainly of a tacit type) is diffused. Starting from this premise, a burgeoning literature on the contribution of those knowledge-intensive business services (KIBS) to regional innovation has emerged. Most of these papers adopt a national perspective, that is, analyse regions in a specific country. On the contrary, comparisons of regional features have been carried out in very few papers: Germany and the UK (Simmie and Strambach, 2006) or Germany and France (Muller and Zenker, 2001) are two examples. The objective of this paper is to take a step further and examine the distribution of knowledge-intensive services (KIS) in the European regions. For so doing we employ the data provided by the Regional Innovation Scoreboard (RIS) 2009. This database provides information on the innovation performance across 194 regions of the European Union and Norway. The methodology employed is known as spatial analysis and evaluates whether there are clusters in the location of KIS in the European regions, which involves three processes. First, to evaluate the existence of spatial autocorrelation by means of global statistics; the Moran's I and the Geary's C. Once verified the existence of positive spatial autocorrelation, it is possible to identify "clusters" of regions with high and low participations of KIS by using a local indicator of spatial autocorrelation (LISA). Finally, employing an econometric model, some potential explanatory factors for the concentration of KIS are examined. The results obtained support the hypothesis that KIS are spatially concentrated and confirm that spatial clusters are different in northern/central and southern/eastern regions. Moreover, a close relationship between location of KIS and regional innovation performance is found.
Background: Cutaneous forms of leishmaniosis due to Leishmania braziliensis have been reported in horses in the New World. Domestic animals play a role in the transmission of the disease. In Costa Rica, human cases of L. braziliensis, L. panamensis and L. infantum have been reported. Objectives: The present report describes five cases of equine cutaneous leishmaniosis in Costa Rica. The aetiological diagnosis was based on the presence of the parasite within the lesions. Methods: Skin biopsies were used to perform histopathological analyses of the lesions. Immunohistochemistry was used to detect the presence of the Leishmania spp. antigens in tissue sections. Laser-capture micro-dissection and quantitative real-time PCR techniques were carried out to detect the pathogen nucleic acid within the microscopic lesions. Results: Histopathological analyses showed a granulomatous inflammation within the dermis, with multi-nucleated giant cells, macrophages, lymphocytes and few neutrophils and eosinophils. We detected the parasite by immunohistochemistry, using a rabbit polyclonal antibody raised against Leishmania spp. However, we could not identify Leishmania spp. by quantitative real-time PCR in formalin-fixed paraffin-embedded tissues, using specific primers for the conserved region in the minicircle of the Leishmania DNA kinetoplast. Conclusions: Our results emphasise the importance of Leishmania spp. not only as a causative agent of equine cutaneous disease in the New World, but also as a possible emerging pathogen. South American camelids are susceptible to tuberculosis, caused mainly by Mycobacterium bovis and M. microti. Despite the tuberculin skin test being the official test for tuberculosis, it has a very low sensitivity in these species (14-20%). Serological tests present the advantages of being rapid, easy to perform and facilitate analysis of large numbers of samples in a short period of time. Novel antigen discovery and evaluation would provide enhanced detection of specific antibodies against members of M. tuberculosis complex. Here, we describe the development and evaluation of an ELISA-type immunoassays to use in the diagnosis of tuberculosis in llamas and alpacas based on P22, a multiprotein complex obtained by affinity chromatography from bovine Purified Protein Derivative (bPPD), that showed high sensitivity and specificity in mice, cattle and goats. This work was performed in two stages. First, a preliminary panel of samples collected from tuberculosis-free (n = 396) and M. bovis-infected herds (n = 56) was assayed, obtaining high specificity (100%) and sensitivity ranging from 63 to 96%. Subsequently, the use of the serological assay was tested using samples from two herds suffering from clinical M. bovis (n = 88) and M. microti (n = 25) infection to evaluate the ability of the ELISA to detect infected animals. 11 out of 88 alpacas were positive to the ELISA in a M. bovis outbreak and 7 out of 25 in a M. microti outbreak. C3 is the central component of the complement system. Upon activation, C3 sequentially generates various proteolytic fragments, C3a, C3b, iC3b, C3dg, each of them exposing novel surfaces, which are sites of interaction with other proteins. C3 and its fragments are therapeutic targets and markers of complement activation. We report the structural and functional characterization of four monoclonal antibodies (mAbs) generated by immunizing C3-deficient mice with a mixture of human C3b, iC3b and C3dg fragments, and discuss their potential applications. This collection includes three mAbs interacting with native C3 and inhibiting AP complement activation; two of them by blocking the cleavage of C3 by the AP C3-converase and one by impeding formation of the AP C3-convertase. The interaction sites of these mAbs in the target molecules were determined by resolving the structures of Fab fragments bound to C3b and/or iC3b using electron microscopy. A fourth mAb specifically recognizes the iC3b, C3dg, and C3d fragments. It binds to an evolutionary-conserved neoepitope generated after C3b cleavage by FI, detecting iC3b/C3dg deposition over opsonized surfaces by flow cytometry and immunohistochemistry in human and other species. European badgers (Meles meles) have been identified as wildlife