Screening of sporulating bacterial strains of genus Bacillus isolated from various natural sources was carried out. The most active antagonistic bacteria were chosen. Probiotic properties of the selected spore-farming Bacillus were investigated: spectrum of antimicrobial action, safety for macroorganism, survival in transit via gastrointestinal tract (bile and acid resistance), lack of inhibiting effect on gut microbiota, native non-plasmid resistance to widely applied antimicrobial agents. The prospects of using the most active cultures as key components of probiotics and feed additives for farm stock, poultry and fish were demonstrated.

Probiotics are live microorganisms present in naturally fermented food products, and also added to other products as supplements to improve the hosts' health and microbial balance. Bacteria are considered as probiotics based on selection criteria that include the ability to survive the transition through the gastrointestinal tract conditions (pH and bile salt concentration), adhesion to the intestinal epithelium, auto-aggregation, and antibiotic resistance. The industrial properties of probiotic bacteria associated with their incorporation into food products are essential for the application of probiotic cultures in the development of functional foods. Probiotic bacteria must survive industrial applications, grow adequately in food products during their shelf life, and be technologically suitable for their incorporation into foods products so that they retain viability and efficacy. The antimicrobial activity of probiotic bacterial strains against foodborne pathogenic bacteria may also be a characteristic parameter for probiotics to be included in the composition of probiotic preparations and probiotic foods. This review discusses the in vitro and industrial properties of bacteria isolated from a variety of fermented food products.

Encapsulation with edible films is a promising approach that may solve the disadvantages associated with the use of bioactive compounds as food additives. This is particularly relevant in the case of probiotics, since their stability in food matrices and in the gastrointestinal tract may be rather poor. Therefore, new cellulose-based edible films have been successfully developed and characterized. Sodium carboxymethyl cellulose (CMC) and hydroxyethyl cellulose (HEC) were used for the film preparation and cross-linked with citric acid (CA) under reasonably mild conditions. Model probiotic bacteria (Lactobacillus rhamnosus GG) were incorporated in the films either during the film formation and casting or after the film synthesis, via bacteria diffusion and adsorption. The later approach could efficiently entrap and preserve viable bacteria. The mechanical properties and swelling ability could be tuned by varying the HEC/CMC ratio and the amount of CA. Moreover, the surface area and total pore volume of the films considerably decreased after cross-linking. The growing demand for dairy products amended with probiotics has led to the exploration of new beneficial microorganisms such as probiotics with beneficial properties. In the present work, the probiotic and antioxidant potential of Lactobacillus fermentum strains isolated from dairy products were evaluated. Strains were investigated for their probiotic properties by performing different tests such as survival in pepsin, low pH, and bile salt, antibacterial activity, and antioxidant potential. These strains were further evaluated for their utilisation in yogurt formation as a probiotic. The isolated strains were identified as L. fermentum Y1, L. fermentum Y2, and L. fermentum C by 16S rRNA sequencing. All strains showed greater survival ability in simulated gastric conditions (pH 2.2 + pepsin) and in the presence of 0.3% bile salt. The highest antibacterial activity was exhibited by L. fermentum Y1 against Bacillus cereus. Among these three strains, L. fermentum Y1 had the highest reducing power, and L. fermentum C had the highest DPPH scavenging activity. All Lactobacillus strains as a single inoculum or in consortium showed significant (p < 0.05) probiotic properties by maintaining pH, titratable acidity, solid content, and high water holding capacity in comparison to the control in the cow yogurt and homogenised milk. The isolated Lactobacillus strains may be a potential source of probiotics in commercial yogurt preparation.

Children aged 3 months to 48 months who present between November 2013 and June 2017 with <72 hours of gastroenteritis symptoms will be assessed for eligibility. A total of 886 children will be randomized (1:1 allocation via an internet based, third party, randomization service) to receive 5 days of a combination probiotic agent (Lactobacillus rhamnosus and L. helveticus) or placebo. All participants, caregivers, and outcome assessors will be blinded to group assignment. The study includes three key outcomes: 1) clinical - the development of moderate to severe disease following an emergency department (ED) evaluation that employs a validated clinical score (Modified Vesikari Scale); 2) safety - side effect; and 3) mechanism - fecal secretory immunoglobulin A levels. Discussion Definitive data are lacking to guide the clinical use of probiotics in children with acute gastroenteritis. Hence, probiotics are rarely prescribed by North American physicians. However, the following current trends obligate an urgent assessment: 1) probiotics are sold as food supplements, and manufacturers can encourage their use while their relevance has yet to be established; 2) North American and European government agencies remain concerned about their value and safety; 3) some institutions are now recommending the routine use of probiotics; and 4) parents of affected children are often providing probiotics. With probiotic consumption increasing in the absence of solid evidence, there is a need to conduct this definitive trial to overcome the limitations of prior work in this field. Using the method of scanning electron microscopy, a comparative study of the particle surface morphology and the elemental composition of the cuticle of Hermetia illucens pupa before and after the application of the culture liquid (CL) of the probiotic strain Lactobacillus acidophilus B 2707 was carried out. At the same time, under the influence of CL, destruction of relatively large particles of the powder of the chitin-protein complex of the cuticle (average particle diameter D 845.3 ± 109.1 μm) and the formation of small fragments (average D 63.4 ± 13.9 μm), including nanoparticles (average D 54 ± 18 nm). A biofilm of lactobacilli enriched with calcium, magnesium and phosphorus was obtained on a sorbent consisting of microparticles of the cuticle of insect pupae and including nanoparticles, which provided the probiotic preparation with high biological efficiency.

