Wetlands of the same kind can present considerable difference in relation to a series of environmental variables that influence on the physicochemical properties and on the biological communities sustained, natural variability scarcely considered in comparative studies. The aim of this work was to provide a first approach to the edaphological, morphological and climatic classification of the forested wetlands from central Chile, unique environments for the conservation of an interesting floral and faunal diversity. 18 wetlands were classified in the Araucanía Region, by means of the B classification procedure by the European Union Water Framework Directive. 21 variables at basin scale were used. Four ecotypes were obtained, which were characterized mainly by microclimates and soil types, though were also relevant morphological variables such as slope, slope of the wetlands, and the basins towards they drain (Wilks's Lambda 3.84). Ecotypes determined were defined by: 1) convergence of moderate marine, warm and Mediterranean climates with silty clay loam soil and high slopes, 2) moderate marine climate with silty loam soil and high slopes, 3) Cold Mediterranean climate with silty loam soil and low slopes and 4) Moderate marine climate, silty clay loam soil and high slope. Classification is in agreement with in situ observations. Nevertheless, it must be validated by both limnological and biological approaches. ; Humedales del mismo tipo pueden presentar diferencias considerables en relación a una serie de variables ambientales que influyen en las propiedades fisicoquímicas y a su vez en las comunidades biológicas que sustentan. Sin embargo, en estudios comparativos, esta variabilidad natural es generalmente poco considerada. El objetivo de este trabajo fue realizar una primera aproximación de una clasificación edafológica, morfológica y climática de los humedales boscosos de la zona central de Chile, ecosistemas únicos para la conservación de una interesante diversidad de flora y fauna. Se clasificaron 18 humedales presentes en la región de la Araucanía, mediante el procedimiento de clasificación B de la Directiva Marco del Agua de la Unión Europea, utilizando 21 variables a escala de cuenca. Se obtuvieron cuatro ecotipos, caracterizados principalmente por el microclima y tipo de suelo, aunque también fueron relevantes variables morfológicas, como la pendiente y la superficie de sus cuencas (Lambda de Wilks 3,84). Los ecotipos determinados estuvieron definidos por: 1) Convergencia de climas marino fresco, cálido y mediterráneo frio, con suelo franco arcillo limoso y pendientes altas, 2) Clima marino fresco con suelo franco limoso y pendientes altas, 3) Clima mediterráneo frio con suelo franco limosos y pendientes bajas y 4) Clima marino fresco, suelo franco arcillo limoso y pendiente alta. La clasificación debe ser posteriormente validada mediante observaciones in situ de variables limnológicas y biológicas.

Las evaluaciones ecotoxicológicas son herramientas fundamentales en las estimaciones de riesgo ambiental y la toma de decisiones de políticas de control y prevención de daños ecológicos. A nivel regional la caracterización ecotoxicológica de sedimentos se determina principalmente evaluando en ellos niveles de contaminantes considerados como prioritarios, sin tener en cuenta factores tales como biodisponibilidad, resuspensión posibles interacciones o impacto sobre estructura y funcionamiento de las comunidades asociadas a sedimentos. En ese sentido los bioensayos son herramientas esenciales para evaluar la calidad del ambiente porque responden de manera integrada a los efectos adversos de mezclas químicas complejas con las que están en contacto y cuya ecotoxicidad interesa conocer. Los bioensayos con anfípodos tienen considerable relevancia en la evaluación toxicológica de agua dulce, siendo Hyalella azteca, de amplia distribución en el hemisferio norte, la especie más utilizada. Los protocolos para bioensayos estandarizados fueron diseñados en el hemisferio norte en función de los requerimientos y biología de esta especie. Además, estos protocolos se centran primordialmente en la evaluación de parámetros tradicionales (mortalidad y crecimiento) sobre los anfípodos, asumiendo que la ausencia de efecto indica que el medio de ensayo es ecotoxicológicamente aceptable. Sin embargo, hay una tendencia mundial a evaluar el impacto de los contaminantes en un sistema acuático particular a través del monitoreo ecotoxicológico con especies nativas. La necesidad de utilizar especies locales se hace evidente, entre