reservoirs for Mycobacterium bovis in the UK and Ireland, and may also have a role in the epidemiology of animal tuberculosis in other European regions. Thus, detection of M. bovis-infected badgers may be required for the purposes of surveillance and monitoring of disease levels in infected populations. Current serological assays to detect M. bovis infection in live badgers, while rapid and inexpensive, show limited diagnostic sensitivity. Here we describe and evaluate new ELISA platforms for the recognition of the P22 multiprotein complex derived from the purified protein derivative (PPD) of M. bovis. The recognition of IgG against P22 multiprotein complex derived from PPD-B was tested by ELISA in the serum of badgers from the UK, Ireland and Spain. TB infection in the badgers was indicated by the presence of M. bovis in tissues by culture and histology at post-mortem examination and TB-free status was established by repeated negativity in the interferon γ release assay (IGRA). In experimentally infected badgers, humoral antibody responses against P22 developed within 45 days post-infection. The ELISA tests showed estimated sensitivity levels of 74-82% in experimentally and naturally infected badgers with specificities ranging from 75% to 100% depending on the badger population tested. The ante-mortem diagnosis of tuberculosis (TB) in ruminants is based mainly on the intradermal tuberculin test and the IFN-γ assay. Antibody (Ab)-based tests have emerged as potential tools for the detection of TB infected animals using serum, plasma, or even milk samples. Oral fluids have also been evaluated as alternative samples with which to detect specific Abs against Mycobacterium bovis in pigs or wild boars, but not in ruminants. The objective of this study was, therefore, to evaluate the performance of an in house-ELISA for TB diagnosis (P22 ELISA) in goats as an experimental model for the diagnosis of TB using oral fluid samples. Oral fluid samples from 64 goats from a TB-infected herd (n = 197) and all the animals from a TB-free herd (n = 113) were analyzed using the P22 ELISA. The estimated sensitivity (Se) and specificity (Sp) were 34.4% (95% CI: 22.4-45.6) and 100% (95% CI: 97.4-100), respectively. The optimal cut-off point was set at 100% according to the ROC analysis. Those animals with a higher level of Abs in their oral fluid attained a higher lesion score (p = 0.018). In fact, when taking into account only the setting of the animals with severe lesions (n = 16), the ELISA showed a Se of 75% (95% CI: 53.7-96.2). Results of the present study suggest that the P22 ELISA is highly specific but has a limited value detecting infected animals in oral fluid samples. BACKGROUND: Serum antibody detection has potential as a complementary diagnostic tool in animal tuberculosis (TB) control, particularly in multi-host systems. The objective of the present study was to assess the specificity (Sp) of an enzyme-linked immunosorbent assay (ELISA) based on the new multiprotein complex P22 for the detection of specific antibodies against the Mycobacterium tuberculosis complex (MTC) in the four most relevant domestic animals acting as MTC hosts: cattle, goat, sheep and pig. We used sera from an officially TB-free (OTF) country, Norway, and from a non-OTF one, Spain. The samples included sera from goats that had been vaccinated against M. avium subsp. paratuberculosis (MAP) and sheep from a herd in which Corynebacterium pseudotuberculosis had been isolated. RESULTS: In cattle, the Sp ranged from 92.5 (IC95% 90.7-94) to 99.4% (IC95% 98.3-99.8) depending on the cut-off used and the origin of the samples (Spain or Norway). Sp in cattle (cut-off point 100) was significantly higher (P < 0.05) for Norwegian samples. By contrast, Sp in goats was consistently low at the 100 cut-off [30.9 (CI95.4-39.5)-78% (CI95% 68.9-85)]. A higher cut-off of 150 improved Sp in Norwegian goats [97% (CI95% 91.6-99)], but still yielded a poor Sp of 56.1% (CI95% 47.3-64.6) in Spanish goats. In Norway at the 100 cut-off the Sp was 58.3 (CI95% 42.2-72.9) and 90.6% (CI95% 81-95.6) in MAP vaccinated and non-vaccinated goats, respectively, indicating interference due to MAP vaccination. Sp in sheep was between 94.4 (CI95% 91.7-96.3) and 100% (CI95% 96.3-100) depending on the cut-off and country, and no diagnostic interference due to infection with C. pseudotuberculosis was recorded. Sp in pigs was 100%, regardless the cut-off point applied, and no significant differences were observed between pigs from Norway and from Spain. Background: Bovine purified protein derivative (bPPD) and avian purified protein derivative (aPPD) are widely used for bovine tuberculosis diagnosis. However, little is known about their qualitative and quantitative characteristics, which makes their standardisation difficult. In addition, bPPD can give false-positive tuberculosis results because of sequence homology between Mycobacterium bovis (M. bovis) and M. avium proteins. Thus, the objective of this study was to carry out a proteomic characterisation of bPPD, aPPD and an immunopurified subcomplex from bPPD called P22 in order to identify proteins contributing to cross-reactivity among these three products in tuberculosis diagnosis. Methods: Trypsin digests of bPPD, aPPD and P22 were analysed by nanoscale liquid chromatography-electrospray ionization tandem mass spectrometry. Mice were immunised with bPPD or aPPD, and their serum was tested by indirect ELISA for reactivity against these preparations as well as against P22. Results: A total of 456 proteins were identified in bPPD, 1019 in aPPD and 118 in P22; 146 of these proteins were shared by bPPD and aPPD, and 43 were present in all three preparations. Candidate proteins that may cause cross-reactivity between bPPD and aPPD were identified based on protein abundance and antigenic propensity. Serum reactivity experiments indicated that P22 may provide greater specificity than bPPD with similar sensitivity for ELISA-type detection of antibodies against M. tuberculosis complex.