The aim of this study was to evaluate the possibility of using Staphylococcus epidermidis cells as a probiotic to treat some skin infections. For this purpose, S. epidermidis Y73, which is an active bacteriocin producer and non-biofilm forming isolate, was selected among 134 skin isolates through primary and secondary screening. Tryptic soya broth was selected as the best medium to support bacteriocin production, while the optimal pH and temperature for S. epidermidis Y73 growth were 7 and 37°C, respectively, which were invested in the formula preparation. Furthermore, the possibility of using this isolate as a probiotic was investigated by preparing 4 potential cellulosic pads with 4 different formulae which were all subjected to an in vitro trial to select the one which is superior to the others in terms of supporting bacteriocin production and cells viability. The shelf life of the pad was estimated and the results showed that the cells remained vital until the 20th week. The selected pad formula was used to treat artificially induced wounds on rabbit skin. The wounds were infected with Staphylococcus aureus, Kocuria rosea and Pseudomonas. aeruginosa. The symptoms in both control and treated animals were recorded and, based on the results; the healing process with the presence of the S. epidermidis Y73 pad was significantly faster compared with that for the control. This research will serve as a base for future studies on using vital cells of S. epidermidis as probiotics and, hence, make a contribution to the current literature on using live cells to treat bacterial skin infections.

Probiotics are used in poultry farming for prevention and treatment of infectious diseases of gastrointestinal tract, immune stimulation, correction of dysbacteriosis of digestive tract, and for replacement of antibiotics in compound feed. Increasing the fiber level in diet for poultry is considered as one of the nutrition strategies with the aim of reduction of the frequency of problems in the gastrointestinal tract as well. The purpose of research is to study the effect of grass meal and probiotic in diet for poultry on the intestinal microbiota, fat deposition and folliculogenesis. The experiment had been conducted on meat-and-egg type of hens of Amrox and Sussex breeds. From 18 weeks of age, within 12 weeks hens of the control groups were fed a normal diet, in the I experimental group 10 % of the usual diet was replaced with alfalfa grass flour, and in the II experimental group 10 % of the diet was replaced with grass flour with addition of enzyme preparation "Cellobacterin-T", produced by Biotrof Ltd. Feeding grass meal along with probiotic positively affected the metabolic processes in body, so the amount of abdominal fat in carcass decreased. Fat reduction exceeded 20 % in poultry of experimental groups. It has been also determined that the number of maturing follicles in the ovaries of hens in experimental groups was 25-64 % higher, which indicates a positive effect of the experimental diet on potential egg production. Analysis of cecum contents in digestive tract of hens using the modern molecular genetic method T-RFLP showed that use of grass meal in diet for poultry along with Cellobacterin-T probiotic contributed to formation of useful microflora, as well as to decrease in the number of undesirable actinomycetes and pathogenic mycoplasmas in gastrointestinal tract of hens.

Background. Today, allergic respiratory diseases in children, including asthma and allergic rhinitis, are a relevant medical and social problem associated with a high risk of disability, a significant reduction in the quality of life of patients and their families, and a significant financial burden on patients and the government. Bronchial asthma and allergic rhinitis often coexist, complicating the course of each other. Children with respiratory allergic diseases are particularly vulnerable to acute respiratory infections. Respiratory viruses, in turn, play an important role in the pathogenesis of bronchial asthma and allergic rhinitis, contributing to the development of diseases and acting as triggers for their exa­cerbations. One of the possible causes of predisposition to frequent acute respiratory infections in children with allergic pathology of the respiratory system may be a lack of nonspecific factors of mucosal immunity of the respiratory tract — endogenous cationic amphiphilic antimicrobial peptides, in particular, cathelicidin and human defensin. According to many studies, these antimicrobial peptides have antiviral, antimicrobial and immunomodulatory effects. However, nowadays there is insufficient data on the state of production of antimicrobial peptides and their impact on respiratory morbidity in children with allergic diseases. At the same time, some literature sources describe the positive effect of individual probiotic strains, in particular Lactobacillus rhamnosus GG, on the overall resistance to respiratory viral infection and the prevention of atopic diseases. The purpose of our research was to study the concentrations of antimicrobial peptides (HbD-2 and LL-37) in the secretion of the mucous membranes of the upper respiratory tract in children with bronchial asthma and allergic rhinitis, to clarify their role in protection against respiratory viral infection and to estimate the effectiveness of a probiotic preparation containing Lactobacillus rhamnosus GG in the prevention of acute respiratory infections and reduction of the frequency of virus-induced exacerbations of allergic pathology. Materials and methods. We conducted a clinical and laboratory examination of 76 children aged 7 to 17 years, 