otras razones, porque el uso de las mismas adaptadas a las características del ambiente, pueden proporcionar resultados mucho más cercanos a la realidad que los obtenidos con una especie exótica. Por lo tanto, debido a que las especies propuestas en los protocolos internacionales no son las mismas que encontramos en nuestros ecosistemas acuáticos, es muy importante evaluar el efecto de los contaminantes sobre especies autóctonas sobre la base del conocimiento biológico, ya que estas pueden mostrar requerimientos y características diferentes a las de las especies estandarizadas en dichos protocolos. Es recomendable que cada región geográfica tenga protocolizado diversos métodos de ensayo donde las especies autóctonas estén representadas. Este trabajo presenta resultados obtenidos a partir de la evaluación de efectos de matriz acuosa y sedimentos contaminados sobre dos especies de anfípodos dulceacuícolas presentes en la región: Hyalella curvispina y Hyalella pseudoazteca, por medio de bioensayos de toxicidad en condiciones controladas de laboratorio. Utilizando organismos de dos estadios de vida, se realizaron ensayos de exposición aguda y subcrónica a dos metales pesados, tóxicos referentes (cobre y cadmio) en matriz acuosa, sedimento procesado y agua de poro extraída del mismo, adaptando una metodología estandarizada para H. azteca. Los puntos finales fueron concentración de metal en tejidos, sobrevivencia y crecimiento, siendo los dos últimos parámetros tradicionales para estos protocolos experimentales. A su vez, se investigó la utilidad del Índice de Masa Corporal (IMC) como nuevo parámetro evaluador de efecto subletal. Con el objetivo de utilizar nuevos puntos finales de evaluación de efecto temprano por exposición a metales en H. curvispina, se determinaron efectos sobre el contenido de glucógeno, proteínas totales, lípidos totales, triglicéridos, arginina fosfato, niveles de peroxidación lipídica (TBARS) y actividad de catalasa (CAT), superóxido dismutasa (SOD) y Na+/K+ ATPasa, en matriz sólida y acuosa. Además se ajustó e implementó para esta especie la técnica de sistema de transporte de electrones (ETS) como biomarcador de estado metabólico. Se llevó a cabo la evaluación de la ecotoxicidad de sedimentos ambientales realizando ensayos con sedimento completo y agua de poro, utilizando a H. curvispina como organismo prueba, evaluando como puntos finales parámetros tradicionales de efecto. A su vez, sobre muestras de sedimento completo se investigó la utilidad de biomarcadores de efecto temprano (niveles de lipoperoxidación y actividad de SOD, CAT y ETS). En relación al efecto del cadmio y cobre en solución H. curvispina probó ser sensible a concentraciones ecológicamente relevantes de ambos metales. Los resultados reafirman su importancia como una especie adecuada para ensayos de evaluación ecotoxicológica en cuerpos de agua dentro de su área de distribución. Con respecto a la toxicidad de ambos metales en bioensayos en matriz sólida, se demostró que ambas especies sufren efectos negativos sobre el crecimiento a concentraciones de Cd y Cu cercanas a los Niveles Guía de Calidad de Sedimentos (SQGs), desarrollados para el hemisferio norte, siendo el IMC el mejor parámetro discriminador de efecto. En cuanto al desarrollo de nuevos puntos finales de efecto temprano para ensayos con anfípodos se confirmó que la exposición a Cu ó Cd, tanto en solución como en sedimento, afectó de manera significativa a la mayoría de los parámetros metabólicos y bioquímicos evaluados. Se postula al ETS como nuevo biomarcador de efecto en bioensayos ecotoxicológicos con H. curvispina. El protocolo aplicado, los parámetros determinados y la especie test elegida, resultaron ser aptos para monitorear la calidad toxicológica de sedimentos ambientales. El uso de biomarcadores demostró una mayor sensibilidad y un más preciso ajuste de evaluación de efectos que el obtenido por el ensayo estándar de sobrevivencia y crecimiento, y se revelan más contundentes en relación a la evaluación de efectos proponiéndose su implementación en estudios de biomonitoreo de sedimentos. Por último, se estableció una línea de base como contribución al incremento del conocimiento biológico de las especies de ensayo, utilizando como criterio el IMC y parámetros del metabolismo energético, comparando el estado general de los animales en medio acuoso y sedimento en situaciones control. Se concluyó que H. curvispina puede ser empleada en bioensayos tanto en matriz sólida como acuosa, pero la especie resulta más apropiada para la realización de bioensayos en presencia de sedimentos. Con respecto a H. pseudoazteca resulta una especie apropiada para su uso en bioensayos en matriz sólida, pero no es recomendada para su utilización en ensayos en matriz acuosa. ; Ecotoxicology assays are fundamental tools for assessing environmental risks and making political to control and prevent environmental damage. At a regional level, ecotoxicological characterization of sediments is determined mostly by analysing the toxic levels present in them that are prioritized, not taking into account variables such as bioavailability, resuspension, possible interactions or impact on the structure and functioning of the communities associated to the sediments. In this sense, bioassays are essential tools for assessing environmental quality because they respond in an integral way to adverse effects of complex chemical mixtures with which they come in contact and of which there is an interest in knowing their ecotoxicity. Bioassays with amphipods are of remarkable relevance for toxicological assessment in freshwater, being Hyalella azteca, widely distributed across the northern hemisphere, the most employed species. Standardized bioassay protocols were designed in the northern hemisphere based on the requirements and biology of this species. Moreover, these protocols are focused mainly on the assessment of traditional parameters (growth and mortality) of amphipods, assuming the absence of an effect implies that the assay medium is ecotoxicologically acceptable. However, there is a worldwide trend to assess toxic impact in a particular aquatic system through ecotoxicological monitoring with native species. The need to use local species becomes evident, among other reasons, because using these adapted to the environmental characteristics can give results which are much closer to reality than those obtained using an exotic species. Therefore, considering the species proposed by international protocols are not the ones we find in our aquatic ecosystems, it is extremely important to assess the effect of pollutants on native species on the basis of biological knowledge, since native species may present requirements and characteristic that are different to those of the standardized species in said protocols. It is advisable that each geographic region have a diversity of standardized assay protocols where native species are represented. This work presents results obtained from the assessment of the effects of polluted sediments and liquid matrix on two freshwater amphipod species present in the region: Hyalella curvispina and Hyalella pseudoazteca, through toxicity bioassays in controlled laboratory conditions. Acute and subchronic exposure to two heavy metals, reference toxics (copper and cadmium) in liquid matrix, processed sediment and groundwater assays were performed, adapting a methodology standardized for H. azteca, using organisms at different life stages. Endpoints were metal tissue concentration, survival and growth, being these last two the traditional parameters for these experimental protocols. At the same time, utility of the body mass index (IMC) as a new parameter to evaluate sublethal effects was assessed. Aiming to use new endpoints to evaluate early effects of metal exposure in H. curvispina, the effects on glycogen, total proteins, total lipids, triglycerides, phosphate arginine, lipids peroxidation levels (TBARS) and catalase activity (CAT), superoxide dismutase (SOD) and Na+/K+ ATPase were determined in both solid and liquid matrix. In this work, the electron transportation system (ETS) technique was adjusted and implemented for this species as a metabolic state biomarker. Environmental sediment ecotoxicity assessment was carried out with complete sediment and pore water assays using H. curvispina as test organism, taking as endpoint the traditional effect parameters. Utility of early effect biomarkers (lipoperoxidation levels and SOD, CAT and ETS activity) was also assessed using complete sediment samples. With regard to the effect over cadmium and copper in solution, H. curvispina showed sensitivity to concentrations of both ecologically relevant metals. The results reasserted its significance as an adequate species for ecotoxicological assessment assays in water bodies inside their area of distribution. As for the toxicity of both metals in bioassays with solid matrix, it showed that both species suffer negative effects on growth when using concentrations of Cd and Cu close to the Sediment Quality Guideline (SQG) levels (developed for the North hemisphere), with