24 of whom were diagnosed with allergic rhinitis, 28 children — bronchial asthma, and 24 — bronchial asthma and allergic rhinitis. The control group consisted of 20 clinically healthy children of matched age and sex. Research methods also included the analysis of respiratory disease patterns, determination of concentrations of antimicrobial peptides: cathelicidin (LL-37), human β-defensin 2 (HbD-2) in the secretion of the upper respiratory tract by ELISA, and statistical analysis. Results. It was found that children with allergic diseases of the respiratory tract are characterized by a higher frequency of acute respiratory infections (30 %) with more frequent involvement of the lower respiratory tract in the inflammatory process (72 %), which causes a longer duration of disease (95 %) compared to healthy children. The children with allergic rhinitis and bronchial asthma had a significant decrease in the concentrations of antimicrobial peptides in the secretion of the upper respiratory tract compared with the control group. It was found that the level of antimicrobial peptide concentrations in the secretion of the upper respiratory tract is most negatively affected by the allergic inflammation severity, accompanied by a lack of symptom control, and the use of basic therapy and no symptoms have a positive effect. However, even in the presence of complete control of the symptoms of allergic diseases, the levels of antimicrobial peptides were significantly lower than in children of the control group. It was found that there is an inverse relationship between the levels of antimicrobial peptides and the frequency and duration of acute respiratory infections in children with allergic diseases, most pronounced in bronchial asthma. Conclusions. Taking a probiotic drug containing a strain of Lactobacillus rhamnosus GG for a month in addition to the basic therapy of allergic rhinitis and bronchial asthma contributed to a significant increase in levels of HbD-2 (average 27 %) and ­LL-37 (average 60 %) in secretions of the upper respiratory tract, which was accompanied by a decrease in the frequency of acute respiratory infections by an average of 26 % (including episodes involving the lower respiratory tract), a decrease in the average duration of acute respiratory infections by 22 % over the next year and a decrease in virus-induced exacerbations by 25.7 %. ; Актуальность. На сегодняшний день аллергические заболевания респираторного тракта, в частности бронхиальная астма и аллергический ринит, у детей являются актуальной медико-социальной проблемой, ассоциированной с высоким риском инвалидизации, существенным снижением качества жизни пациентов и их семей, а также значительной финансовой нагрузкой на пациентов и государство. Бронхиальная астма и аллергический ринит часто сосуществуют, обременяя течение друг друга. Дети, больные респираторными аллергическими заболеваниями, являются особенно уязвимыми к развитию острой респираторной вирусной инфекции (ОРВИ), а респираторные вирусы, в свою очередь, играют важную роль в патогенезе бронхиальной астмы и аллергического ринита, способствуя развитию заболеваний и выступая триггерами их обострений. Одной из возможных причин склонности к частым ОРВИ у детей с аллергической патологией дыхательной системы может быть недостаточность неспецифических факторов мукозального иммунитета респираторного тракта — эндогенных катионоактивных амфифильных антимикробных пептидов, в частности кателицидина (LL-37) и человеческих дефензинов. По данным ряда исследований, эти антимикробные пептиды проявляют противовирусное, антимикробное и иммуномодулирующее действие. При этом данных о состоянии продукции антимикробных пептидов и их влиянии на респираторную заболеваемость у детей с аллергическими заболеваниями в современной литературе недостаточно. В то же время в ряде литературных источников описано положительное влияние отдельных пробиотических штаммов, в частности Lactobacillus rhamnosus GG, на общую резистентность к респираторной вирусной инфекции и профилактику атопических заболеваний. Целью нашей работы стало изучение уровней антимикробных пептидов (человеческого β-дефензина 2 (HbD-2) и LL-37) в секрете слизистых оболочек верхних дыхательных путей у детей с бронхиальной астмой и аллергическим ринитом, выяснение их роли в защите от респираторной вирусной инфекции у данного контингента больных и оценка эффективности применения пробиотического препарата, содержащего Lactobacillus rhamnosus GG, с целью профилактики ОРВИ и снижения частоты вирус-индуцированных обострений аллергической патологии. Материалы и методы. Нами было проведено клинико-лабораторное обследование 76 детей в возрасте от 7 до 17 лет, из которых у 24 был диагностирован аллергический ринит, у 28 детей — бронхиальная астма, а у 24 — бронхиальная астма и аллергический ринит. Группу контроля составили 20 клинически здоровых детей соответствующего возраста и пола. Кроме общепринятых клинических, методы исследования включали анализ паттернов респираторной заболеваемости, определение уровней антимикробных пептидов LL-37, HbD-2 в секрете верхних дыхательных путей методами иммуноферментного и статистического анализа. Результаты. Установлено, что дети с аллергическими заболеваниями респираторного тракта характеризуются большей частотой ОРВИ (на 30 %) с более частым вовлечением в воспалительный процесс нижних дыхательных путей (на 72 %), что обусловливает более длительную продолжительность заболеваний (на 95 %) по сравнению со здоровыми детьми. У детей с аллергическим ринитом и бронхиальной астмой обнаружено существенное снижение уровней антимикробных пептидов в секрете верхних дыхательных путей по сравнению с