the IMC being the best parameter for effect discrimination. With regard to developing new early effect endpoints for assays with amphipods, it was confirmed that the exposition to both metals significantly affected most of the intermediary metabolism parameters and biochemical oxidative damage evaluated, both in solution and in sediment. ETS is presented as a biomarker of effect in ecotoxicological bioassays with H. curvispina. The applied protocol, the determined parameters and the test species chosen were adequate for monitoring the toxicological quality of environmental sediments. The use of biomarkers showed a higher sensitivity and a more precise effect evaluation adjustment than the one obtained through the use of the standard survival and growth assay. Since they are also more telling in regards to the effects evaluation, its implementation in sediment biomonitoring studies is proposed. Finally, a baseline was established as contribution to the increment of biological knowledge of the test species, using the IMC and parameters from the energetic metabolism as criteria, comparing the general state of the animals in water and sediment media in control situations. It is concluded that H. curvispina can be used in bioassays with both solid and water matrix, but the species is more adequate for carrying out bioassays in presence of sediments. With regard to the H. pseudoazteca, it is an adequate species to be used in solid matrix bioassays, but it is not recommended for its use in water matrix trials. ; Fil:Giusto, Anabella. Late 20th. Century malacology in Mexico represents the fusion of two lines of action from the previous century: natural history museums established in the 18th. Century and field collecting that began in the 16th Century. Museum activity suffered from political influence, mainly in the 19th century. Field work by expeditions from Europe and the USA were important around the 19th century (e.g. the large works by Fischer and Crosse, 1870-1902; von Martens, 1890-1901; Strebel, 1873-1882 and Díaz de León, 1912), but continental mollusc research done by Mexicans was very sporadic (specially in the 19th Century, less in the 20s and 50s of the 20th century) until 1980. ; Dos vertientes de desarrollo, que al final del siglo XX se fusionan, son los antecesores de la malacología continental actual en México. Una vertiente comprende la actividad desempeñada en los museos de historia natural (que inicia hacia el final del siglo XVIII) y la otra son las exploraciones del territorio mexicano (que inician hacia finales del siglo XVI). La actividad en los museos está muy influenciada por los acontecimientos políticos (principalmente en el siglo XIX) y tal vez refleja la inestabilidad de esa época. La investigación realizada por mexicanos con respecto a moluscos continentales es esporádica. Inicia hacia el final del siglo XIX y, ya en el siglo XX se realizan algunos trabajos, entre los años veinte y cincuenta; pero es hasta después de 1980 cuando surge la investigación formal del grupo. En cambio, el espíritu exploratorio de europeos y estadounidenses, tuvo su máxima expresión en el siglo XIX, aunque antes y después de ese siglo también hubieron exploraciones. El resultado de esas exploraciones dará lugar a varios inventarios monumentales (Fischer y Crosse 1870-1902, von Martens 1890-1901, Strebel 1873-1882 y Díaz de León 1912) y descripción de nuevas especies en múltiples publicaciones la más de las veces foráneas.
ABSTRACT In Colombia many rivers and other inland aquatic systems, especially into the Andean Region (the most populated of the country), suffer several anthropogenic impacts related to land use and land cover compromising their water quality and quantity, and the provision of other goods and ecosystem services for the future. It is imperative that the Colombian government may initiate new and updated policies, plans, programs in constant consultation and cooperation with other institutions, civilian and environmental authorities, and researchers from the country (or elsewhere), to obtain amply responsibility on the sustainable river management, especially in this region. Thus, a more integrative watershed river assessment and related riverine issues are necessaries for the current freshwater use and management, and before that new development projects going to start. Colombia is a geographically diverse country mainly due to the Andean terrain, which creates a river system that combines different orders of drainages, physiography and water types, located in four areas: Caribbean, Pacific, Orinoco and Amazon. Hydrographic areas or basins are classified based on the watershed and have been treated regionally only in terms of political division to manage watersheds sections. A regional classification of freshwater ecosystems that combines both geographical and biological criteria, does not exist for Colombia, so the territorial planning and decision-makers do not consider aquatic ecosystems as a unit in their management guidelines. Therefore, the objective of this research was to propose continental hydro-biological units based on hydro-geographic and biological criteria, regrouping official hydrographic areas according to their fish species composition and the interpretation of their drainage network and geomorphological characteristics. There are 28 ecoregions proposed: six for the Caribbean region, eight for the Andean, five for the Pacific and nine for cis-Andean (five for Orinoco and four for Amazonas). These ecoregions can be used as a tool of territorial planning. ; Colombia es un país geográficamente heterogéneo debido principalmente a la orografía andina, la cual propicia una red hidrográfica que combina diferentes órdenes de drenajes, fisiografías y tipos de agua, ubicados en cuatro vertientes: Caribe, Pacífico, Orinoquia y Amazonia. La actual presi?n que actividades productivas tales como miner?a, generaci?n de energ?a el?ctrica, urbanizaci?n, construcci?n de obras civiles, expansi?n de la frontera agr?cola ejercen sobre los ecosistemas estrat?gicos es muy grave, m?s a?n teniendo en cuenta que, a nivel local y regional, muchas de estas ?reas ?nicas no se han delimitado y por esta raz?n no son incluidas en los planes de ordenamiento territorial. La regi?n del Oriente antioque?o es una de las zonas de mayor crecimiento econ?mico en el pa?s, lo cual ha tra?do consigo una constante migraci?n de personas y la necesidad de un incremento sustancial en los servicios ecosist?micos de provisi?n. Esta tesis tiene como objetivo investigar los efectos del etanol y metales pesados (cadmio y mercurio) en los patrones diarios de dos especies acuáticas clave: pez cebra (Darío rerio) y anfípodo gammarus (G. aequicauda y G. chevreuxi). La investigación se realizó en dos niveles organizativos: comportamental y molecular. Con este fin, hemos establecido cuatro objetivos específicos: 1. Investigar los efectos al etanol en el pez cebra en función en su ritmo circadiano. 2. Determinar la existencia de un ritmo diario de la toxicidad del cadmio en el pez cebra. 3. Caracterizar en Gammarus chevreuxi los efectos en la actividad locomotora debido a sedimentos contaminados con mercurio. 4. Evaluar la calidad ambiental de los sedimentos de la cuenca sur del Mar Menor influenciados por históricas actividades mineras. Por lo tanto, estos nuevos datos tiene la intención de señalar la importancia de aplicar un enfoque multidisciplinario para estudios toxicológicos, así como elevar la importancia de la elección del momento adecuado para realizar el ensayo de toxicidad Metodología Los experimentos llevados a cabo en la presente investigación siguieron la legislación española sobre protección de animales y prácticas de laboratorio y fue aprobado tanto por el Comité Nacional de Protección de los Animales y el Comité de Bioética de la Universidad de Murcia - Los muestreo de crustáceos se realizaron en las zonas próximas a la costa mediante el uso de redes de 5 µ de luz de maya. Y su trasporte con aireación y refrigeración . Los muestreos de pez cebra se realizaron en las cámaras del departamento de Fisiología animal. - El etanol se adquirió a través de distribuidores comerciales y los sedimentos tóxicos directamente de las zonas de estudio mediante muestreos con barca. Se almacenaran en refrigeradores a una temperatura de 4ºC. - Los test de mortalidad y comportamiento se realizaran en cámaras herméticas donde se permite el control de las condiciones de fotoperiodo y temperatura. - Las condiciones del agua antes y después de las exposiciones se medió con electrodos de pH y Oxígeno. - Lo análisis de la actividad ante exposiciones subletales se realizaron en las mismas cámaras y mediante la utilización de equipos de filmación sensibles a las luces infrarrojas, permitiendo así mantener las condiciones de oscuridad en los experimento que sea necesarios. - Determinación de metales pesados en tejidos, utilizamos La Espectroscopia de Emisión en Plasma Acoplado Inductivamente (ICP-AES) con Potencia RF: 1200W, Velocidad de bomba: 30r.p.m., Flujo de gas aux.:0.5l/min, Flujo de gas de nebulización: 0.5 l/min, Flujo de gas de refrigeración: 14 l/min, Flujo de gas de purga: Normal. - Para el estudio de expresión de diferentes genes realizamos la técnica de PCR cuantitativa utilizando SYBR Green PCR Master Mix (Applied Biosystems, Foster City, CA) y un aparato ABI Prism 7500 (Applied Biosystems, Foster City, CA). Conclusiones 1. La exposición de pez cebra mediante baño a etanol a diferentes horas del día muestra un ritmo de toxicidad que refleja una mayor tasa de mortalidad en larvas durante el día comparado con los efectos durante la noche. Así mismo en adultos de pez cebra se observan efectos en su actividad natatoria y posición en la columna de agua siendo significativamente más severos durante la fase de luz. 2. La exposición de pez cebra mediante baño a cadmio durante diferentes horas del día reflejó un incremento de la mortalidad y acumulación del cadmio en tejidos durante la mitad de la fase de luz, así como una mayor reducción de la actividad locomotora cuando el pez es expuesto a concentraciones subletales, además la expresión de mt1 muestra un pico antes al final de la fase de luz. Todos estos resultados indican que el pez cebra es más vulnerable al cadmio durante la fase de luz que en la fase de oscuridad. 3. La exposición de Gammarus chevreuxi mediante sedimentos contaminados con mercurio mostró un efecto deletéreo en la actividad natatoria incluso en concentraciones consideradas no letales. Dichos efectos fueron más acusados durante el la fase de luz, que corresponde con la más activa del animal. 4. Nuestros resultados sugieren que la actividad locomotora y el número de surfacings se presentan cono una señal temprana de la interacción de los factores co-estresantes temperatura y sedimentos contaminados. Los parámetros comportamentales estudiados en Gammarus. aequicauda y Gammarus. chevreuxi tienen una conexión clara con la supervivencia de los organismos y pueden tener una relevancia ecológica. This thesis aims at investigating the effects of Ethanol and heavy metals (cadmium and mercury) on the daily patterns of two key aquatic species: zebrafish (Dario rerio) and amphipod gammarus (G. aequicauda and G. chevreuxi). The research was conducted in two organizative levels: Physiology behavior and molecular. With this purpose we have established four particular objectives: 1. To investigate time-dependent effects of ethanol exposure in zebrafish. 2. To determine the existence of a daily rhythm of cadmium toxicity in zebrafish. 3. To characterise the effects of locomotor activity to mercury-contaminated sediments in Gammarus chevreuxi. 4. To evaluate the environmental quality of the soft-bottoms from the southern basin of the Mar Menor lagoon influenced by historical mining activities. Hence, this new data intends to point out the importance of applying a multidisciplinary approach to toxicology studies, and also raise the importance of the time experimental setting of toxicity test. Methodology The experiments performed in the present research followed Spanish legislation on Animal Welfare and Laboratory Practices and was approved by both the National Committee on Animal Welfare and the Bioethics Committee of the University of Murcia. - The sampling of gammarids were conducted in the nearshore areas by using mesh of 5 μ. Their transportation was with aeration and cooling. The zebrafish samplings were performed in Animal Physiology's chambers of the department. - Ethanol was purchased by authorized distributors, and toxic sediments were extracted directly from the study areas by boat sampling. Sediments extraction was with cores and dredge. They were stored in the refrigerator at 4 ° C. - The test of mortality and behavior was conducted in chambers where control of the photoperiod and temperature conditions allowed. - Water conditions before and after exposure was measured with pH electrodes and oxygen. - The analysis of activity at sublethal exposures were conducted in the same chamber and using equipment sensitive to infrared light filming, thereby maintaining the conditions of darkness in the experiment is necessary. - Determination of heavy metals in tissues, we used emission spectroscopy Inductively Coupled Plasma (ICP-AES) RF Power: 1200W, pump speed: 30r.pm, aux gas flow: 0.5l / min gas flow. The exposure of Gammarus chevreuxi by mercury-contaminated sediments showed a deleterious effect on the swimming activity even considered non-lethal concentrations. In both cases a loss of freshwater occurs. So in order to minimize this effect, the construction of a retention dam was suggested in a previous work.