группой контроля. На уровни антимикробных пептидов в секрете верхних дыхательных путей в наибольшей степени негативно влияет степень активности аллергического воспаления, сопровождающегося отсутствием контроля симптомов, а положительно — прием препаратов базисной терапии и отсутствие симптомов. Вместе с тем даже при наличии полного контроля симптомов аллергических заболеваний, то есть при минимальном уровне аллергического воспаления в дыхательных путях, уровни антимикробных пептидов были достоверно ниже, чем у детей контрольной группы. Установлено, что существует обратная связь между уровнями антимикробных пептидов и частотой и продолжительностью ОРВИ у детей с аллергическими заболеваниями, которая в наибольшей степени выражена при бронхиальной астме. Выводы. Прием пробиотического препарата, содержащего штамм Lactobacillus rhamnosus GG, в течение 1 месяца дополнительно к базисной терапии аллергического ринита и бронхиальной астмы способствовал достоверному повышению уровней HbD-2 (в среднем на 27 %) и LL-37 (в среднем на 60 %) в секрете верхних дыхательных путей, что сопровождалось снижением частоты ОРВИ в среднем на 26 % (в том числе и эпизодов с привлечением нижних дыхательных путей), уменьшением средней продолжительности ОРВИ на 22 % в течение следующего года и снижением вирус-индуцированных обострений на 25,7 %. ; Актуальність. На сьогодні алергічні захворювання респіраторного тракту, зокрема бронхіальна астма й алергічний риніт, у дітей є актуальною медико-соціальною проблемою, асоційованою з високим ризиком інвалідизації, суттєвим зниженням якості життя пацієнтів та їх сімей, а також значним фінансовим навантаженням на пацієнтів і державу. Бронхіальна астма й алергічний риніт часто співіснують, обтяжуючи перебіг одне одного. Діти, хворі на респіраторні алергічні захворювання, особливо вразливі до розвитку гострої респіраторної вірусної інфекції (ГРВІ), а респіраторні віруси, в свою чергу, відіграють важливу роль у патогенезі бронхіальної астми й алергічного риніту, сприяючи розвитку захворювань і виступаючи тригерами їх загострень. Однією з можливих причин схильності до частих ГРВІ у дітей з алергічною патологією респіраторної системи може бути недостатність неспецифічних факторів мукозального імунітету респіраторного тракту — ендогенних катіоноактивних амфіфільних антимікробних пептидів, зокрема кателіцидину (LL-37) та людських дефензинів. За даними низки досліджень, дані антимікробні пептиди проявляють противірусну, антимікробну та імуномодулюючу дію. При цьому даних щодо стану продукції антимікробних пептидів та їх впливу на респіраторну захворюваність у дітей з алергічними захворюваннями в сучасній літературі недостатньо. В той же час в низці літературних джерел описано позитивний вплив окремих пробіотичних штамів, зокрема Lactobacillus rhamnosus GG, на загальну резистентність до респіраторної вірусної інфекції та профілактику атопічних захворювань. Метою нашої роботи стало вивчення рівнів антимікробних пептидів (людського β-дефензину 2 (HbD-2) та LL-37) у секреті слизових оболонок верхніх дихальних шляхів у дітей з бронхіальною астмою й алергічним ринітом, з'ясування їх ролі у захисті від респіраторної вірусної інфекції у даного контингенту хворих та оцінка ефективності застосування пробіотичного препарату, що містить Lactobacillus rhamnosus GG, з метою профілактики ГРВІ та зниження частоти вірус-індукованих загострень алергічної патології. Матеріали та методи. Нами було проведено клініко-лабораторне обстеження 76 дітей віком від 7 до 17 років, з яких у 24 був діагностований алергічний риніт, у 28 дітей — бронхіальна астма, а у 24 — бронхіальна астма й алергічний риніт. Групу контролю становили 20 клінічно здорових дітей відповідного віку та статі. Окрім загальноклінічних, методи дослідження включали аналіз патернів респіраторної захворюваності, визначення рівнів антимікробних пептидів LL-37, HbD-2 у секреті верхніх дихальних шляхів методами імуноферментного та статистичного аналізу. Результати. Встановлено, що діти з алергічними захворюваннями респіраторного тракту характеризуються більшою частотою ГРВІ (на 30 %) з більш частим залученням у запальний процес нижніх дихальних шляхів (на 72 %), що обумовлює більшу тривалість захворювань (на 95 %) порівняно із здоровими дітьми. У дітей з алергічним ринітом і бронхіальною астмою виявлено суттєве зниження рівнів антимікробних пептидів у секреті верхніх дихальних шляхів порівняно з групою контролю. На рівні антимікробних пептидів у секреті верхніх дихальних шляхів найбільшою мірою негативно впливає ступінь активності алергічного запалення, що супроводжується відсутністю контролю симптомів, а позитивно — прийом препаратів базисної терапії та відсутність симптомів. Разом з тим навіть за наявності повного контролю симптомів алергічних захворювань, тобто при мінімальному рівні алергічного запалення в дихальних шляхах, рівні антимікробних пептидів були вірогідно нижчими, ніж у дітей контрольної групи. Встановлено, що існує зворотний зв'язок між рівнями антимікробних пептидів і частотою та тривалістю ГРВІ у дітей з алергічними захворюваннями, найбільш виражений при бронхіальній астмі. Висновки. Прийом пробіотичного препарату, що містить штам Lactobacillus rhamnosus GG, протягом 1 місяця додатково до базисної терапії алергічного риніту та бронхіальної астми сприяв вірогідному підвищенню рівнів HbD-2 (в середньому на 27 %) та LL-37 (в середньому на 60 %) у секреті верхніх дихальних шляхів, що супроводжувалось зниженням частоти ГРВІ в середньому на 26 % (в тому числі й епізодів із залученням нижніх дихальних шляхів), зменшенням середньої тривалості ГРВІ на 22 % протягом наступного року та зниженням вірус-індукованих загострень на 25,7 %.

Background. Today, allergic respiratory diseases in children, including asthma and allergic rhinitis, are a relevant medical and social problem associated with a high risk of disability, a significant reduction in the quality of life of patients and their families, and a significant financial burden on patients and the government. Bronchial asthma and allergic rhinitis often coexist, complicating the course of each other. Children with respiratory allergic diseases are particularly vulnerable to acute respiratory infections. Respiratory viruses, in turn, play an important role in the pathogenesis of bronchial asthma and allergic rhinitis, contributing to the development of diseases and acting as triggers for their exa­cerbations. One of the possible causes of predisposition to frequent acute respiratory infections in children with allergic pathology of the respiratory system may be a lack of nonspecific factors of mucosal immunity of the respiratory tract — endogenous cationic amphiphilic antimicrobial peptides, in particular, cathelicidin and human defensin. According to many studies, these antimicrobial peptides have antiviral, antimicrobial and immunomodulatory effects. However, nowadays there is insufficient data on the state of production of antimicrobial peptides and their impact on respiratory morbidity in children with allergic diseases. At the same time, some literature sources describe the positive effect of individual probiotic strains, in particular Lactobacillus rhamnosus GG, on the overall resistance to respiratory viral infection and the prevention of atopic diseases. The purpose of our research was to study the concentrations of antimicrobial peptides (HbD-2 and LL-37) in the secretion of the mucous membranes of the upper respiratory tract in children with bronchial asthma and allergic rhinitis, to clarify their role in protection against respiratory viral infection and to estimate the effectiveness of a probiotic preparation containing Lactobacillus rhamnosus GG in the prevention of acute respiratory infections and reduction of the frequency of virus-induced exacerbations of allergic pathology. Materials and methods. We conducted a clinical and laboratory examination of 76 children aged 7 to 17 years, 24 of whom were diagnosed with allergic rhinitis, 28 children — bronchial asthma, and 24 — bronchial asthma and allergic rhinitis. The control group consisted of 20 clinically healthy children of matched age and sex. Research methods also included the analysis of respiratory disease patterns, determination of concentrations of antimicrobial peptides: cathelicidin (LL-37), human β-defensin 2 (HbD-2) in the secretion of the upper respiratory tract by ELISA, and statistical analysis. Results. It was found that children with allergic diseases of the respiratory tract are characterized by a higher frequency of acute respiratory infections (30 %) with more frequent involvement of the lower respiratory tract in the inflammatory process (72 %), which causes a longer duration of disease (95 %) compared to healthy children. The children with allergic rhinitis and bronchial asthma had a significant decrease in the concentrations of antimicrobial peptides in the secretion of the upper respiratory tract compared with the control group. It was found that the level of antimicrobial peptide concentrations in the secretion of the upper respiratory tract is most negatively affected by the allergic inflammation severity, accompanied by a lack of symptom control, and the use of basic therapy and no symptoms have a positive effect. However, even in the presence of complete control of the symptoms of allergic diseases, the levels of antimicrobial peptides were significantly lower than in children of the control group. It was found that there is an inverse relationship between the levels of antimicrobial peptides and the frequency and duration of acute respiratory infections in children with allergic diseases, most pronounced in bronchial asthma. Conclusions. Taking a probiotic drug containing a strain of Lactobacillus rhamnosus GG for a month in addition to the basic therapy of allergic rhinitis and bronchial asthma contributed to a significant increase in levels of HbD-2 (average 27 %) and ­LL-37 (average 60 %) in secretions of the upper respiratory tract, which was accompanied by a decrease in the frequency of acute respiratory infections by an average of 26 % (including episodes involving the lower respiratory tract), a decrease in the average duration of acute respiratory infections by 22 % over the next year and a decrease in virus-induced exacerbations by 25.7 %. ; Актуальность. На сегодняшний день аллергические заболевания респираторного тракта, в частности бронхиальная астма и аллергический ринит, у детей являются актуальной медико-социальной проблемой, ассоциированной с высоким риском инвалидизации, существенным снижением качества жизни пациентов и их семей, а также значительной финансовой нагрузкой на пациентов и государство. Бронхиальная астма и аллергический ринит часто сосуществуют, обременяя течение друг друга. Дети, больные респираторными аллергическими заболеваниями, являются особенно уязвимыми к развитию острой респираторной вирусной инфекции (ОРВИ), а респираторные вирусы, в свою очередь, играют важную роль в патогенезе бронхиальной астмы и аллергического ринита, способствуя развитию заболеваний и выступая триггерами их обострений. Одной из возможных причин склонности к частым ОРВИ у детей с аллергической патологией дыхательной системы может быть недостаточность неспецифических факторов мукозального иммунитета респираторного тракта — эндогенных катионоактивных амфифильных антимикробных пептидов, в частности кателицидина (LL-37) и человеческих дефензинов. По данным ряда исследований, эти антимикробные пептиды проявляют противовирусное, антимикробное и иммуномодулирующее действие. При этом данных о состоянии продукции антимикробных пептидов и их влиянии на респираторную заболеваемость у детей с аллергическими заболеваниями в современной литературе недостаточно. В то же время в ряде литературных источников описано положительное влияние отдельных пробиотических штаммов, в частности Lactobacillus rhamnosus GG, на общую резистентность к респираторной вирусной инфекции и профилактику атопических заболеваний. Целью нашей работы стало изучение уровней антимикробных пептидов (человеческого β-дефензина 2 (HbD-2) и LL-37) в секрете слизистых оболочек верхних дыхательных путей у детей с бронхиальной астмой и аллергическим ринитом, выяснение их роли в защите от респираторной вирусной инфекции у данного контингента больных и оценка эффективности применения пробиотического препарата, содержащего Lactobacillus rhamnosus GG, с целью профилактики ОРВИ и снижения частоты вирус-индуцированных обострений аллергической патологии. Материалы и методы. Нами было проведено клинико-лабораторное обследование 76 детей в возрасте от 7 до 17 лет, из которых у 24 был диагностирован аллергический ринит, у 28 детей — бронхиальная астма, а у 24 — бронхиальная астма и аллергический ринит. Группу контроля составили 20 клинически здоровых детей соответствующего возраста и пола. Кроме общепринятых клинических, методы исследования включали анализ паттернов респираторной заболеваемости, определение уровней антимикробных пептидов LL-37, HbD-2 в секрете верхних дыхательных путей методами иммуноферментного и статистического анализа. Результаты. Установлено, что дети с аллергическими заболеваниями респираторного тракта характеризуются большей частотой ОРВИ (на 30 %) с более частым вовлечением в воспалительный процесс нижних дыхательных путей (на 72 %), что обусловливает более длительную продолжительность заболеваний (на 95 %) по сравнению со здоровыми детьми. У детей с аллергическим ринитом и бронхиальной астмой обнаружено существенное снижение уровней антимикробных пептидов в секрете верхних дыхательных путей по сравнению с группой контроля. На уровни антимикробных пептидов в секрете верхних дыхательных путей в наибольшей степени негативно влияет степень активности аллергического воспаления, сопровождающегося отсутствием контроля симптомов, а положительно — прием препаратов базисной терапии и отсутствие симптомов. Вместе с тем даже при наличии полного контроля симптомов аллергических заболеваний, то есть при минимальном уровне аллергического воспаления в дыхательных путях, уровни антимикробных пептидов были достоверно ниже, чем у детей контрольной группы. Установлено, что существует обратная связь между уровнями антимикробных пептидов и частотой и продолжительностью ОРВИ у детей с аллергическими заболеваниями, которая в наибольшей степени выражена при бронхиальной астме. Выводы. Прием пробиотического препарата, содержащего штамм Lactobacillus rhamnosus GG, в течение 1 месяца дополнительно к базисной терапии аллергического ринита и бронхиальной астмы способствовал достоверному повышению уровней HbD-2 (в среднем на 27 %) и LL-37 (в среднем на 60 %) в секрете верхних дыхательных путей, что сопровождалось снижением частоты ОРВИ в среднем на 26 % (в том числе и эпизодов с привлечением нижних дыхательных путей), уменьшением средней продолжительности ОРВИ на 22 % в течение следующего года и снижением вирус-индуцированных обострений на 25,7 %. ; Актуальність. На сьогодні алергічні захворювання респіраторного тракту, зокрема бронхіальна астма й алергічний риніт, у дітей є актуальною медико-соціальною проблемою, асоційованою з високим ризиком інвалідизації, суттєвим зниженням якості життя пацієнтів та їх сімей, а також значним фінансовим навантаженням на пацієнтів і державу. Бронхіальна астма й алергічний риніт часто співіснують, обтяжуючи перебіг одне одного. Діти, хворі на респіраторні алергічні захворювання, особливо вразливі до розвитку гострої респіраторної вірусної інфекції (ГРВІ), а респіраторні віруси, в свою чергу, відіграють важливу роль у патогенезі бронхіальної астми й алергічного риніту, сприяючи розвитку захворювань і виступаючи тригерами їх загострень. Однією з можливих причин схильності до частих ГРВІ у дітей з алергічною патологією респіраторної системи може бути недостатність неспецифічних факторів мукозального імунітету респіраторного тракту — ендогенних катіоноактивних амфіфільних антимікробних пептидів, зокрема кателіцидину (LL-37) та людських дефензинів. Numerose formulazioni probiotiche sono reperibili sul mercato e vendute sotto forma di integratori alimentari in grado di migliorare il benessere fisico del consumatore. In realtà, per molti preparati probiotici esistono specifici health claims, preventivamente approvati dall'EFSA, che vengono utilizzati per indirizzare il consumatore al loro impiego nella prevenzione o la cura di differenti malattie. Dunque, in presenza di un effetto salutistico dichiarato, il formulato probiotico dovrebbe contenere particolari ceppi, con adeguate cariche di microrganismi vivi e vitali, in grado di agire a livello intestinale per alleviare i sintomi o coadiuvare la cura di specifiche patologie. Non a caso, fra i criteri dettati per l'uso appropriato del termine "probiotico", c'è l'accertamento dello specifico effetto benefico che il microrganismo, o il pool di microrganismi, è in grado di apportare al consumatore. Altro importante criterio di valutazione è rappresentato dalla capacità di sopravvivenza al transito gastroenterico per arrivare nel colon con elevate cariche di microrganismi vivi e vitali. Tuttavia, a differenza dei farmaci, che sono intensamente ispezionati dagli organismi di controllo per verificarne la sicurezza d'uso e l'efficacia, i probiotici sono controllati con minore rigore. Di conseguenza, è molto più semplice approfittare della lacuna legislativa esistente in tale ambito per immettere sul mercato nuovi prodotti probiotici la cui sicurezza ed efficacia sono state spesso solo sommariamente testate. Alla luce di tali premesse, la prima parte del lavoro di tesi di dottorato ha inteso valutare la corrispondenza dei ceppi dichiarati in etichetta con quelli realmente presenti in differenti formulati probiotici farmaceutici, valutandone anche la sopravvivenza quando sottoposti agli stress tipici del transito gastrointestinale. Tale valutazione, a differenza di quanto riportato in numerosi studi consultati, è stata condotta eliminando la fase preliminare di rivitalizzazione delle preparazioni liofilizzate, dal momento che la loro ingestione da parte del consumatore, e il successivo passaggio lungo il tratto gastrointestinale, non prevede questa condizione. L'attività successiva ha quindi previsto la valutazione della sopravvivenza alla conservazione e al transito gastroenterico simulato di alcuni ceppi probiotici commerciali quando integrati in una matrice alimentare, al fine di valutarne le capacità protettive. Da questo punto di vista, gli alimenti possono essere in grado di influenzare positivamente la sopravvivenza dei microrganismi nel tratto gastrointestinale, svolgendo pertanto un ruolo importante di veicolo dei probiotici con la dieta. Il loro impiego può divenire un approccio vincente dal punto di vista economico e lodevole per il carattere nutrizionale, a patto che la scelta dell'alimento ricada su matrici adeguatamente presenti nelle diete dei consumatori e che, eventualmente, siano già in possesso di componenti di interesse salutistico nonché accattivanti per le più ampie fasce di età. In tale ottica, la scelta è ricaduta sul cioccolato fondente. I risultati ottenuti hanno tuttavia evidenziato che tale matrice alimentare, nonostante la ricchezza in ingredienti di pregio dal punto di vista nutrizionale, emozionale e salutistico, non può costituire un veicolo preferenziale per proteggere i microrganismi durante il loro viaggio verso l'intestino. Da questa considerazione è scaturita un'ultima attività di ricerca, caratterizzata dalla valutazione dell'influenza di ciascun fattore di stress gastroenterico sulla sopravvivenza di ceppi di Lactobacillus rhamnosus isolati da matrici alimentari, campioni fecali e probiotici del commercio. Tale indagine ha restituito risultati particolarmente interessati non solo con riguardo alle condizioni che maggiormente influenzano la vitalità dei ceppi durante il transito ma anche alle matrici di origine alimentare (caseina) in grado di svolgere un ragguardevole effetto protettivo contro le ingiurie dovute a tali condizioni. ; Many probiotic formulations are available on the market and sold in the form of food supplements that can improve physical health of the consumer. In fact, for many probiotic preparations, there are specific health claims, previously approved by EFSA, that are used to direct the consumer to their use in the prevention or treatment of different diseases. Therefore, in presence of a healthy effect stated, the formulation should contain particular probiotic strains, with appropriate amount of live and viable microorganisms, able to act in the intestine to relieve symptoms or assist the treatment of specific pathologies. Not surprisingly, among the criteria dictated by the appropriate use of the term "probiotic", there is the determination of the specific beneficial effect that the microorganism, or the pool of microorganisms, is able to give to the consumer. Another important evaluation criterion is represented by the ability to survive to the gastrointestinal transit, in order to arrive to the colon with high amount of live and viable microorganisms. However, unlike drugs, which are intensively inspected by the inspection authorities to ensure safe use and efficacy, probiotics are controlled less rigorously. As a result, it is much easier to take advantage of the existing legislative gap in this area, to bring to the market new probiotic products whose safety and efficacy have been often only slightly tested. In light of these circumstances, the first part of the PhD research was aimed to the identification of microorganisms composing different lactic acid bacteria based pharmaceutical formulations and the ascertainment of their ability to survive gastrointestinal (GI) stresses, the main requisite to produce beneficial effects. This evaluation, unlike what is reported in numerous studies available in literature, was conducted without a preliminary rehydration step, since their ingestion by the consumer, and the next step through the gastrointestinal tract, does not contemplate this condition. The next activity was focused on the evaluation of viability, after the conservation and simulated gastrointestinal transit, of some commercial probiotic strains integrated in a food matrix, in order to evaluate the protective capacity. From this point of view, food may be able to positively affect the survival of microorganisms in the gastrointestinal tract, thus playing an important role as a vehicle of probiotics in the diet. Their use can therefore become a winning approach from an economic and nutritional standpoint, as long as the food choice falls on matrices that are not only adequately present in consumers' diets, but also already have healthy interesting components and are appealing enough to the widest age range of people. In this context, our choice fell on dark chocolate. However, the results showed that this food matrix, although is rich in valuable ingredients from the nutritional, healthy and emotional point of view, can not be a preferential vehicle to protect the organisms during their passage through the intestine. From this consideration, in fact, derived one last activity: the assessment of the influence of each gastrointestinal stress factor on the survival of Lactobacillus rhamnosus strains isolated from food matrices, fecal samples and probiotics pharmaceutical formulations. That analysis furnished results particularly interesting not only with regard to the conditions that mostly affect the viability of the strains during transit but also to food-borne matrices (casein) that are able to play a substantial protective effect against the injuries caused by these conditions. ; Dottorato di ricerca in Biotecnologia degli alimenti (XXVIII ciclo)

ABSTRACT: Dairy products are an adequate alternative as functional foods since they present excellent nutritional value, have wide acceptance with consumers, and are potential matrices for inoculation of probiotic microorganisms. The present study aimed to develop a fermented milk beverage with fruits and a probiotic culture included, to evaluate the acceptance of this product among consumers to define the ideal formulation, and to perform Lactobacillus acidophilus counting in order to characterize the product as a probiotic food. For the formulation of the beverage, milk, whey, sugar, and pasteurized orange and papaya pulps were used as main ingredients. The beverage formulation consisted of mixing the fruit pulps with a dairy substrate of milk and whey which was previously fermented using a mixed commercial probiotic ABT-5 culture. Six formulations were tested. The milk substrate was maintained at 70%. These formulations consisted of 50% of mineral water and 50% of skimmed milk with a concentration of probiotics of 0.5g/L. Sugar concentration varied between 40g, 70g, or 100g. As for flavoring, the juice was composed of 180mL of papaya and 120mL of orange, or 150mL of papaya and 150mL of orange. Beverages were prepared and then stored at 5±1°C. These products were evaluated after 24 hours of preparation/storage by 90 tasters (consumers) for acceptance and sensory characterization. Data were analyzed by ANOVA. The differences between samples were evaluated using the Tukey test at a significance level of 5%. Acceptance of the beverage samples among tasters varied from 5.1 to 6.9 and ranged between the hedonistic terms I neither liked nor disliked it and somewhat enjoyed it in a 9-point hedonic scale. The enumeration of Lactobacillus acidophilus LA-5 ranged from 7.83 to 8.09log CFU/mL in the first week of refrigerated storage. Our results suggested that the beverages analyzed were suitable for inoculation of probiotics and met the legislation requirements. Therefore, these beverages should be classified as probiotic products. Samples with 10% sugar were the least preferred by the study participants whereas beverages containing 40g, 70g and 180mL of papaya juice and 120mL of orange juice and the sample with 70g of sugar, 150mL of papaya juice, and 150mL of orange juice did not differ significantly from each other in terms of acceptance and were; therefore, the 3 formulations that received the highest means of acceptance by consumers.

Bifidobacterium longum INIA P132 and Bifidobacterium infantis INIA P731, isolated from infant-faeces, were investigated in this work. Regarding the probiotic and technological potential of the bifidobacteria, both were resistant to gastrointestinal tract simulated conditions. B. longum showed high survival upon freezing and thawing as well as lyophilisation and was able to grow in milk. B. infantis had higher adhesion capacity to human Caco-2 cells than the commercial probiotic Bifidobacterium animalis BB12 strain. Moreover, both bacteria secrete heteropolysaccharides (HePS) composed of rhamnose, galactose and glucose. In a dextran sodium sulphate-induced enterocolitis model in zebra fish larvae, treatment with each HePS preparation resulted in a decrease of the larval mortality. In addition, the HePS from B. longum immunomodulated in vitro human macrophages treated with the inflammatory Escherichia coli O111:B4 lipopolysaccharide.
